Sequencing of the transcriptome from embryonic and larval hemocytes in gcm knock down background

ABSTRACT: Hemocytes were sorted from stage 16 embryos and wandering 3rd instar larvae in which gcm was knocked down using gcmRNAi and the driver srp(hemo)Gal4. The experiments were done in triplicate. The wild type controls at stage 16 embryo and wandering larvae are available under this assession number: E-MTAB-8702.

Project description:The aim of this experiment is to compare the transcriptome of Drosophila hemocytes at two developmental stages: in stage 16 embryo, mostly free from any immune challenge and in larvae, which grow in bacteria rich environment. Transgenes inducing the expression of RFP in the hemocytes were used to sort the hemocytes from the two stages before carrying out the sequencing.

RNA-seq of Drosophila melanogaster neurons, glia and hemocytes at embryonic stage 16 and third instar wandering larvae

Project description:We produced bulk RNA sequencing data on Drosophila neurons, glia and hemocytes at embryonic and larval stages to extrapolate the most significant and specific features that identify a cell type as a whole. We also tracked changes in the expression in the two stages. We identified stable features that define a cell type functionally and developmentally as well as plastic features including stage-specific metabolic states, which likely depend on the environment.

Mis-expression of Tramtrack in Drosophila embryos

Project description:We profiled the transcriptome of Drosophila melanogaster embryos in ttk2D50 embryos or after over-expression using btl-GAL4; UAS-ttk, respectively. We further isolated cells that express btl-enh-RFPmoe (Cabernard and Affolter 2005) and FACS sorting, and profiled their transcriptomes in the same genetic backgrounds. A total of 19 samples were analysed with 2-3 replicates each. For each embryos and sorted cells, the genotypes were: ttk2D50 btl-enh-RFPmoe (mutant; embryos: 2, cells: 2), btl-enh-RFPmoe (mutant control; embryos: 2, cells: 3), btl-GAL4; UAS-ttk, btl-enh-RFPmoe (over-expression; embryos: 3, cells: 2), btl-GAL4; btl-enh-RFPmoe (over-expression control; embryos: 3, cells: 2).

Bulk RNAseq of Anopheles gambiae hemocytes upon Toll pathway overactivation

Project description:Toll pathway is a key mediator of antiplasmodial immunity and its mechanism of action is dependent on hemocytes (mosquito white blood cells). Anopheles gambiae were injected with dsRNA for Cactus, an inhibitor of the Toll pathway. Cactus silencing over activates the Toll pathway. The goal of this experiment is to evaluate hemocyte transcriptional changes between control mosquitoes injected with a non-related dsRNA (dsLacZ and dsCactus injected mosquitoes. We also divide hemocytes from control and experimental conditions into 2 groups: bound (hemocytes that attach to glass - granulocyte populations) and unbound (hemocytes that remain in suspension - prohemocytes and oenocytoids). Files from Unbound fractions are labelled: UB and from bound fractions are labelled B. Some of the samples were run twice and have 2 gz files.

From the fat body to the hemolymph: Profiling tick immune and storage proteins through transcriptomics and proteomics

Project description:Ticks are blood-feeding arachnids that are known to transmit various pathogenic microorganisms to their hosts. During blood feeding, ticks activate their metabolism and immune system to efficiently utilise nutrients from the host's blood and complete the feeding process. In contrast to insects, in which the fat body is known to be a central organ that controls essential metabolic processes and immune defence mechanisms, the function of the fat body in tick physiology is still relatively unexplored. To fill this gap, we sought to uncover the repertoire of genes expressed in the fat body associated with trachea (FB/Tr) by analyzing the transcriptome of individual, partially fed (previtellogenic) Ixodes ricinus females . The resulting catalog of individual mRNA sequences reveals a broad repertoire of transcripts encoding proteins involved in nutrient storage and distribution, as well as components of the tick immune system. To gain a detailed insight into the secretory products of FB/Tr specifically involved in inter-tissue transport and humoral immunity, the transcriptomic data were complemented with the proteome of soluble proteins in the hemolymph of partially fed female ticks. Among these proteins, the hemolipoglyco-carrier proteins were predominant. When comparing immune peptides and proteins from the fat body with those produced by hemocytes, we found that the fat body serves as a unique producer of certain immune components. Finally, time-resolved transcriptional regulation of selected immune transcripts from the FB/Tr was examined in response to experimental challenges with model microbes and analysed by RT-qPCR. Overall, our data show that the fat body of ticks, similar to insects, is an important metabolic tissue that also plays a remarkable role in immune defense against invading microbes. These findings improve our understanding of tick biology and its impact on the transmission of tick-borne pathogens.

SWI/SNF complex controls self-renewal and differentiation in stem cell lineages but how this function relates to tumorigenesis is currently unclear. Here, we use Drosophila neuroblasts to demonstrate that the SWI/SNF component Osa (ARID1) prevents tumorigenesis in stem cell lineages by ensuring correct unidirectional lineage progression. Ecdysone and Mediator trigger a metabolic switch uncoupling cell cycle from cell growth to end proliferation in Drosophila neural stem cells

Project description:Stem cells are highly abundant and proliferate rapidly during early development but become a rare population in most adult organs. The molecular mechanisms causing stem cells to exit proliferation at a specific time are not well understood. Here, we show that changes in energy metabolism induced by the steroid hormone Ecdysone initiate an irreversible cascade of events leading to cell cycle exit in Drosophila neural stem cells. We show that the timely induction of oxidative phosphorylation and the mitochondrial respiratory chain are required in neuroblasts to uncouple cell cycle progression from cell growth. This results in a progressive reduction in neuroblast cell size and ultimately in terminal differentiation. Neuroblasts isolated from brain tumors fail to undergo this shrinkage process and this may explain why they are immortalized. Our findings show that cell size control can be modified by systemic hormonal signaling and reveal a unique connection between metabolism and proliferation control in stem cells. Regulation of Drosophila brain development and organ growth by the Minibrain/Rala signaling network

Project description:Using affinity purification-mass spectrometry (AP-MS), we identified the novel Mnb binding partners Ral interacting protein (Rlip) and RALBP1 associated Eps domain containing (Reps). Rlip and Reps physically and genetically interact with Mnb, and the three proteins may form a ternary complex. Mnb phosphorylates Reps, and human DYRK1A binds to the Reps orthologs REPS1 and REPS2. Furthermore, Mnb engages the small GTPase Ras-like protein A (Rala) to regulate brain and wing development. This work uncovers a previously unrecognized early role of Mnb in the neuroepithelium and defines the functions of the Mnb/Reps/Rlip/Rala signaling network in brain development.
