MicroRNA profiling by array of paired samples tumor tissue/normal adjacent tissue from colon cancer patients

ABSTRACT: Ten arrays were performed on the RNA extracts from 10 patients' samples, each of them contained the paired samples tumor tissue/ normal adjacent tissue.

Project description:Breast malignancy is a serious threat to women’s health around the world. However, for invasive, ductal carcinomas, and mammary fibroadenoma, there is an urgent demand for better breast malignancy-linked biomarkers. The current study was designed to identify diagnostic and/or therapeutic protein biomarkers for breast tumors. iTRAQ proteomic analysis was employed to characterize differentially expressed protein biomarkers. We identified several potential protein biomarkers that can be recommended for further testing as diagnostic and/or therapeutic targets in breast tumors.

MicroRNA profile differences in Hepatitis B virus (HBV) positive and negative hepatocellular carcinoma

Project description:MicroRNAs (miRNAs) exhibit essential regulatory functions related to cell growth, apoptosis, development and differentiation. Dysregulated expression of miRNAs is associated with a wide variety of human diseases. As such miRNA signatures are valuable as biomarkers for disease and for making treatment decisions. Hepatitis B virus (HBV) is a major risk factor for hepatocellular carcinoma (HCC). Here we screened for miRNAs in chronic HBV associated HCC. To evaluate the effect of HBV infection on the change in expression of miRNAs, 12 pairs of samples from HCC and non-tumor tissues (including 6 HBV-positive HCC and 6 HBV-negative HCC and their non-tumor tissues) were collected. The extracted RNAs were evaluated to detect the expression of miRNAs. Using ANOVA to screen the differential expression of miRNAs at P-value â¤ 0.01, fold change â¥ 2 or â¤ 0.5, 225 miRNAs were detected.

The expression of microRNAs in embryoid bodies derived from mouse embryonic stem cells

Project description:Mouse embryonic stem cells can spontaneously differentiate and assemble into a spherical embryoid body (EB) during suspension culture. The initial study aims to identify the up-regulated or down-regulated microRNAs during the differentiation process of pluripotent stem cells. From the microRNA profiling, we will focus on the microRNAs associated with mesodermal and endothelial differentiation of embryonic or induced pluripotent stem cells. In this study, the EBs were collected at 0 and 10-day differentiation of ESCs (ATCC: CRL-1934). The miRNA transcripts listed in Sanger miRBase Release 19.0 was detected by microarray.
