Systematic analysis of gene expression in human brains before and after death.

Project description:BACKGROUND: Numerous studies have employed microarray techniques to study changes in gene expression in connection with human disease, aging and evolution. The vast majority of human samples available for research are obtained from deceased individuals. This raises questions about how well gene expression patterns in such samples reflect those of living individuals. RESULTS: Here, we compare gene expression patterns in two human brain regions in postmortem samples and in material collected during surgical intervention. We find that death induces significant expression changes in more than 10% of all expressed genes. These changes are non-randomly distributed with respect to their function. Moreover, we observe similar expression changes due to death in two distinct brain regions. Consequently, the pattern of gene expression differences between the two brain regions is largely unaffected by death, although the magnitude of differences is reduced by 50% in postmortem samples. Furthermore, death-induced changes do not contribute significantly to gene expression variation among postmortem human brain samples. CONCLUSION: We conclude that postmortem human brain samples are suitable for investigating gene expression patterns in humans, but that caution is warranted in interpreting results for individual genes.

Gene expression in primate postnatal brain through lifespan - prefrontal cortex

Project description:We investigated molecular changes during human, chimpanzee, and rhesus macaque postnatal brain development at the transcriptome, proteome, and metabolome levels in two brain regions: the prefrontal cortex (PFC) that is involved in several human-specific cognitive processes, and the cerebellar cortex (CBC) that may be functionally more conserved. We find a nearly three-fold excess of human-specific gene expression changes in PFC compared to CBC. The most prominent human-specific mRNA expression pattern in the PFC is a developmental delay of approximately 5 years in the expression of genes associated with learning and memory, such as synaptic transmission and long-term potentiation. This pattern is supported by correlated changes in concentrations of proteins and the respective neurotransmitters and its magnitude is beyond the shift expected from the life-histories of the species. Mechanistically, it might be driven by change in timing of expression of four or more transcription factors. We speculate that delayed synaptic maturation in PFC may play a role in the emergence of human-specific cognitive abilities. MicroRNA expression in primate postnatal brain through lifespan

Project description:While multiple studies have reported the accelerated evolution of brain gene expression in the human lineage, the mechanisms underlying such change remain poorly understood. Here we address this issue from a developmental perspective, by analyzing mRNA and microRNA (miRNA) expression in two brain regions within macaques, chimpanzees and humans throughout their lifespan. We find that developmental profiles of trans-regulators, such as miRNA, as well as their target genes, show the fastest rates of human-specific evolutionary change. Changes in expression of a few key regulators may be a major driving force behind human brain evolution. Role of omega-3 fatty acids regulating the brain functions and associated disorders: Transcriptomic analysis of the effect of a fish oil enriched diet on murine brains

Project description:Based on the comprehensive inhibition of a pool of inflamamsome and apoptosome networks, FD likely reduces the amyloidal burden, attenuates oxidative stress and assist somatostatin activation. Thus FD is a potential therapeutic agent for Alzheimer’s disease, Parkinson’s disease and affective disorder A total of 10 mice were randomly chosen from a strain of C57BL/6j.Those mice consumed two diets customized n-3 PUFA enriched diet (FD) and standard lab diet (SD )from weaning age until 5 month. Hemi brain samples were harvested from these mice and Transcriptomic Expression Analysis, Cluster and Functional analysis, Real time polymerized chain reaction and assay to validate microarray results were done to collect data. A comparative analysis of the diet compositions reveals relatively comparable amount of major nutrients and consumable calories derived from both diets .The major difference between the two diet types emerges from the percentage of n-3 PUFA and n-6 PUFA present in each type .
