Synergistic Antibacterial Activity of Benzalkonium Bromide and Cu-Bearing Duplex Stainless Steel against Pseudomonas aeruginosa.

ABSTRACT: The bactericide benzalkonium bromide is widely used to kill Pseudomonas aeruginosa, which causes microbiologically influenced corrosion (MIC). However, the extensive use of benzalkonium bromide will enhance bacterial drug resistance and cause environmental pollution. In this study, benzalkonium bromide combined with Cu-bearing 2205 duplex stainless steel (2205-Cu DSS) was used to kill Pseudomonas aeruginosa; the germicidal rate of the combination of benzalkonium bromide and 2205-Cu DSS was 24.2% higher than that of using benzalkonium bromide alone, after five days. The antibacterial efficacy was evaluated using an antibacterial test and biofilm observation. The results showed that, in the presence of P. aeruginosa, the combination of 23.44 ppm benzalkonium bromide and 2205-Cu DSS showed the best antibacterial efficacy.

Project description:Friction taper stitch welding (FTSW) is a novel technique that uses multiple inserts to conceal surface crack in a given substrate. The inserts are rotated and forced to fill the crack as plasticized material, and forge with the substrate in solid-state. The process is well suited for alloys such as duplex stainless steel, which suffers degradation of properties during fusion welding. A detailed experimental and theoretical investigation is presented here on FTSW of a duplex stainless steel (DSS). The experimental results show the presence of a ferrite-rich phase along the interface. The results computed by the numerical process model reveal a direct influence of thermal cycle in the amount of ferrite along the joint interface. The welded joint shows near homogeneous structure and properties similar to those of the substrate.

Cu-bearing stainless steel reduces cytotoxicity and crystals adhesion after ureteral epithelial cells exposing to calcium oxalate monohydrate.

Project description:Calcium oxalate monohydrate (COM), which is the main component of encrustation, may result in cell membrane injury. In addition, cellular damage is suggested to be the primary event attributing to COM crystal binding. To study the interaction between cells and crystals after incubating with a Cu-bearing stainless steel (316L-Cu SS), MTS and flow cytometric analyses were used to assess the cellular responses. The results confirmed that 316L-Cu SS could inhibit cytotoxicity and cellular apoptosis of ureteral epithelial cells (UECs) after COM treatment. Furthermore, molecular expressions of Cu/Zn superoxide dismutase (CuZnSOD), which were evaluated by western blot analysis and real-time quantitative PCR (qPCR), indicated that 316L-Cu SS could inhibit the oxidative stress attributing to up-regulating of CuZnSOD. Moreover, the crystal adhesion cytokine CD44 was examined with western blot and qPCR, and the corresponding hyaluronic (HA) secreted into the medium was measured by enzyme-linked immunosorbent assay (ELISA). All results were confirmed that the expressions of cells cultured with 316L-Cu SS were down-regulated, demonstrating the inhibitory performance of 316L-Cu SS against crystal adhesion.

Microbiologically Influenced Corrosion of 2707 Hyper-Duplex Stainless Steel by Marine Pseudomonas aeruginosa Biofilm.

Project description:Microbiologically Influenced Corrosion (MIC) is a serious problem in many industries because it causes huge economic losses. Due to its excellent resistance to chemical corrosion, 2707 hyper duplex stainless steel (2707 HDSS) has been used in the marine environment. However, its resistance to MIC was not experimentally proven. In this study, the MIC behavior of 2707 HDSS caused by the marine aerobe Pseudomonas aeruginosa was investigated. Electrochemical analyses demonstrated a positive shift in the corrosion potential and an increase in the corrosion current density in the presence of the P. aeruginosa biofilm in the 2216E medium. X-ray photoelectron spectroscopy (XPS) analysis results showed a decrease in Cr content on the coupon surface beneath the biofilm. The pit imaging analysis showed that the P. aeruginosa biofilm caused a largest pit depth of 0.69 μm in 14 days of incubation. Although this was quite small, it indicated that 2707 HDSS was not completely immune to MIC by the P. aeruginosa biofilm.

Surface and Electrochemical Performance Variations of 2205 Duplex Stainless Steel in Multistep Surface Processing.

Project description:This study aims to elucidate the impact of different surface processing techniques on the corrosion resistance of 2205 duplex stainless steel. The samples were subsequently treated by mechanical grinding (Sgrd), water-sandpaper sanding (Swap), and mechanical polishing (Spol) and characterized by XRD, AFM, XPS, and a series of electrochemical tests. Due to the grinding-induced residual compressive stress and low donor density in the passive film, Sgrd exhibited the lowest corrosion current among the three. Nevertheless, the lattice defects and the high content of Cr(III) hydroxide not only weakened the stability of the passive film but also resulted in the decreased trend of corrosion resistance. Sanding and mechanical polishing with a lower power input removed the residual stress left by grinding and inhibited the occurrence of selective oxidation. The lower surface roughness facilitated the formation of homogeneous passive layers with fewer defects, which can enhance the pitting resistance and provide a gradually enhanced protection from corrosion.

Micro-deformation evolutions of the constituent phases in duplex stainless steel during cyclic nanoindentation.

Project description:Cyclic elastoplastic deformation behaviors of austenite phase and ferrite phase in a duplex stainless steel were investigate by load-controlled cyclic nanoindentation with a Berkovich indenter. During the tests, the maximum penetration depth per cycle increased rapidly with cycle number at transient state, and reached stable at quasi-steady state. Plastic dissipated energy was quantitatively proved to be the driving force for the propagation of deformation zones during cyclic nanoindentation tests. Transmission electron microscopy combined with FIB was used to reveal the deformation mechanisms of both phases underneath indents with cycles. After quasi-static single loading, nucleation and concentration of dislocations were observed in both austenite phase and ferrite phase under the indenter. After cyclic loading, dislocations propagated to further regions in both phases. Besides, slip bands were generated within single nanoindentation and propagated during the subsequent cyclic nanoindentation. The sizes of the deformation regions for both phases under the indents after cyclic indentation observed by TEM were consistent with those calculated by the expansion model of spherical cavity.

Electrodepositing Ag on Anodized Stainless Steel for Enhanced Antibacterial Properties and Corrosion Resistance.

Project description:Antibacterial stainless steels have been widely used in biomedicine, food, and water treatment. However, the current antibacterial stainless steels face challenges in balancing corrosion resistance and antibacterial effectiveness, limiting their application range and lifespan. In this study, an oxide layer sealed with antibacterial Ag particles was constructed on the surface of 304 stainless steel through anodizing and electrodeposition, and the process parameters were optimized for achieving long-term antibacterial properties. The electrochemical tests demonstrated that the composite coating effectively enhanced the corrosion resistance of 304 stainless steel. The X-ray photoelectron spectroscopy analysis revealed the close binding mechanism between the Ag particles and the micropores in the oxide layer. Furthermore, the antibacterial stainless steel has an antibacterial rate of 99% against Escherichia coli (E. coli) and good biocompatibility. This study provides an effective approach for designing efficient, stable, and safe antibacterial stainless steel.

Real Time Imaging of Deuterium in a Duplex Stainless Steel Microstructure by Time-of-Flight SIMS.

Project description:For more than one century, hydrogen assisted degradation of metallic microstructures has been identified as origin for severe technical component failures but the mechanisms behind have not yet been completely understood so far. Any in-situ observation of hydrogen transport phenomena in microstructures will provide more details for further elucidation of these degradation mechanisms. A novel experiment is presented which is designed to elucidate the permeation behaviour of deuterium in a microstructure of duplex stainless steel (DSS). A hydrogen permeation cell within a TOF-SIMS instrument enables electrochemical charging with deuterium through the inner surface of the cell made from DSS. The outer surface of the DSS permeation cell exposed to the vacuum has been imaged by TOF-SIMS vs. increasing time of charging with subsequent chemometric treatment of image data. This in-situ experiment showed evidently that deuterium is permeating much faster through the ferrite phase than through the austenite phase. Moreover, a direct proof for deuterium enrichment at the austenite-ferrite interface has been found.

Tribocorrosion Failure Mechanism of TiN/SiOx Duplex Coating Deposited on AISI304 Stainless Steel.

Project description:TiN/SiOx duplex coatings were synthesized on AISI304 stainless steel by plasma immersion ion implantation and deposition (PIIID) followed by radio frequency magnetron sputtering (RFMS). The microstructure and tribocorrosion failure behaviors of the duplex coatings were investigated by X-ray diffraction, X-ray photoelectron spectroscopy, scanning electron microscopy, atomic force microscopy, reciprocating-sliding tribometer, and electrochemical tests. The as-deposited duplex coating had a two-layered columnar growth structure consisting of face-centered cubic TiN and amorphous SiOx. Sliding tests showed that the TiN interlayer had good adhesion with the substrate, but the SiOx layer suffered from severe delamination failure. Friction force induced a number of micro-cracks in the coating, which provided channels for the diffusion of NaCl solution. The tribocorrosion test showed that the duplex coating exhibited a lower wear-performance in NaCl solution than in ambient atmosphere. Multi-scale chloride ion corrosion occurred simultaneously and substantially degraded the bonding strength of the columnar crystals or neighboring layers. Force-corrosion synergy damage eventually led to multi-degradation failure of the duplex coating. The presented results provide a comprehensive understanding of the tribocorrosion failure mechanism in coatings with duplex architecture.

SCC of 2304 Duplex Stainless Steel-Microstructure, Residual Stress and Surface Grinding Effects.

Project description:The influence of surface grinding and microstructure on chloride induced stress corrosion cracking (SCC) behavior of 2304 duplex stainless steel has been investigated. Grinding operations were performed both parallel and perpendicular to the rolling direction of the material. SCC tests were conducted in boiling magnesium chloride according to ASTM G36; specimens were exposed both without external loading and with varied levels of four-point bend loading. Residual stresses were measured on selected specimens before and after exposure using the X-ray diffraction technique. In addition, in-situ surface stress measurements subjected to four-point bend loading were performed to evaluate the deviation between the actual applied loading and the calculated values according to ASTM G39. Micro-cracks, initiated by grinding induced surface tensile residual stresses, were observed for all the ground specimens but not on the as-delivered surfaces. Loading transverse to the rolling direction of the material increased the susceptibility to chloride induced SCC. Grinding induced tensile residual stresses and micro-notches in the as-ground surface topography were also detrimental.
