Transcription profiling by array of homozygous-bred gastric intrinsic factor knockout mice before and after Citrobacter rodententium infection

ABSTRACT: Knockout mice were generated that harbour a mutation in the gene encoding gastric intrinsic factor (GIF) a transporter protein necessary for the absorption of vitamin B12/cobalamin (Cbl) from the intestine. The levels of Cbl in the blood of these gif-/- mice exibited a maternal influence dependent on whether they were bred from heterozygous (high Cbl) or homozygous (low Cbl) mothers and this correlated with their susceptibility to infection challenge with Citrobacter rodententium (C.rodentium). Microarrray analysis was conducted on total messenger RNA (mRNA) extracted fron the homzygous-bred gif-/- and wild-type mice with the aim to identify any abnormal genomic expression patterns.
