Gene expression profiling of human mammary artery media during aging

Project description:Our goal was to better understand the genes and pathways implicated during arterial media aging. We therefore compared the gene expression profile of human mammary artery media between Young (<60, 8 patients) and old (>75, 7 patients) patients. The microarray data, generated using the Illumina sentrix Ref8-V2 platform, was normalized and analyzed using the IlluminaGUI package (Eggle and Schultze, 2007). We used 8 samples from the "Young" groups and 7 samples from the "old" group. No technical replicates were used.

Transcription profiling by array of human clonal primary fibroblasts from patients with CDKL5 mutations

Project description:Mutations in the human CDKL5 gene have been shown to cause infantile spasms, as well as Rett syndrome-like phenotype. Because CDKL5 is subjected to X chromosome inactivation (XCI), individual cells from CDKL5 mutation girls either express the wild-type or mutant allele, likely resulting in different consequences at both the cellular and molecular levels. To identify these consequences, we carried out gene expression profiling on clonal populations derived from primary cultures of three patientsM-^R fibroblasts expressing either allele. A total of 16 up-regulated and 20 down-regulated genes were identified. The differentially expressed gene products, mostly involved in differentiation and oxidative stress may be related to a mechanism underlying mental retardation and epilepsy. Among these differentially expressed proteins, the apoptosis signal-regulated kinase MAP3K5 expression was found to be altered in non-neuronal, but also in neuronal CDKL5 deficient cells. Due to the fact that MAP3K5 activates MAP kinase pathway, which mediates signals leading to both differentiation and survival in neuronal cells, we suggest that a CDKL5 deficit may induce changes in synaptic plasticity in patientM-^Rs brain.

Transcription profiling of rat heart after treatment with PETN or NTG

Project description:Male Wistar rats (6 weeks, 250g), (Charles River, Sulzfeld, Germany) were equipped with micro osmotic pumps model 2001 from Alzet (Cupertino, CA) containing 400mM PETN dissolved in DMSO or the solvent and infusion was maintained for four days at 1?l/h (10.5?g/kg/min). For direct comparison, rats were also infused with 450mM NTG (6.6?g/kg/min in ethanol or the solvent as a control). <br>After 4d rats were sacrificed by exsanguinations under Isofluran anesthesia (5% inhalant in room air), the heart was rapidly excised, placed in ice-cold Krebs-HEPES-solution (composition in g/l: 5.78 NaCl, 0.35 KCl, 0.37 CaCl2, 0.30 MgSO4, 2.1 NaHCO3, 0.14 K2HPO4, 5.21 HEPES and 2.0 D-glucose) and dissected.<br>Total RNA was isolated from the hearts. Direct labeled cDNA was synthezised and hybridized to Rat OpArray Microarray (total genom expression profiling).

Gene expression profile analysis of F2 cross between Fat and Lean chickens

Project description:The aim of this study was to identify genes involved in the variation of the muscle glycogen content at death (estimated through the glycolytic potential, GP), a determining factor in the meat quality of chicken. The gene expression profiles of the Pectoralis major muscle were established using microarrays. We compared animals showing extreme GP values, issued from a F2 cross between Fat and Lean lines. A total of 254 genes were differentially expressed between F2 birds with low or high GP. Several of these genes were validated by qRT-PCR. For the genes with human orthologs, annotation analyses were performed and mainly revealed pathways involved in carbohydrate, fatty-acid and protein metabolism. The relationship between gene expression and meat quality has to now be validated by further e-QTL studies on the F2 population. 8 high GP samples compared to 8 low GP samples, 4 of these were dye-swapped.

Biological replicates: 5 control replicates, 5 Losrtan-treated animals. All the RNA samples were pooled to provide a reference RNA for a two-color experiment.
