Meiotic crossover frequency varies within genomes, which influences genetic diversity and adaptation. In turn, genetic variation within populations can act to modify crossover frequency in cis and trans. Project description:Genomic DNA from 180 Col/Bur F2 individuals, and 186 taf4b/Bur F2 individuals, was extracted by CTAB and used to generate sequencing libraries as previously described (Ziolkowski et al, 2017 Genes & Dev). Sequencing data was analysed to identify crossovers using the TIGER pipeline as previously described (Rowan et al, 2015 G3 (Bethesda); Yelina et al, 2015 Genes & Dev).

Therefore R3327-AT1 Dunning prostate carcinoma and Walker-256 mammary carcinoma cells were incubated at pH 7.4 and 6.6 for 24 and the miRNA expression was analyzed by NGS.

Differentially expressed genes analysis between Eupolyphaga sinensis Walker (ESW) and Taspine derivate

Project description:To gain insights into the mechanisms of Eupolyphaga sinensis Walker (ESW) and taspine derivate on inhibition to HCC, we have employed whole genome microarray expression profiling as a discovery platform to identify different genes between ESW-treated sample and control. At the same time, the differences were investigated between taspine derivate and the same control. SMMC-7721 cells were cultivated in the absence or presence of 0.1mg/mL ESWE and Taspine derivate of 2.5x10-5mol/L for 48 h, followed by the Agilent Whole Human Genome Oligo Microarray. The PLG, PKCM-NM-2 and IL3RA genes confirmation of Microarray analysis was confirmed by Real-time PCR. Transcription profiling by array of Aedes aegypti infected with Wolbachia strains wMelPop and wMel

Project description:Wolbachia is a vertically transmitted intracellular bacteria that infect most than 60% of insect species. The strains wMelPop and wMel were introduced in the dengue virus vector Aedes aegypti, naturally not infected by Wolbachia. Recently, it was shown that those two strains inhibit dengue virus replication into their new host, A. aegypti (Moreira et al. 2009 and Walker et al. in preparation). The aim of this project is to look at the transcriptional response of Aedes aegypti to infection with wMel and wMelPop and try to find some genes or pathway potentially involved in the viral interference.Four laboratory lines of A. aegypti were used throughout this study. The PGYP1 and Mel2 lines were generated by transinfection with wMelPop and wMel strains respectively. PGYP1.tet and Mel2tet lines were treated with the antibiotic tetracycline and cured from Wolbachia infection (McMeniman et al., 2009 and Walker et al in preparation). The Mosquitoes were reared under standard laboratory conditions (26 ± 2 °C, 12:12 light/dark cycle, 75% relative humidity). Mosquito larvae were fed 0.1mg/larvae of TetraMin Tropical Tablets once a day. Adults were transferred to cages (measuring 30 x 30 x 30 cm) at emergence at 400 individuals per cage. Adults were supplied with a basic diet of 10% sucrose solution (Turley et al., 2009).

Project description:NLRC5 is a member of the NLR family of proteins. The observation that NLRC5 is found in the nucleus prompted us to perform a gene array to identify putative target genes of NLRC5. We generated Jurkat T cell lines that stably express either the wild-type or mutant forms of NLRC5 harboring mutations in the nucleotide binding domain (NBD): Walker A (deficient in nucleotide binding), Walker B (deficient in nucleotide hydrolysis), and the combined Walker AB, carrying both mutations. Site-directed mutagenesis was used to create the NLRC5 NBD mutants: Walker A (K234A), Walker B (E311Q), and Walker AB (both mutations).
