This study aimed to determine the gene expression profile of these conditions, in addition to potential biomarkers for closed-cervix pyometra, the most severe condition Total RNA was extracted from 69 fresh endometrium samples collected from 21 healthy female dogs during diestrus, 16 CEH, 15 mucometra and 17 pyometra (eight open and nine closed-cervix). Global gene expression was detected using the Affymetrix Canine Gene 1.0 ST Array

Circulating microRNA profiling by array in dogs with myxomatous mitral valve degeneration (MMVD) and pulmonic stenosis (PS) compared with healthy dogs

Project description:In this study, we aimed to demonstrate expression profiles of circulating microRNAs (miRNAs) in dogs with eccentric or concentric cardiac hypertrophy, and investigate whether there is a difference in miRNA expression according to the type of cardiac hypertrophy. Dogs with myxomatous mitral valve degeneration (MMVD) or pulmonic stenosis (PS) were included in this study, which are the two representative diseases of eccentric or concentric cardiac hypertrophy in dogs, respectively. Circulating miRNAs were isolated from the serum samples of five dogs with MMVD, five dogs with PS, and five healthy dogs. The circulating miRNA expression levels of dogs with MMVD or PS were compared with those of the healthy dogs by microarray analysis (Affymetrix GeneChip miRNA 4.0), using two independent parameters, a fold change cut-off of > 1.5 (up or down regulation) and p-value of < 0.05.

Gene expression in canine uterine bacterial infection

Project description:The study was undertaken to analyze the effect of canine uterine bacterial infection (Pyometra) on local gene expression patterns in the uteri. The results show profound upregulation of various expected genes related to both the innate and innate immune system. In addition, there was a marked upregulation of many genes associated with proteolysis. Moreover, the bacterial disease resulted in downregulation of a large number of homeobox and zinc finger transcription factors. Effects of ad libitum feeding on canine skeletal muscle gene expression

Project description:Skeletal muscle, as a large and insulin sensitive tissue, is an important contributor to metabolic homeostasis and energy expenditure. Obese dogs exhibit skeletal muscle insulin resistance, but the causes of these changes are unclear. Thus, we used canine microarrays to analyze gene expression profiles of skeletal muscle tissue collected from obese dogs, after 24 wk of ad libitum feeding. Skeletal muscle tissue samples were collected from 9 intact female beagles (4 yr-old; 4 control; 5 ad libitum) after 24 wk of ad libitum feeding.

Affymetrix expression profile array data of miR-206 affected genes in left superior fat pads in canines

Project description:Mongrel dogs of either sex were randomly divided into 2 groups: 1) infection with control lentiviruses group (lenti-control, n = 6), in which control lentiviruses were injected into left superior fat pads;2) only infection with miR-206 overexpressed lentiviruses group (lenti-miR-206, n = 6), in which miR-206 lentiviruses were injected into left superior fat pads. We used microarrays to detail the global programme of mRNA expression underlying miR-206 overexpression and identified distinct classes of up-regulated and down-regulated genes during this process. Microarray-based mRNA expression profiling was performed of miR-206 overexpressed group and control group using the Affymetrix Array (CA, USA). Equal quantities of RNAs from 3 dogs were mixed to generate one sample for each experimental group.

Project description:RNA Seq data showed 49 differentially altered mRNAs in estrus and diestrus stages of buffalo cell free saliva, that includes 10 mRNAs commonly present in both estrus and diestrus stages, whereas 12 and 27 mRNAs were found to be exclusively present either at the estrus or the diestrus stage respectively. In addition, four mature miRNAs were predicted to be differentially altered in the estrus as compared to the diestrus phase of buffaloes.

Project description:The aim of the study was to analyse differences in serum and urinary proteomes between healthy dogs and dogs with renal dysfunction in babesiosis using label-based high-resolution quantitative proteomic approach. In the study 8 healthy control dogs (group H) and 22 dogs with naturally occurring babesiosis were enrolled. Dogs with babesiosis were divided into 3 groups: group A consisted of 6 non-azotaemic dogs (serum creatinine <140 μmol/L) with normal UPC (UPC < 0.5), group B of 10 non-azotaemic dogs with proteinuria (UPC > 0.5) and group C of 6 azotaemic dogs (serum creatinine > 140 μmol/L) and significant proteinuria (UPC > 2).

CEH-60/PBX regulates vitellogenesis and cuticle formation through interaction with UNC-62/MEIS in Caenorhabditis elegans

Project description:The onset of sexual maturity involves dramatic changes in physiology and gene expression in many animals. A textbook example is the production of enormous amounts of yolk proteins destined for the nascent oocytes, termed vitellogenesis. We previously identified the rapidly evolving PBC-class Hox-cofactor CEH-60/PBX as necessary for abundant vit transcription in C. elegans. Differential proteomics data shows that all vitellogenin proteins are indeed drastically downregulated in ceh-60 mutant animals and suggest an additional role for CEH-60’s involvement cuticle structure, innate immunity and stress response.
