RNA-seq of chicken spleens after LPS injection

ABSTRACT: The acute phase response is poorly studied in chickens, but its examination can possibly supply valuable markers for the identification of subclinical disease and assessment of disease severity. We therefore injected ten six weeks-old Lohmann Selected Leghorn chickens i.v. with 10µg LPS per kg body weight (n=5) or the same volume of PBS as controls (n=5) and collected spleens after three hours. Spleens were kept in ice cold PBS and RNA was isolated immediately. After quality control of RNA (RIN>9.0), a cDNA library was prepared and submitted to Illumina high-throughput sequencing.
