Transcription profiling of Arabdiopsis plants over-expressing MKK3DD or MKK4DD to identify genes acting downstream of MAP kinase kinase

ABSTRACT: To identify genes acting downstream of MAP kinase kinase, and thus map its signal, we used the steroid-inducible overexpression system. The constitutive active form MKK3 (MKK3DD) or MKK4 (MKK4DD) was cloned into the glucocorticoid-inducible vector pTA7002 and transformed using Agrobacterium method into Arabidopsis Col-0 plants. The transgenic plants transformed empty vector was used as control plants (Cont).

The plant hormone jasmonic acid (JA) plays a key role in the environmental stress responses and developmental processes of plants. Although ATMYC2/JASMONATE-INSENSITIVE1 (JIN1) is a major positive regulator of JA-inducible gene expression and essential for JA-dependent developmental processes in Arabidopsis thaliana, molecular mechanisms underlying the control of ATMYC2/JIN1 expression remain largely unknown. Here, we identify a mitogen-activated protein kinase (MAPK) cascade, MAPK KINASE 3 (MKK  ...[more]
