Guanidine Acetic Acid Alters Tissue Bound Amino Acid Profiles and Oxidative Status in Finishing Pigs.

ABSTRACT: This study aims to investigate the effects of guanidine acetic acid (GAA) on carcass traits, plasma biochemical parameters, tissue antioxidant capacity, and tissue-bound amino acid contents in finishing pigs. Seventy-two 140-day-old (body weight 86.59 ± 1.16 kg) crossbred pigs (Duroc × Landrace × Large White) were randomly assigned into four treatments with six replicate pens and three pigs per pen, which were fed the basal diets supplemented with 0, 0.05%, 0.10%, or 0.15% GAA, respectively. The plasma glucose concentration decreased, and creatine kinase activity and levels of GAA and creatine increased with the dietary GAA concentration. GAA linearly improved creatine content in the longissimus thoracis muscle (LM) and heart. The activities of superoxide dismutase, total antioxidant capacity, and glutathione peroxidase increased linearly in tissue or/and plasma, while the contents of malondialdehyde and protein carbonyl decreased linearly. GAA improved the contents of multiple-bound amino acids (such as proline or isoleucine) in the myocardium and LM. In conclusion, GAA enhanced the plasma biochemical parameters, oxidative status, and bound amino acid profiles of the heart and LM in finishing pigs.

Project description:BackgroundWith increasing health awareness among consumers, the demand for healthier, tastier, higher quality and nutritional value pork is increasing. It has been shown that different dietary starch sources can alter the carcass traits and meat quality. However, research on the effects of different starch sources with clear different amylose/amylopectin ratio on the amino acid and fatty acid composition in Longissimus thoracis (L. thoracis) muscle of pigs is limited. This study aimed to investigate the effects of different dietary starch sources on carcass traits, meat quality, muscle amino acid and fatty acid composition, and the mRNA expression levels of genes involved in lipid metabolism and muscle fiber characteristics in finishing pigs. A total of 72 Duroc × Landrace × Large White barrows were randomly allocated to 3 different dietary treatment groups with 8 replicate pens/group and 3 pigs per pen. Tapioca starch (TS), corn starch (CS), and pea starch (PS), with amylose/amylopectin ratio of 0.11, 0.25, and 0.44, respectively, were used as their dietary starch sources for 40 days.ResultsResults showed that the PS diet significantly increased (P < 0.05) the final body weight, average daily gain, loin-eye area, and fat-free lean index compared with the TS diet, but significantly decreased (P < 0.05) the feed to gain ratio and backfat thickness. Compared with the TS diet, PS diet also increased (P < 0.05) the pH45 min, marbling scores, the content of intramuscular fat and inosine monophosphate in the L. thoracis, and decreased (P < 0.05) the drip loss and shear force. In addition, compared with the TS diet, PS diet increased (P < 0.05) the proportions of flavor amino acids, DHA, EPA, and n-3 polyunsaturated fatty acid (PUFA) in the L. thoracis compared with TS diet, but decreased (P < 0.05) the ratio of n-6/n-3 PUFA. Furthermore, compared with the TS diet, PS diet also upregulated (P < 0.05) the lipogenic genes (FAS, LPL, SCD, ACCα) and myosin heavy-chain (MyHC)-IIa mRNA expression levels compared with the TS diet, but downregulated (P < 0.05) the CPT1B and MyHC-IIb mRNA levels.ConclusionsIn conclusion, these results provided compelling evidence that the different dietary starch source altered the carcass traits, meat flavor and quality in finishing pigs, and consumption of a diet with higher amylose/amylopectin ratio results in the production of a healthy, higher quality, and nutritional value pork.

Project description:This study was conducted to investigate the effect of protease inclusion level in two different ages on the apparent (AID) and standardized (SID) ileal digestibility of crude protein (CP) and amino acids (AAs) in soybean meal (SBM) fed to growing-finishing pigs. Ten cannulated pigs (21 ± 2 kg) were assigned to experimental diets in a duplicate 5 × 5 Latin square design. In phase I (23 to 30 kg-pigs, 90 ± 17 d of age), ileal digesta was collected in five periods of 7 d (5 d adaptation and 2 d ileal digesta collection). In phase II, (50 to 65 kg-pigs, 140 ± 17 d of age), ileal digesta was collected in 5 more periods of 7 d. For both phases, a corn starch-based diet was formulated with SBM as the sole source of CP and AA and containing titanium as an indigestible marker. Protease was supplemented at 0, 15,000, 30,000, and 45,000 NFP/kg of feed (0, 25, 50, and 75 g/ton of ProAct 360). A nitrogen-free diet was used to estimate basal ileal endogenous AA losses. Pigs were fed at 4% of their body weight, which was adjusted at the end of each period. Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of dietary protease supplementation in each phase. In phase I, increasing levels of protease resulted in a linear increase (P < 0.10) in SID for the 7/11 indispensable AA (Except Arg, His, Met + Cys, and Trp) and the average of all dispensable AA. In phase II, the SID of Ile, Leu, Met, Met + Cys, Val, the average of all indispensable AA, and 4/7 dispensable AA were quadratically increased (P < 0.10). In most cases, supplementation with 30,000 NFP/kg of feed (50 g/ton) resulted in the greatest increase in AA digestibility. However, the linear response in phase I for some AA suggests that diets for younger pigs could be supplemented with a greater level (45,000 NFP/kg or 75 g/ton of feed). Interestingly, younger pigs had consistently increased (P < 0.10) SID of CP and 15/18 AA (Except Arg, Cys, and Ser), being ~5.6% greater for indispensable AA when compared to older pigs. In conclusion, dietary protease supplementation can increase the SID of AA in SBM in both growing and finishing periods. Pig age can potentially influence AA digestibility, possibly related to a greater ileal endogenous AA flow in younger pigs. However, this fact warrants further investigation.
