Project description:A plasmon-enhanced fluorescence-based antibody-aptamer biosensor - consisting of gold nanoparticles randomly immobilized onto a glass substrate via electrostatic self-assembly - is described for specific detection of proteins in whole blood. Analyte recognition is realized through a sandwich scheme with a capture bioreceptor layer of antibodies - covalently immobilized onto the gold nanoparticle surface in upright orientation and close-packed configuration by photochemical immobilization technique (PIT) - and a top bioreceptor layer of fluorescently labelled aptamers. Such a sandwich configuration warrants not only extremely high specificity, but also an ideal fluorophore-nanostructure distance (approximately 10-15 nm) for achieving strong fluorescence amplification. For a specific application, we tested the biosensor performance in a case study for the detection of malaria-related marker Plasmodium falciparum lactate dehydrogenase (PfLDH). The proposed biosensor can specifically detect PfLDH in spiked whole blood down to 10 pM (0.3 ng/mL) without any sample pretreatment. The combination of simple and scalable fabrication, potentially high-throughput analysis, and excellent sensing performance provides a new approach to biosensing with significant advantages compared to conventional fluorescence immunoassays.

Project description:A predictive model is proposed that quantitatively describes the synergistic behavior of the electrical conductivities of CNTs and graphene in CNT:graphene hybrids. The number of CNT-to-CNT, graphene-to-graphene, and graphene-to-CNT contacts is calculated assuming a random distribution of CNTs and graphene particles in the hybrids and using an orientation density function. Calculations reveal that the total number of contacts reaches a maximum at a specific composition and depends on the particle sizes of the graphene and CNTs. The hybrids, prepared using inkjet printing, are distinguished by higher electrical conductivities than that of 100% CNT or graphene at certain composition ratios. These experimental results provide strong evidence that this approach involving constituent element contacts is suitable for investigating the properties of particulate hybrid materials.

Project description:Highly active and stable bimetallic Au-Pd catalysts have been extensively studied for several liquid-phase oxidation reactions in recent years, but there are far fewer reports on the use of these catalysts for low-temperature gas-phase reactions. Here we initially established the presence of a synergistic effect in a range of bimetallic Au-Pd/CeZrO4 catalysts, by measuring their activity for selective oxidation of benzyl alcohol. The catalysts were then evaluated for low-temperature WGS, CO oxidation, and formic acid decomposition, all of which are believed to be mechanistically related. A strong anti-synergy between Au and Pd was observed for these reactions, whereby the introduction of Pd to a monometallic Au catalyst resulted in a significant decrease in catalytic activity. Furthermore, monometallic Pd was more active than Pd-rich bimetallic catalysts. The nature of the anti-synergy was probed by several ex situ techniques, which all indicated a growth in metal nanoparticle size with Pd addition. However, the most definitive information was provided by in situ CO-DRIFTS, in which CO adsorption associated with interfacial sites was found to vary with the molar ratio of the metals and could be correlated with the catalytic activity of each reaction. As a similar correlation was observed between activity and the presence of Au0* (as detected by XPS), it is proposed that peripheral Au0* species form part of the active centers in the most active catalysts for the three gas-phase reactions. In contrast, the active sites for the selective oxidation of benzyl alcohol are generally thought to be electronically modified gold atoms at the surface of the nanoparticles.

Project description:Crucial to effective optical trapping is the ability to precisely control the nature of force/potential to be attractive or repulsive. The nature of particles being trapped is as important as the role of laser parameters in determining the stability of the optical trap. In this context, hybrid particles comprising of both dielectric and metallic materials offer a wide range of new possibilities due to their tunable optical properties. On the other hand, femtosecond pulsed excitation is shown to provide additional advantages in tuning of trap stiffness through harnessing optical and thermal nonlinearity. Here we demonstrate that (metal/dielectric hybrid) core/shell type and hollow-core type nanoparticles experience more force than conventional core-type nanoparticles under both continuous-wave and, in particular, ultrafast pulsed excitation. Thus, for the first time, we show how tuning both materials properties as well as the nature of excitation can impart unprecedented control over nanoscale optical trapping and manipulation leading to a wide range of applications.

Project description:Phenylboronic acids can form tight covalent bonds with diol-containing biomolecules. In this work, water-dispersed bifunctional gold nanoparticles (AuNPs) modified with ferrecene (Fc)-derivatized peptides and 4-mercaptophenylboronic acids (MBA) (denoted as Fc-MBA-AuNPs) were synthesized and characterized by UV/vis spectrometry and transmission electron microscopy. To demonstrate the application and the analytical merits of the nanoparticles in biosensing, glycoprotein avidin was tested as a model analyte. Specifically, avidin was captured by the biotin-covered gold electrode via the strong biotin-avidin interaction. Then, Fc-MBA-AuNPs were attached by the captured avidin through the formation of tight covalent bonds between the boronic acid moieties of Fc-MBA-AuNPs and the oligosaccharides of avidin. As a result, a detection limit of 0.2 pM was achieved. We believe that the bifunctional nanoparticles would found many applications in amplified detection of diol-containing species by rational design of the surface chemistry of electrode.

Project description:Gold(II) complexes are rare, and their application to the catalysis of chemical transformations is underexplored. The reason is their easy oxidation or reduction to more stable gold(III) or gold(I) complexes, respectively. We explored the thermodynamics of the formation of [AuII (L)(X)]+ complexes (L=ligand, X=halogen) from the corresponding gold(III) precursors and investigated their stability and spectral properties in the IR and visible range in the gas phase. The results show that the best ancillary ligands L for stabilizing gaseous [AuII (L)(X)]+ complexes are bidentate and tridentate ligands with nitrogen donor atoms. The electronic structure and spectral properties of the investigated gold(II) complexes were correlated with quantum chemical calculations. The results show that the molecular and electronic structure of the gold(II) complexes as well as their spectroscopic properties are very similar to those of analogous stable copper(II) complexes.

Project description:Biosensors for the determination of glucose concentration have a great significance in clinical diagnosis, and in the food and pharmaceutics industries. In this research, short-chain polyaniline (PANI) and polypyrrole (Ppy)-based nanocomposites with glucose oxidase (GOx) and 6 nm diameter AuNPs (AuNPs(6 nm)) were deposited on the graphite rod (GR) electrode followed by the immobilization of GOx. Optimal conditions for the modification of GR electrodes by conducting polymer-based nanocomposites and GOx were elaborated. The electrodes were investigated by cyclic voltammetry and constant potential amperometry in the presence of the redox mediator phenazine methosulfate (PMS). The improved enzymatic biosensors based on GR/PANI-AuNPs(6 nm)-GOx/GOx and GR/Ppy-AuNPs(6 nm)-GOx/GOx electrodes were characterized by high sensitivity (65.4 and 55.4 μA mM-1 cm-2), low limit of detection (0.070 and 0.071 mmol L-1), wide linear range (up to 16.5 mmol L-1), good repeatability (RSD 4.67 and 5.89%), and appropriate stability (half-life period (τ1/2) was 22 and 17 days, respectively). The excellent anti-interference ability to ascorbic and uric acids and successful practical application for glucose determination in serum samples was presented for GR/PANI-AuNPs(6 nm)-GOx/GOx electrode.

Project description:Vitrimers represent a new class of smart materials. They are covalently crosslinked like thermosets, yet they can be reprocessed like thermoplastics. The underlying mechanism is the rapid exchange reactions which form new bonds while breaking the old ones. So far, heating is the most widely used stimulus to activate the exchange reaction. Compared to heating, light not only is much more convenient to achieve remote and regional control, but can also offer fast healing. Gold nanospheres are excellent photothermal agents, but they are difficult to disperse into vitrimers as they easily aggregate. In this paper, we use polydopamine to prepare gold nanospheres. The resultant polydopamine-coated gold nanospheres (GNS) can be well dispersed into epoxy vitrimers, endowing epoxy vitrimers with light responsivity. The composites can be reshaped permanently and temporarily with light at different intensity. Efficient surface patterning and healing are also demonstrated.

Project description:As a kind of excellent photoluminescent material, carbon quantum dots have been extensively studied in many fields, including biomedical applications and optoelectronic devices. They have been dispersed in polymer matrices to form luminescent films which can be used in LEDs, displays, sensors, etc. Owing to the total internal reflection at the flat polymer/air interfaces, a significant portion of the emitted light are trapped and dissipated. In this paper, we fabricate free standing flexible PVA films with photoluminescent carbon quantum dots embedded in them. We disperse silica microspheres at the film surfaces to couple out the total internal reflection. The effects of sphere densities and diameters on the enhancement of photoluminescence are experimentally investigated with a homemade microscope. The enhancement of fluorescence intensity is as high as 1.83 when the film is fully covered by spheres of 0.86 [Formula: see text]m diameter. It is worth noting that the light extraction originates from rather the scattering of individual spheres than the diffraction of ordered arrays. The mechanism of scattering is confirmed by numerical simulations. The simulated results show that the evanescent wave at the flat PVA/air interface can be effectively scattered out of the film.
