Effect of obesity on genome methylation of lung carcinoma cells grown as subcutaneous tumors in young male C57BL/6 mice

ABSTRACT: Luciferase-expressing Lewis lung carcinoma (LLC-Luc/LL-2; ATCC CRL-1642-LUC2) or KPN1.1 lung carcinoma (DOI 0.1016/j.crmeth.2021.100080) cell-lines were grown as subcutaneous tumors after injection in syngeneic male C57BL/6 of ~6 months age that had been on normal or high-fat diet for ~4 months and which they were kept on during the rest of the experiment. About 19-29 days later, tumors were harvested to isolate the carcinoma cells by flow cytometry to extract DNA/RNA from for genome methylation and transcriptome RNA sequencing. All LLC-Luc samples were from one mouse experiment, all but one (sample 12) of the KPN1.1 samples were from the same mouse experiment. DNA isolation and array experiments were performed in one batch for all samples.
