RNA-seq of human brain microvascular endothelial (hBME) cells infected with Toxoplasma gondii RH tachyzoite for 6, 24 and 48 hours versus control

ABSTRACT: hBME cells were infected with T. gondii RH tachyzoites for 6, 24 and 48 hours at multiplicity of infection (MOI) of 2. At each time points, total RNA were extracted using Qiagen RNeasy MIni Kit.The quality and quantity of extracted RNA was determined using the Agilent® RNA 6000 Nano kit and 2100 Bioanalyser All samples with RQN (RNA quality number) values of 9.0 or above were processed for sequencing. Messenger RNA (mRNA) libraries were prepared using Illumina’s TruSeq Stranded mRNA HT Kit [RS-122-2103]. Sequencing was performed as 150 bp paired end reads on Illumina’s HiSeq4000 platform according to Illumina specifications. mRNA sequencing was performed by Oxford Genomics Centre, The Wellcome Trust Centre for Human Genetics, University of Oxford, United Kingdom.
