Exon Array analysis of WT1(+KTS) mutant mice

ABSTRACT: This exon array analysis was performed on mice carrying splice specific mutations at the end of exon9 of the Wilms tumor supressor gene that interfere with the production of WT1(+KTS) isoforms (Hammes et al., 2001; Cell 106:309) 4 wildtype and 4 wt1(+KTS) mutant E18.5 mouse embryos were transcardially perfused with magnetic beads, glomeruli isolated and RNA extracted and analyzed for differential expression on Affymetrix Exon Arrays

Monocular deprivation by TTX-injection (at P12-13 and again at P13-14), followed by manual sorting of m-Citrin-labeled Layer 4 Visual Cortex Star Pyramid neurons in deprived and non-deprived hemispheres. RNA was extracted using PicoPure RNA Isolation Kit, reverse transcribed, and amplified using a standard T7 IVT protocol (Affymetrix Small Sample Target Labeling Assay Version II).

Project description:To characterize gene expression in Gata6 positive epidermal cells we analyzed a Gata6 reporter mouse in which the endogenous Gata6 promoter drives expression of mTomato. We performed flow cytometry followed by transcriptome analysis. We compared four subpopulations of telogen epidermal cells: Gata6+/Itga6+ cells, Gata6+/itga6- cells, CD34+/Itga6+ cells (which are Gata6-) and all remaining Itga6+ cells (Gata6-/CD34-). The RNA was isolated from age and sex matched mice. Epidermal overexpression of âNLef1 leads to deregulation of epidermal stem cell fate.

Project description:In adult K14ÎNLef1 mouse, the overexpression of âNLef1, a B-catenin dominat negative, in basal keratinocytes leads to the conversion of hair follicles into multilayered epithelial cysts and ectopic sebaceous gland. To uncover in vivo changes in gene expression associated to âNLef1 activity, we compared the expression profiles of unfractionated keratinocytes in wild type and K14ÎNLef1 transgenic mice. Cells were collected from 9.5 week old mice, when the hair follicle are in the resting (telogen) phase of the hair growth cycle. At this stage the âNLef1 phenotype is not yet evident enabling us to detect early molecular events. We compared the expression profiles of unfractionated keratinocytes in wild type and K14ÎNLef1 transgenic 9.5 week old mice. Three biological replicates for each cell population were analyzed.

Gene expression data of cultured primary microglia from neonatal control and Cstb-/- mice.

Project description:Cystain B (Cstb) is a ubiquitously expressed cysteine protease inhibitor and mutations of the CSTB gene lead to the neurodegenerative disease progressive myoclonus epilepsy of Unverricht-Lundorg type (EPM1). We are interested in the microglia-specific, Cstb-dependent gene-expression changes in mice and in this data set, we include gene-level expression data from cultured primary microglia of control and Cstb-/- mice extracted from neonatal mice at P5 and we identified 156 differentially-expressed genes in Cstb-/- microglia. We analyzed 8 samples in total and we compared 4 control microglia samples to 4 Cstb-/- microglia samples. Genes with a fold change > 1.3 and with p < 0.05 in T-test with BH correction were considered differentially expressed.
