Project description:Advanced breast cancer is characterised by enhanced tumour adaptability to therapeutic pressure and the metastatic microenvironment. Targeting epi-transcriptomic modulators may reverse cellular adaptability, offering new therapeutic strategies to treat metastatic disease. This study looks into the dynamic adaptations that occur in cancer cells in response to therapeutic pressure and metastatic evolution by profiling mRNA epi-transcriptomic modifications in models of disease progression. ER positive (ER+) cell models were used to represent progressive breast cancer, MCF7 (endocrine sensitive), LY2 (endocrine resistant) and T347 (derived from an ER-positive, treatment resistant brain metastatic patient tumour) cells. MeRIP sequencing was undertaken to determine genome-wide RNA methylated regions.

Transcriptome differences in 14 primary breast tumour samples (n = 14 samples) are uncovered through this comparative RNA-seq analysis of patients that responded well to therapy (n = 7) and patients who had disease recurrence on endocrine treatment (n = 7). RNA sequencing data is deposited as log2 transformed median-of-ratios (DESeq2) normalised counts gene expression values (44934 genes IDs; n = 14 tumour samples).

FGFR1 underlies obesity-associated progression of estrogen receptor-positive breast cancer after estrogen deprivation.

Project description:Obesity increases breast cancer mortality by promoting resistance to therapy. Here, we identified regulatory pathways in estrogen receptor-positive (ER-positive) tumors that were shared between patients with obesity and those with resistance to neoadjuvant aromatase inhibition. Among these was fibroblast growth factor receptor 1 (FGFR1), a known mediator of endocrine therapy resistance. In a preclinical model with patient-derived ER-positive tumors, diet-induced obesity promoted a similar gene expression signature and sustained the growth of FGFR1-overexpressing tumors after estrogen deprivation. Tumor FGFR1 phosphorylation was elevated with obesity and predicted a shorter disease-free and disease-specific survival for patients treated with tamoxifen. In both human and mouse mammary adipose tissue, FGF1 ligand expression was associated with metabolic dysfunction, weight gain, and adipocyte hypertrophy, implicating the impaired response to a positive energy balance in growth factor production within the tumor niche. In conjunction with these studies, we describe a potentially novel graft-competent model that can be used with patient-derived tissue to elucidate factors specific to extrinsic (host) and intrinsic (tumor) tissue that are critical for obesity-associated tumor promotion. Taken together, we demonstrate that obesity and excess energy establish a tumor environment with features of endocrine therapy resistance and identify a role for ligand-dependent FGFR1 signaling in obesity-associated breast cancer progression.

Targeting the USP7-CDK1 axis suppresses estrogen receptor-positive breast cancer progression.

Project description:Estrogen receptor-positive breast cancer (ERPBC) accounts for approximately 70% of breast cancers in women worldwide. The therapeutic strategy process for ERPBC is well-established and significantly reduces the mortality rate. The discovery of new therapeutic targets remains essential for ERPBC patients with metastasis or endocrine resistance. This study indicated that USP7 is highly expressed in ERBPC and promotes tumor progression and metastasis. Inhibition of USP7 activity repressed proliferation, induced apoptosis, suppressed migration and invasive activities, and reversed the epithelial-mesenchymal transition of ERPBC. Mass spectrometry analysis indicated that USP7 regulates CDK1 expression, which is highly expressed and correlates with a poor overall survival rate in ERPBC. USP7 directly interacts with CDK1 and regulates its stability. The combined inhibition of USP7 and CDK1 by GNE-6776 and Ro-3306 synergistically represses the malignant process and metastasis of ERPBC. These findings proved that targeting USP7 and CDK1 is a potential strategy for overcoming endocrine resistance in patients with advanced ERPBC.

A Functional Survey of the Regulatory Landscape of Estrogen Receptor-Positive Breast Cancer Evolution.

Project description:Only a handful of somatic alterations have been linked to endocrine therapy resistance in hormone-dependent breast cancer, potentially explaining ∼40% of relapses. If other mechanisms underlie the evolution of hormone-dependent breast cancer under adjuvant therapy is currently unknown. In this work, we employ functional genomics to dissect the contribution of cis-regulatory elements (CRE) to cancer evolution by focusing on 12 megabases of noncoding DNA, including clonal enhancers, gene promoters, and boundaries of topologically associating domains. Parallel epigenetic perturbation (CRISPRi) in vitro reveals context-dependent roles for many of these CREs, with a specific impact on dormancy entrance and endocrine therapy resistance. Profiling of CRE somatic alterations in a unique, longitudinal cohort of patients treated with endocrine therapies identifies a limited set of noncoding changes potentially involved in therapy resistance. Overall, our data uncover how endocrine therapies trigger the emergence of transient features which could ultimately be exploited to hinder the adaptive process. Significance: This study shows that cells adapting to endocrine therapies undergo changes in the usage or regulatory regions. Dormant cells are less vulnerable to regulatory perturbation but gain transient dependencies which can be exploited to decrease the formation of dormant persisters.
