Transcriptional profiling of visual stimulation-activated mouse retinal cells using RNA-Seq

ABSTRACT: We investigated the retinal transcriptional response to visual stimulation using phosphorylated ribosomal protein S6 immunoprecipitation (pS6-IP), a method to enrich RNA expressed in activated cells. Mice were exposed to moving, high-contrast vertical gratings for twelve hours per day over a period of two weeks. At the same time, control mice were placed in an environment with stationary white walls. At the end of the experiment, the neural retina was dissected and homogenized, followed by pS6-IP, extraction of immunoprecipitated RNA, and RNA-seq.
