Gene expression analysis of coronal pulp and apical pulp complexes

ABSTRACT: We performed this study to compare the gene expression pattern between coronal pulp and apical pulp complex There were no studies about gene expression of apical pulp complex tissue before. So We performed this study to confirm the gene expression of apical pulp complex tissues. Coronal pulp tissues were collected from 11 healthy subjects. Gene expression profiles were conducted using cDNA microarray analysis. Apical pulp complex tissues were collected from 18 healthy subjects. Gene expression profiles were conducted using cDNA microarray analysis.

Transcription profiling of human pulp and odontoblasts reponse to TGF-beta1

Project description:Transforming growth factor beta 1 (TGF-Î²1) is the most extensively studied growth factor in dentin-pulp complex, with pleiotropic effects on pulp response and healing. Our main objective was to analyze the expression profile of pulp tissue and odontoblasts, and the effects of TGF-Î²1 on these profiles in cultured human pulp and odontoblasts with a specific interest in the anti- and pro-inflammatory cytokines. Experiment Overall Design: Pulp tissues and odontoblasts were cultured for different time periods, and microarray was performed to both cultured and native samples to detect the effects of TGF-Î²1. Expression of various interleukins (IL) were confirmed by RT-PCR, and in +/- TGF-Î²1 treated pulps also by antibody array.

Comparative gene expression analysis of the human dental pulp in deciduous and permanent teeth

Project description:Human deciduous and permanent teeth exhibit different developmental processes, morphologies, histological characteristics and life cycles. In addition their pulp tissues react differently to external stimuli, such as the pulp sensitivity test, dental trauma and pulp therapy materials. These differences are attributable to their genetic backgrounds. Therefore the purpose of this study is to compare the differences of dental pulp in deciduous and permanent teeth. Pulp samples were obtained from permanent premolars (n=6, aged 11-14 years) and deciduous teeth (n=6, aged 11-14 years). Comparative cDNA microarrary analysis revealed several differences in gene expression between the deciduous and permanent pulp tissues. Each GSM record represents a pulp sample pooled from two teeth samples.

TAILS N-terminomics of the human dental pulp stroma and the odontoblast layer

Project description:We report the targeted analysis of the human dental pulp stroma and the odontoblast layer using the positional proteomics technique TAILS N-terminomics, which allowed us to capture both naturally blocked and unblocked protein N-termini, and to identify differences between e.g. the dental pulp proteomes of older and younger donors. Noteworthy, these differences were most pronounced in the odontoblast region. Note: This study is a follow-up on PXD002264.

Project description:We report the targeted analysis of the human dental pulp proteome and N-terminome using the positional proteomics technique TAILS, which allowed us to capture both naturally blocked and unblocked protein N-termini. Furthermore, by using a proteomically barely studied human tissue in combination with a non-shotgun proteomics approach, we were able to identify many so far missing proteins (PE level 2-4), and established together with our previously published studies on human erythrocytes (Lange et al. 2014) and platelets (Prudova et al. 2014), a general workflow suitable for the in-depth and high-throughput analysis of protein N-termini in human specimen, as envisioned by the Chromosome-centric Human Proteome Project.

Proteomic profile of human stem cells from dental pulp and periodontal ligament

Expression status of mRNA for sex hormone receptors in human dental pulp cells and the response to sex hormones in the cells

Project description:Objectives: Sex hormone receptors are reported to be present in human dental pulp (HDP) cells. The purpose of this study was to examine the biological significance of estrogen and androgen receptors (ER and AR, respectively) in HDP cells. Design: We isolated HDP cells expressing ER- and AR-mRNAs and investigated the expression status of the receptors and the response to sex hormones in the cells. Results: HDP cells expressing ER- and/or AR-mRNAs had the ability to form alizarin red S-positive nodules in which calcium and phosphorus were deposited in vitro and to differentiate into odontoblasts-like cells and dentin-like tissue in vivo. Individual clones isolated from HDP cells exhibited a different expression pattern of mRNA for ER and AR. Some clones expressed ERM-NM-1- and/or ERM-NM-2-mRNAs and the others coexpressed ER- and AR-mRNAs. Using the Ingenuity software, we found that 17M-NM-2-estradiol (E2) and dihydrotestosterone (DHT) could act directly on HDP cells through ER- or androgen signaling-mediated mechanisms. E2 or DHT stimulated the mRNA expression for genes related to odontogenesis of dentin-containing teeth and odontoblast differentiation, suggesting that ER and AR in HDP cells may be involved in dentinogenesis. Conclusions: Our findings provide new insights into the biological significance of sex hormone receptors in HDP cells. Gene expression profiles of human dental pulp cells derived from one male patient were compared between cells treated with 10^-6 M 5alpha-dihydrotestosterone and cells treated with vehicle. Each sample has one replicate. Gene expression profiles of human dental pulp cells derived from one female patient were compared between cells treated with 10^-9 M 17beta-estradiol and cells treated with vehicle. Each sample has one replicate.
