Methylsiloxanes from Vehicle Emissions Detected in Aerosol Particles.

ABSTRACT: Methylsiloxanes have gained growing attention as emerging pollutants due to their toxicity to organisms. As man-made chemicals with no natural source, most research to date has focused on volatile methylsiloxanes from personal care or household products and industrial processes. Here, we show that methylsiloxanes can be found in primary aerosol particles emitted by vehicles based on aerosol samples collected in two tunnels in São Paulo, Brazil. The aerosol samples were analyzed with thermal desorption-proton transfer reaction-mass spectrometry (TD-PTR-MS), and methylsiloxanes were identified and quantified in the mass spectra based on the natural abundance of silicon isotopes. Various methylsiloxanes and derivatives were found in aerosol particles from both tunnels. The concentrations of methylsiloxanes and derivatives ranged 37.7-377 ng m^-3, and the relative fractions in organic aerosols were 0.78-1.9%. The concentrations of methylsiloxanes exhibited a significant correlation with both unburned lubricating oils and organic aerosol mass. The emission factors of methylsiloxanes averaged 1.16 ± 0.59 mg kg^-1 of burned fuel for light-duty vehicles and 1.53 ± 0.37 mg kg^-1 for heavy-duty vehicles. Global annual emissions of methylsiloxanes in vehicle-emitted aerosols were estimated to range from 0.0035 to 0.0060 Tg, underscoring the significant yet largely unknown potential for health and climate impacts.

Project description:BackgroundEpidemiological evidence supports the association between exposure to ambient particulate matter (PM) and cardiovascular diseases. Chronic exposure to ultrafine particles (UFP; Dp <100 nm) is reported to promote atherosclerosis in ApoE knockout mice. Atherogenesis-prone factors induce endothelial dysfunction that contributes to the initiation and progression of atherosclerosis. We previously demonstrated that UFP induced oxidative stress via c-Jun N-terminal Kinases (JNK) activation in endothelial cells. In this study, we investigated pro-inflammatory responses of human aortic endothelial cells (HAEC) exposed to UFP emitted from a diesel truck under an idling mode (UFP1) and an urban dynamometer driving schedule (UFP2), respectively. We hypothesize that UFP1 and UFP2 with distinct chemical compositions induce differential pro-inflammatory responses in endothelial cells.ResultsUFP2 contained a higher level of redox active organic compounds and metals on a per PM mass basis than UFP1. While both UFP1 and UFP2 induced superoxide production and up-regulated stress response genes such as heme oxygenease-1 (HO-1), OKL38, and tissue factor (TF), only UFP2 induced the expression of pro-inflammatory genes such as IL-8 (2.8 +/- 0.3-fold), MCP-1 (3.9 +/- 0.4-fold), and VCAM (6.5 +/- 1.1-fold) (n = 3, P < 0.05). UFP2-exposed HAEC also bound to a higher number of monocytes than UFP1-exposed HAEC (Control = 70 +/- 7.5, UFP1 = 106.7 +/- 12.5, UFP2 = 137.0 +/- 8.0, n = 3, P < 0.05). Adenovirus NF-kappaB Luciferase reporter assays revealed that UFP2, but not UFP1, significantly induced NF-kappaB activities. NF-kappaB inhibitor, CAY10512, significantly abrogated UFP2-induced pro-inflammatory gene expression and monocyte binding.ConclusionWhile UFP1 induced higher level of oxidative stress and stress response gene expression, only UFP2, with higher levels of redox active organic compounds and metals, induced pro-inflammatory responses via NF-kappaB signaling. Thus, UFP with distinct chemical compositions caused differential response patterns in endothelial cells.

Project description:Exposure to vehicular emissions has been linked to numerous adverse health effects. In response to the arising concerns, near-road monitoring is conducted to better characterize the impact of mobile source emissions on air quality and exposure in the near-road environment. An intensive measurement campaign measured traffic-related air pollutants (TRAPs) and related data (e.g., meteorology, traffic, regional air pollutant levels) in Atlanta, along one of the busiest highway corridors in the US. Given the complexity of the near-road environment, the study aimed to compare two near-road monitors, located in close proximity to each other, to assess how observed similarities and differences between measurements at these two sites inform the siting of other near-road monitoring stations. TRAP measurements, including carbon monoxide (CO) and nitrogen dioxide (NO2), are analyzed at two roadside monitors in Atlanta, GA located within 325m of each other. Both meteorological and traffic conditions were monitored to assess the temporal impact of these factors on traffic-related pollutant concentrations. The meteorological factors drove the diurnal variability of primary pollutant concentration more than traffic count. In spite of their proximity, while the CO and NO2 concentrations were correlated with similar diurnal variations, pollutant concentrations at the two closely sited monitors differed, likely due to the differences in the siting characteristics reducing the dispersion of the primary emissions out of the near-road environment. Overall, the near-road TRAP concentrations at all sites were not as elevated as seen in prior studies, supporting that decreased vehicle emissions have led to significant reductions in TRAP levels, even along major interstates. Further, the differences in the observed levels show that use of single near-road observations will not capture pollutant levels representative of the local near-road environment and that additional approaches (e.g., air quality models) are needed to characterize exposures.
