The needles were isolated by year emergence (whorl) in four groups. 0 (2012), 1 (2011), 2 (2010) and 3 (2009). The samples were harvested at 1245 m of altitude at Los Reales de Sierra Bermeja (Spain) (30S X:303.095 Y:4.039.618) one time per month. Reference design. 6 biological replicates that were pooled two by two resulting in 3 samples for the hybridization. The common reference samples was a pool of RNA samples from January. The reference sample was labeled with Cy5 and the test samples with Cy3.

Microarray based molecular marker detection

Project description:Fungal resistant and susceptible maize genotypes were subjected to Aspergillus flavus spore inoculation and kernels around the infected area were collected 4 days after inoculation. Uninoculated kernels were also collected at 4 days. Microarray experiment was performed to compare the transcriptional profiles of the different genotypes and interpret the genes involved in the associated resistance of the individual genotype to further characterize them as potential molecular markers for resistance.

Project description:Arabidopsis Col-0 seedlings grown in liquid culture to growth stage 1.0 were treated with 5 M-5M Benzyladenine and incubated for the indicated time points. Roots and shoots of the seedlings were then harvested separately by taking each individual seedling with a soft forceps at the hypocotyl, dipping it into liquid nitrogen and breaking off the root at the inner wall of a collection tube that was swimming in liquid nitrogen. The shoot part was then striped from the forceps at the edge of another collection tubes.
