Characterization of yap7 in Saccharomyces cerevisiae and the human pathogen Candida glabrata

ABSTRACT: This study aimed at characterizing the targets of the bZIP transcription factor Yap7 in two yeast species: Saccharomyces cerevisiae and the human pathogen Candida glabrata. Transcriptome analyses were thus conducted in wild type and yap7 knock out strains using Agilent microarrays.

The labelled cDNA from treated cells were competitively hybridized on microarrays versus cDNA from mock treated cells.

We performed these experiments in two different growth conditions: glycerol as sole carbon source (as Hap4 is supposed to be involved in the induction of respiratory genes in non-fermentative growth conditions) or glucose with iron excess (2 mM FeSO4) as Hap5 is supposed to be involved in the high iron response independently of Hap4.
