Complex Energy Landscapes of Self-Assembled Vesicles.

ABSTRACT: The field of supramolecular chemistry has witnessed tremendous progress in bringing the system away from equilibrium for traditionally inaccessible structures and functions. Vesicular assemblies with complex energy landscapes and pathways, which are reminiscent of diverse cellular vesicles like exosomes, remain exceedingly rare. Here, relying on the activation of oligo(ethylene glycol) (OEG) interdigitation and the encoded conformational freedom in monodisperse Janus dendrimers, we reveal a rich landscape and a pathway selection of distinct vesicles. The interdigitation can be selectively switched on and off using temperature ramps, and the critical temperatures can be further determined by molecular design. Our findings suggest that synthetic vesicles, with different energy states and unexpected transition pathways, emulate dynamic cellular vesicles in nature. We anticipate that vesicles with an activated OEG corona conformation will open new routes for nanomedicine and advanced materials.

Project description:High performance organogel polyelectrolytes were synthesized by super acid catalyst step-growth polycondensation of isatin and the non-activated multiring aromatic p-terphenyl. Subsequently, a chemical modification reaction was carried out to obtained quaternary ammonium functionalized polyelectrolytes through a nucleophilic substitution reaction with (3-bromopropyl)trimethylammonium bromide and potassium carbonate at room temperature. Different functionalization degrees were obtained by controlling the molar ratio of the polymer and the modification agent. The organogel polyelectrolytes were formed due to the high phase segregation and self-assembling observed owing to the amphiphilic character of the material (hydrophobic backbone and hydrophilic fragment grafted). The organogel polyelectrolytes were used to fabricate supercapacitors using two commercial graphite electrodes. These polyelectrolytes displayed good ionic conductivity without the use of another doping agent such as salts, acids or ionic liquids. In this work, a strong correlation of functionalization degree and ionic conductivity of the polyelectrolytes and capacitance of the supercapacitors was observed. The ionic conductivity of the polyelectrolytes reached 0.46 mS cm-1 for the 100% functionalization degree, meanwhile the polyelectrolyte with the 10% functionalization degree shows 0.036 mS cm-1. Li-doped polyelectrolytes showed higher ionic conductivity due the presence of extra ionic charges (2.26 and 0.2 mS cm-1 for the polyelectrolytes with the 100% and 10% of functionalization degree, respectively). The principal novelty of this work lies in the possibility of modulating the ionic conductivity of organogels and the capacitance of supercapacitors by chemical modifications. The capacitance of the supercapacitors was 1.17 mF cm-2 for the 100% functionalized polyelectrolyte and is higher in comparison with the polyelectrolyte with 10% functionalization degree (0.68 mF cm-2) measured at a discharge current of 52 μA cm-2 by galvanostatic charge discharge technique. Additionally, when lithium salt (lithium triflate) was added, the polyelectrolytes retained a gel consistency, increasing the ionic conductivity and capacitance. For the doped polyelectrolytes, the areal capacitance reaches 1.37 mF cm-2 for the 100% functionalization degree polyelectrolyte with lithium triflate. These organogel polyelectrolytes open the possibility to design flexible and all solid-state supercapacitors without the risk of leakage.

Project description:Effective delivery of luminescent probes for cell imaging requires both cell membrane permeation and directing to discrete target organelles. Combined, these requirements can present a significant challenge for metal complex luminophores, that have excellent properties as imaging probes but typically show poor membrane permeability. Here, we report on highly luminescent Ruthenium polypyridyl complexes based on the parent; [Ru(dpp)2(x-ATAP)](PF6)2 structure, where dpp is 4,7-diphenyl-1,10-phenanthroline and x-ATAP is 5-amino-1,10-phenanthroline with pendant alkyl-acetylthio chains of varying length; where x is 6; 5-Amido-1,10-phenanthroline-(6-acetylthio-hexanyl). 8; 5-Amido-1,10-phenanthroline-(8-acetylthio-octanyl). 11; 5-Amido-1,10-phenanthroline-(11-acetylthio-undecanyl); and 16; 5-Amido-1,10-phenanthroline-(16-acetylthio-hexadecanyl). Soluble in organic media, the alkyl-acetylthiolated complexes form nanoaggregates of low polydispersity in aqueous solution. From dynamic light scattering the nanoaggregate diameter was measured as 189 nm and 135 nm for 5 × 10-6 M aqueous solutions of [Ru(dpp)2(N∧N)](PF6)2 with the hexadecanoyl and hexanyl tails respectivly. The nanoaggregate exhibited dual exponential emission decays with kinetics that matched closely those of the [Ru(dpp)2(16-ATAP)]2+ incorporated into the membrane of a DPPC liposome. Cell permeability and distribution of [Ru(dpp)2(11-ATAP)]2+ or [Ru(dpp)2(16-ATAP)]2+ were evaluated in detail in live HeLa and CHO cell lines and it was found from aqueous media, that the nanoaggregate complexes spontaneously cross the membrane of mammalian cells. This process seems, on the basis of temperature dependent studies to be activated. Fluorescence imaging of live cells reveal that the complexes localize highly specifically within organelles and that organelle localization changes dramatically in switching the pendent alkyl chains from C16 to C11 as well as on cell line identity. Our data suggests that building metal complexes capable of self-assembling into nano-dimensional vesicles in this way may be a useful means of promoting cell membrane permeability and driving selective targeting that is facile and relatively low cost compared to use of biomolecular vectors.
