Transcriptional profile of small intestinal organoids from C57BL/6 (WT) and Opa1KO mice

ABSTRACT: Small intestinal organoids generated from Opa1KO mice and Opa1fl/fl. Organoids were generated from young adult mice and stimulated in vitro with tamoxifen 4-oht for 3 days. Opa1KO organoids are smaller and more round than their WT counterparts. Organoids were collected and their transcriptome was extracted for RNAseq.

Project description:Small intestine organoids generated from young adult mice were passaged once on day 3. Medium was changed again on day 7 and, one day later, organoids were stimulated for 20 hours with either 25ng/ml IFN-β (R&D Systems), or 25ng/ml IFN-γ (BioLegend) or with 200ng/ml IFN-λ2 (PeproTech) or were left untreated (unstimulated group).

RNA-seq analysis of human forebrain organoids irradiated with moderate to high doses of ionizing radiation

Project description:Prenatal irradiation can cause neurodevelopmental defects which are characterized by a reduction in brain size (microcephaly). The underlying molecular mechanisms in humans have so far not been studied. Here, we leveraged human forebrain organoids as a model for human embryonic brain development to investigate time- and dose-dependent effects of radiation on organoid growth. For this, organoids of 14 days and 56 days old were irradiated with acute X-ray doses of 0.5 Gy or 2 Gy and compared to controls. Using bulk RNA-seq at different early (6 h and 24 h) and late (14 days) time points after irradiation, we investigated mechanisms of radiation-induced growth defects which resulted from activation of the DNA damage response (cell cycle arrest, DNA repair, apoptosis), premature differentiation and the coordinated repression of primary microcephaly genes.

RNA-Seq of human spinal cord organoids generated in Matrigel and alginate

Project description:This experiment compares the transcriptome profile of human spinal cord organoids generated from iPSCs in Matrigel, 1% alginate hydrogel, or 2% alginate hydrogel. Spinal cord organoids were generated from human iPSCs and then encapsulated in the respective hydrogels before further maturation. Organoids were harvested on days 30, 60, and 90 for each group. In addition to the differences between hydrogel groups, neuronal and glial markers at each time point were also assessed for the development of the organoids.
