Single-cell RNA sequencing of PBMCs from five healthy (HC) preterm infants and ten preterm infants with bronchopulmonary dysplasia(BPD)

ABSTRACT: We procured PBMCs whole blood from five HC preterm infants and five preterm infants with BPD. PBMCs were extracted using a density gradient centrifugation method. Initially, 10ml of peripheral blood was mixed with an equal volume of physiological saline, then carefully layered onto Ficoll solution (T10124, from Shangbao Biotech Co., Ltd., Shanghai). After centrifugation at 2,000 rpm for 20 minutes, the cells stratified due to differences in density, with PBMCs positioned between the red blood cells and plasma. Subsequently, the intermediate layer containing PBMCs was gently collected, washed several times with physiological saline to remove residual medium and red blood cells, and finally, PBMCs were isolated and collected through centrifugation.

Preterm birth increases endogenous human milk peptide release compared to term birth

Project description:Hundreds of naturally occurring milk peptides have been found in term human milk. Whether there are differences between the levels of these peptides between term and preterm milk remains unknown. Premature milk is produced before complete maturation of the mammary gland and is under the influence of a different hormonal milieu, which could change enzymatic activity and protein expression within the mammary gland and result in an altered peptide profile. We employed nano-liquid chromatography tandem mass spectrometry to identify naturally occurring peptides in term and premature milks at multiple time-points across lactation and compare the abundances of these peptides. We found that preterm milks produced more unique peptide sequences than term milks on average (359 vs. 286). The peptides identified were searched for overlapping sequences with an in-house database of known functional peptides. Specifically, we found that both term and preterm milks contain peptides overlapping with known sequences with antimicrobial, opioid antagonist and immunomodulatory actions. We also compared the enzymes involved in degradation of both milks via bioinformatic analysis. This analysis reveals that plasmin is more active in preterm milk than term milk.

The gene expression profile of peripheral blood mononuclear cells (PBMCs) of infants (6 to 11 months) immunized with an inactivated EV71 vaccine or placebo

Project description:To study the gene expression after administrating an inactivated EV71 vaccine, we have employed whole human genome microarray expression profiling to emphasize the gene expression profile of the immune system in response to an inactivated EV71 vaccine in humans and confirmed that such an immune response was generated as the result of the positive mobilization of the immune system. Human peripheral blood from this vaccine or placebo was isolated, extracted the RNA and screened by microarray. The results showed significantly greater neutralizing antibody and specific T cell responses in vaccine group after two inoculations on days 0 and 28. Additionally, more than 600 functional genes that were up- or down-regulated in PBMCs were identified by the microarray assay, and these genes included 68 genes associated with the immune response in vaccine group. The vaccine inoculation group and the placebo control group, originated from the cohort of phase II clinical trial, consisted of 20 subjects (aged from 6-11 months, average: 9.6 months). The trial was randomized, double-blinded and placebo-controlled, and all of the parents or guardians of the subjects were required to sign an informed consent explaining the experiment.

Temporal development of necrotizing enterocolitis in the preterm pig

Project description:Necrotizing enterocolitis (NEC), a serious gastrointestinal disease that afflicts 5-10% of preterm infants, often progresses rapidly from mild food intolerance into extensive haemorrhage, inflammation and necrosis. Events leading to NEC have remained poorly defined. Similar disease characteristics are observed in preterm pigs 24-48 h after feeding formula. Using this model, we aimed to characterize the temporal development of NEC, and describe the functional and immunological response of the preterm intestine preceding NEC. Keywords: time course Pigs from treatment groups TPN (n=5), and 8 h (n=5) and 24 h (n=5-6) FORM and COLOS were randomly selected for microarray analysis. Equal amounts of total distal small intestinal RNA from all pigs was pooled to make the reference sample. Samples and reference pool were labelled with Oyster 550 and 650, respectively. The in-house spotted porcine oligonucleotide microarray version 4 (POM4) is a low density microarray consisting of 384 different oligonucleotide probes representing more than 200 different immune related genes and eight different array control oligonucleotides (ArrayControl; Ambion, NÃ¦rum, Denmark). The immunologically relevant 60-70mer oligonucleotide probes represent interferons and interleukins (and receptors), chemokines (and receptors), acute phase proteins, apoptosis-related factors and sequences with relevance to Toll-like receptors and their intracellular signalling pathways.

Proteomics analysis of stool samples from preterm neonates with mass spectrometry for predicting necrotizing enterocolitis

Project description:This study aimed to investigate the potential of fecal proteomics signatures for NEC prediction. Stools were collected from NEC infants up to 10 days before their diagnosis and matched with those from non-NEC controls for three tested periods. Stool samples were analyzed with liquid chromatography-tandem mass spectrometry using SWATH acquisition and a cross-compatible proteomic software platform to perform label-free quantification. ROC curves and principal component analysis were used to explore discriminating information provided by potential markers and sets of markers were evaluated with classical clustering and classification methods to evaluate their capacity to sort the samples giving to their actual outcomes. We identified a specific series of markers that predicted all NEC from non-NEC infants at least a week in advance. Our study shows that multiplexed proteomic signature detection constitutes a favorable approach for the early detection of NEC.

Allergen Shedding in Human Milk from Mothers of Preterm Infants: Proteomic and Peptidomic Feasibility and Pilot Analysis

Project description:There is little information regarding the allergen content of milk feeds in the preterm population. Previous studies have evaluated specific proteins/peptides via ELISA, but no studies have performed a broad analysis of the allergenic peptide content and protease activity of milk feeds in this population. Preterm infants spend a critical window of time for immune development in the Newborn Intensive Care Unit (NICU), and may receive fortified donor milk, maternal milk or formula feeds via nasogastric tube or bottle instead of fresh breastmilk via breastfeeding.

Amniotic fluid and necrotizing enterocolitis in preterm pigs

Project description:Preterm neonates are susceptible to gastrointestinal (GI) disorders such as necrotizing enterocolitis (NEC). Maternal milk, and especially colostrum, protects against NEC via growth promoting, immunomodulatory and antimicrobial factors. The fetal enteral diet, amniotic fluid (AF), contains similar bioactive components and we hypothesized that postnatal AF administration would reduce inflammatory responses and NEC in preterm neonates. Thirty preterm pigs (92% gestation) were delivered by caesarean section and fed total parental nutrition (TPN) for 48 h followed by enteral porcine colostrum (COLOS, n=7), infant formula (FORM, n=13) or formula + porcine AF (AF, n=10). Using a previously validated model of NEC in preterm pigs, we determined the structural, functional, microbiological and immunological responses to AF when administered prior to and after introduction of a suboptimal enteral formula diet. Gene expression patterns in 34 decidua samples of women in term and preterm labour

Project description:This study evaluates the gene expression patterns in 36 decidual samples of women having gone through different types of child birth. The samples were profiled on Illumina Human HT-12_v4 BeadArrays. The analysis includes quality control and exploratory analysis of the dataset, followed by the identification of differentially expressed genes (DEGs) and finally, functional analysis for enrichment of KEGG pathways and GO terms amongst the DEGs. The statistical analysis was performed for six comparisons: Term labour (TL) vs Term not labour (TNL), TL vs Preterm labour (PTL), TL vs Preterm non-labour (PTNL), TNL vs PTL, TNL vs PTNL and PTL vs PTNL. This study evaluates the gene expression levels from decidual samples obtained from women giving birth with different types of labour. The dataset was composed of 36 Illumina Human HT-12_v4 expression BeadArrays.
