Transcription profiling of mouse pituitary glands from wild type, Hesx1Cre/+;Ctnnb1GOF/+ and Hesx1Cre/+;Ctnnb1GOF/+;Rosa26Hesx1/+ animals

ABSTRACT: comparisons between wild type, Hesx1Cre/+;Ctnnb1GOF/+ and Hesx1Cre/+;Ctnnb1GOF/+;Rosa26Hesx1/+ 17.5 dpc pituitaries. RNA was extracted from anterior posterior and intermediate lobes using the RNeasy micro kit (Qiagen) with on-column DNaseI digestion. RNA was labeled using the Affymetrix two-cycle labeling protocol and hybridised to MOE430 2.0 arrays.

Transcription profiling of mouse small intestine from PlagL2-/- animals reveals transcription factor PlagL2 regulates steps in chylomicron metabolism

Project description:Enterocytes assemble dietary lipids into chylomicron particles that are taken up by intestinal lacteal vessels and peripheral tissues. Although chylomicrons are known to assemble in part within membrane secretory pathways, the modifications required for efficient vascular uptake are unknown. We report that the transcription factor Pleomorphic adenoma gene-like 2 (PLAGL2) is essential for this aspect of dietary lipid metabolism. PlagL2-/- mice die from post-natal wasting owing to failure of fat absorption. Lipids modified in the absence of PlagL2 exit from enterocytes but fail to enter interstitial lacteal vessels. Dysregulation of enterocyte genes closely linked to intracellular membrane transport identified candidate regulators of critical steps in chylomicron assembly. PlagL2 thus regulates essential and poorly understood aspects of dietary lipid absorption and its deficiency represents an authentic animal model with implications for amelioration of obesity or the metabolic syndrome. Experiment Overall Design: Total RNA was extracted from 4 knockout and 4 wild-type mouse small intestines at 18.5 dpc using the Macherey-Nagel Nucleospin kit. cRNA synthesis and labeling, hybridization to Affymetrix (Santa Clara, CA) MOE430 2.0 expression arrays, and data acquisition occurred on the Affymetrix GeneChip Instrument System.

Project description:To identify candidate genes regulated by Atx ablation, transcriptional profiles were generated using murine MOE430 2.0 oligonucleotide arrays. Total RNA from Atx+/+ and Atx-/- E9.5 C57BL/6 mouse embryos was analyzed using six separate arrays. 644 probe sets identified with reproducible ANOVA (p value <0.05, FDR p value <0.05 and mean actual intensity fold change ±1.5) expression changes for comparisons between Atx-/- and Atx+/+ embryos.

Project description:Despite a considerable literature concerning the molecular pathogenesis of pituitary tumors, the mechanisms of pituitary tumors development and progression remain unknown. Four SAGE cDNA libraries were constructed using a pool of mRNA obtained from five GH-, two ACTH-secreting, and four non secreting pituitary tumors (NS), and three normal pituitaries from patients who had accidental death, using I-SAGE kit (Invitrogen). The aim of this study was to evaluate the differential gene expression profile by SAGE genes in different subtypes of pituitary tumors to contribute for understanding of pituitary tumorigenesis. A potential impact of SARS-CoV-2 on pituitary glands and pituitary neuroendocrine tumors.

Project description:IntroductionAngiotensin-converting enzyme 2 (ACE2) is the receptor of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The effects of SARS-CoV-2 on normal pituitary glands function or pituitary neuroendocrine tumors (PitNETs) have not yet been elucidated. Thus, the present study aimed to investigate the potential risks of SARS-CoV-2 infection on the impairment of pituitary glands and the development of PitNETs.MethodsPitNETs tissues were obtained from 114 patients, and normal pituitary gland tissues were obtained from the autopsy. The mRNA levels of ACE2 and angiotensin II receptor type 1 (AGTR1) were examined by quantitative real-time PCR. Immunohistochemical staining was performed for ACE2 in 69 PitNETs and 3 normal pituitary glands. The primary tumor cells and pituitary cell lines (MMQ, GH3 and AtT-20/D16v-F2) were treated with diminazene aceturate (DIZE), an ACE2 agonist, with various dose regimens. The pituitary hormones between 43 patients with SARS-CoV-2 infection were compared with 45 healthy controls.ResultsPituitary glands and the majority of PitNET tissues showed low/negative ACE2 expression at both the mRNA and protein levels, while AGTR1 showed high expression in normal pituitary and corticotroph adenomas. ACE2 agonist increased the secretion of ACTH in AtT-20/D16v-F2 cells through downregulating AGTR1. The level of serum adrenocorticotropic hormone (ACTH) was significantly increased in COVID-19 patients compared to normal controls (p < 0.001), but was dramatically decreased in critical cases compared to non-critical patients (p = 0.003).ConclusionsThis study revealed a potential impact of SARS-CoV-2 infection on corticotroph cells and adenomas.
