Transcription profiling of two human cellular models of erythroid differentiation: K562 erythroid-leukemia cells treated with 1uM arabinofuranosyl-cytosine (AraC), and CD34+ stem cells treated with 7U/ml erythropoietin

ABSTRACT: In this study we investigated two cellular models of erythroid differentiation: K562 erythroid-leukemia cells treated with 1uM arabinofuranosyl-cytosine (AraC), and CD34+ stem cells treated with 7U/ml erythropoietin. Time course experiments were performed in both analysed models. Total RNAs were isolated and used for probe synthesis and hybridization on Affymetrix human U133A arrays. We compared the two models of erythropoiesis regarding their functional similarity. Furthermore, we identified differentially expressed proteins in each model, assigned gene-ontology annotations and ordered them into clusters. Finally, we investigated which pathways were affected during the differentiation processes using GenMapp.
