Transcription profiling of Fusarium head blight resistance in Sumai-3 and two susceptible NILs

Project description:Microarray gene expression analysis were performed to identify genes responding to Fusarium graminearum inoculation and genes that show a differential regulation between the resistant c.v. Sumai-3 and the susceptible near isogenic lines. Sumai-3 is a Chinese wheat cultivar, the most commonly used source of resistance to Fusarium head blight (FHB) disease. NIL-3 and NIL-4 are near isogenic lines susceptible to FHB and are derived from a cross between Sumai-3 and Chuan980, a susceptible wheat cultivar, followed by seven generations of marker assisted backcrossing with the recurrent parent Sumai-3 and selection for FHB susceptibility in each generation by artificial inoculation with F. graminearum. A combination of DNA markers including simple sequence repeats (SSRs) and amplified fragment length polymorphisms (AFLPs) were used to screen for DNA polymorphisms between Sumai-3 and its susceptible NILs. Polymorphic markers were mapped using a mapping population. The analysis indicated 7 polymorphisms between the susceptible NILs and the resistant Sumai-3. The analysis indicated that NIL-3 differs from Sumai-3 at chromosome 3BS and NIL-4 differs from Sumai-3 at chromosome 2AL region.

Project description:Infectious pancreatic necrosis (IPN) is a serious viral disease that causes significant economic losses in salmon aquaculture. To characterize the host-pathogen relationship in IPN, we analysed transcriptional profiles of salmon head kidney (SHK-1) cells infected with infectious pancreatic necrosis virus (IPNV) at three timepoints over six days (at 1, 3 & 6 days post infection. The transcriptome was investigated using the TRAITS / SGP 16950-feature Atlantic salmon cDNA microarray, which is enriched for genes with functions related to the immune response.

Transcription profiling of human CD4+ leukemia Jurkat T-cells in which the PI3K pathway is constitutively turned-on and FOXO1 transcriptional activity strongly repressed

Project description:One major effect of PI3-kinase activation downstream of the serine/threonine kinase Akt is the phosphorylation of the transcription factor FOXO1 and its neutralization. FOXO1 has several ubiquitous targets genes in many cell types that control cell quiescence, oxydative stress or apoptosis. However, it has been demonstrated that FOXO1 also has specific targets depending of the cellular context. The role of FOXO1 to regulate specific genes in T lymphocytes has not been investigated yet. To examine this point, we used the CD4+ leukemia Jurkat T-cell line, in which the PI3K pathway is constitutively turned-on and FOXO1 transcriptional activity strongly repressed. These cells were transduced with lentiviruses coding for a constitutively active form of FOXO1 fused to GFP and having the three AKT phosphorylation sites mutated to alanine (FOXO1-AAA-GFP) to restore its transcriptional activity. GFP-transduced cells were used as a control and the gene activation levels in the two cell populations analyzed 48 hours post-infection.
