Transcription profiling of human mono or co-cultured HMEC-1 and HK-2 cells to identify the influence of microvascular endothelial cells on transcriptional regulation of proximal tubular epithelial cells

ABSTRACT: The effect of the presence of HMEC-1 cells on HK-2 cell gene expression was investigated. Cells were cultured in mono or co-culture on opposite sides of aluminium oxide filters, 0.2 ?µM pore size. These conditions provide a close proximity but not direct contact. RNA from HK-2 cells was assayed with Affymetrix HGU133 Plus 2 arrays. 3 Biological samples were used per group.

ORGANISM(S): Homo sapiens

In the renal cortex the peritubular capillary network and the proximal tubular epithelium cooperate in solute and water reabsorption, secretion, and inflammation. However, the mechanisms by which these two cell types coordinate such diverse functions remain to be characterized. Here we investigated the influence of microvascular endothelial cells on proximal tubule cells, using a filter-based, noncontact, close-proximity coculture of the human microvascular endothelial cell line HMEC-1 and the h  ...[more]
