Project description:Amblyseius eharai is a generalist predatory mite that consumes spider mites, rust mites, thrips, and pollen, with a high adaptability to various plants. To better understand ecological and behavioral aspects of this species, we investigated its functional response to different stages of two-spotted spider mite, Tetranychus urticae. Furthermore, we compared its environmental adaptability with that of other referenced phytoseiids using a temperature-dependent model of the intrinsic rate of increase. We were able to calculate the functional response parameters of both sexes of A. eharai when preying on eggs or larvae of T. urticae and, for females only, when preying on the deutonymph of T. urticae. Among the various combinations tested herein, A. eharai females preying on T. urticae larvae had the highest attack rate and shortest handling time. For eggs of T. urticae, A. eharai showed a lower attack rate; however, its handling time for eggs was significantly shorter compared to other phytoseiids. Using T. urticae larva as a prey, the attack rate of female A. eharai was higher and the handling time of both sexes of this species was shorter than those of other phytoseiid mites. Amblyseius eharai populations can show maximum performance quickly due to this species' lower optimal temperature for population growth (28.1°C) compared to other phytoseiid mites. Thus, we provided evidence that this predatory mite has the potential to be a new, effective biological control agent of greenhouse pests such as T. urticae due to its high predation capacity and low optimal temperature.

Project description:Environmental factors like temperature have a great impact on the predation potential of biological control agents. In the present study, the functional response of the predatory mite Neoseiulus californicus (Acari: Phytoseiidae) to the pest mite Tetranychus urticae (Acari: Tetranychidae) at moderate to high temperatures under laboratory conditions was determined. The study aimed to understand the prey-predator interaction under different temperatures and prey densities. Five constant temperatures (24 °C, 27 °C, 30 °C, 33 °C, and 36 °C), and thirteen prey densities (4, 5, 8, 10, 12, 15, 16, 20, 24, 25, 30, 32, and 40) of each stage (adult, nymph, larvae, and egg stage) were employed in the experiment. Observations were made 24 h after the start of each experiment. Results revealed that the predatory mites showed type II functional response to adult females of T. urticae, whereas type I to other stages (nymphs, larvae, and eggs) of T. urticae. The predation capability of adult predatory mites on T. urticae was significant at 24-36 °C. The instantaneous attack rate (a) of N. californicus increased and the handling time (Th) decreased with an increase in temperature. The maximum attack rate was recorded at 36 °C (1.28) for the egg stage. The longest handling time was (0.78) for the larval stage of T. urticae at 30 °C. Daily consumption increased with increasing prey density. Maximum daily consumption was observed at 33 °C (30.00) at the prey density of 40. Searching efficiency decreased with the increase in prey density but was found to increase with the rise in temperature. N. californicus was found to be voracious on the larval and egg stages. Conclusively, the incorporation of N. californicus at earlier stages (larvae and eggs) of T. urticae would be beneficial under warm conditions because managing a pest at its initial stage will save the crop from major losses. The results presented in this study at various temperatures will be helpful in different areas with different temperature extremes. The results of the functional response can also be applied to mass rearing, quality testing, and integrated pest management programmes.

Project description:The two-spotted spider mite, Tetranychus urticae Koch, has a broad host plant range and presents an extreme capacity for developing pesticide resistance, becoming a major economic pest in agriculture. Anticholinesterase insecticides still account for a big part of global insecticide sales. However, there is a growing concern about the serious resistance problems of anticholinesterase insecticides and their nontarget toxicity. In this study, structure-based virtual screening was performed to discover selective AChE inhibitors from the ChemBridge database, and 39 potential species-specific AChE inhibitor were obtained targeting T. urticae AChE, but not human AChE. Among them, compound No. 8 inhibited AChE from T. urticae, but not from human, and had an inhibitory activity comparable to that of eserine. Compound No. 8 had dose-dependent toxicity to T. urticae in glass slide-dipping assay and had significant mite control effects in a pot experiment, but required a high concentration to achieve similar control effects to spirodiclofen. The toxicity evaluation suggested that compound No. 8 had no acute toxicity on pollinator honey bees and natural predator N. californicus and did not affect strawberry growth in our assay. Compound No. 8 is a potential lead compound for developing novel acaricides with reduced nontarget toxicity.

Project description:BackgroundTetranychus urticae is a highly polyphagous species with a cosmopolitan distribution that has the status of pest in more than 100 economically significant crops all over the world. Despite a number of previous efforts to isolate genetic markers, only a reduced set of microsatellite loci has been published. Taking advantage of the whole genome sequence of T. urticae that recently became available; we isolated and characterized a new set of microsatellite loci and tested the level of polymorphism across populations originating from a wide geographical area.ResultsA total of 42 microsatellite sequences widespread in the T. urticae genome were identified, the exact position in the genome recorded, and PCR amplification of microsatellite loci tested with primers defined here. Fourteen loci showed unambiguous genotype patterns and were further characterized. Three multiplex polymerase chain reaction sets were optimized in order to genotype a total of 24 polymorphic loci, including 10 previously published Tetranychus-specific loci. The microsatellite kits successfully amplified 686 individuals from 60 field populations for which we assessed the level of genetic diversity. The number of alleles per locus ranged from 3 to 16 and the expected heterozygosity values ranged from 0.12 to 0.81. Most of the loci displayed a significant excess of homozygous and did not model the Hardy-Weinberg equilibrium. This can be explained by the arrhenotokous mode of reproduction of T. urticae.ConclusionsThese primers represent a valuable resource for robust studies on the genetic structure, dispersal and population biology of T. urticae, that can be used in managing this destructive agricultural pest.

Project description:Soybean is one of the most important sources of food, protein, and oil in the world. Reductions in grain number and quality are caused by different biotic stresses. One of the most common is the phytophagous mite Tetranychus urticae Koch (Acari: Tetranychidae), which inhibits plant growth and grain production. The identification of plant responses to early and late T. urticae infestation is important for a better understanding of the mite-plant interaction. We therefore aimed to evaluate the physiological and molecular responses of soybean plants to mite infestation for 5 and 21 days. Visual and microscopic symptoms of leaf damage, H2O2 accumulation, and lipid peroxidation increased consistently throughout the infestation period, while shoot length/dry weight, chlorophyll level, and number of days to reach specific developmental stages were negatively affected by T. urticae infestation. Using proteomic analysis, we identified 185 and 266 differentially abundant proteins after early (5 days) and late (21 days) mite infestation, respectively, which suggests a complex remodeling of diverse metabolic pathways. GO, KEGG, and protein-protein interaction analyses indicated that photorespiration, chlorophyll synthesis, amino acid metabolism, Krebs cycle/energy production, mitochondrial translation, nucleotide salvage, PS II assembly, and reductive pentose-P cycle are all impacted after both early and late infestation. Specific metabolic pathways modified only after early infestation include cell wall modification, cytoskeleton composition, cell division, and lysine/histidine metabolism, while JA biosynthesis, antioxidant system, S-adenosyl methionine cycle, PS II repair, cysteine/methionine/glutathione/ascorbate/-linolenic acid/selenocompound metabolism, arginine biosynthesis, and proteasome are modified only after late infestation. These differentially abundant proteins can be used as biotechnological tools in future breeding programs aiming at increased resistance to mite infestation.

Project description:BackgroundTwo colour forms of the two-spotted spider mite (Tetranychus urticae Koch) coexist in China: a red (carmine) form, which is considered to be native and a green form which is considered to be invasive. The population genetic diversity and population genetic structure of this organism were unclear in China, and there is a controversy over whether they constitute distinct species. To address these issues, we genotyped a total of 1,055 individuals from 18 red populations and 7 green populations in China using eight microsatellite loci.ResultsWe identified 109 alleles. We found a highly significant genetic differentiation among the 25 populations (global FST = 0.506, global FST {ENA} = 0.473) and a low genetic diversity in each population. In addition, genetic diversity of the red form mites was found to be higher than the green form. Pearson correlations between statistics of variation (AR and HE) and geographic coordinates (latitude and longitude) showed that the genetic diversity of the red form was correlated with latitude. Using Bayesian clustering, we divided the Chinese mite populations into five clades which were well congruent with their geographic distributions.ConclusionsSpider mites possess low levels of genetic diversity, limit gene flow between populations and significant and IBD (isolation by distance) effect. These factors in turn contribute to the strong subdivision of genetic structure. In addition, population genetic structure results don't support the separation of the two forms of spider mite into two species. The morphological differences between the two forms of mites may be a result of epigenetic effects.

Project description:Spider mites of the genus Tetranychus are difficult to identify due to their limited diagnostic characters. Many of them are morphologically similar and males are needed for species-level identification. Tetranychus urticae is a common interception and non-regulated pest at New Zealand's borders, however, most of the intercepted specimens are females and the identification was left at Tetranychus sp. Consequently, the shipments need to be fumigated. DNA sequencing and PCR-restriction fragment length polymorphism (PCR-RFLP) protocols could be used to facilitate the accurate identification. However, in the context of border security practiced in New Zealand, insect identifications are required to be provided within four hours of receiving the samples; thus, those molecular methods are not sufficient to meet this requirement. Therefore, a real-time PCR TaqMan assay was developed for identification of T. urticae by amplification of a 142 bp Internal Transcribed Spacer (ITS) 1 sequence. The developed assay is rapid, detects all life stages of T. urticae within three hours, and does not react with closely related species. Plasmid DNA containing ITS1 sequence of T. uritcae was serially diluted and used as standards in the real-time PCR assay. The quantification cycle (Cq) value of the assay depicted a strong linear relationship with T. urticae DNA content, with a regression coefficient of 0.99 and efficiency of 98%. The detection limit was estimated to be ten copies of the T. urticae target region. The assay was validated against a range of T. urticae specimens from various countries and hosts in a blind panel test. Therefore the application of the assay at New Zealand will reduce the unnecessary fumigation and be beneficial to both the importers and exporters. It is expected that the implementation of this real-time PCR assay would have wide applications in diagnostic and research agencies worldwide.

Project description:Typhlodromus pyri Scheuten and Euseius finlandicus (Oudemans) are important predators of phytophagous mites. The present laboratory study aimed to determine whether both species can develop and reach maturity feeding on spider mites occurring on willows, i.e., Schizotetranychus schizopus (Zacher), Schizotetranychus garmani Pritchard & Baker, and Tetranychus urticae Koch, and on Brassica napus L. pollen. The predators' development, reproduction and demographic parameters were significantly affected by diet. The data suggest that rape pollen can be useful in mass rearing of E. finlandicus but is completely unsuitable as alternative food for T. pyri. Short development time and high values of population parameters achieved by T. pyri feeding on larvae and protonymphs of S. schizopus and by E. finlandicus feeding on juvenile stages of S. garmani indicate great suitability of these preys as food for the phytoseiids, and make both predatory species promising biocontrol agents in spider mite control on willows.

Project description:BackgroundThe tomato red spider mite, Tetranychus evansi Baker & Pritchard (Acari: Tetranychidae), is an agricultural pest of solanaceous crops. Although T. evansi is of South American subtropical origin, it has recently expanded its distribution range to many tropical and temperate areas around the world. Its potential distribution range in response to scenarios of global warming was recently modeled, confirming its current and possible future distributions. Here, we experimentally investigated the biological traits of T. evansi in the context of the current and future global warming (2100) scenarios. Using an environmental simulation system, we tested the life-history traits of T. evansi under current summer temperatures (as of June, July, and August 2016) and under expected temperature increases based on two IPCC scenarios: RCP2.6 (+ 1 °C) and RCP8.5 (+ 3.7 °C). The mites were introduced into each scenario on 1 June and their sequential progeny were used for testing in each following month.ResultsThe mite could develop and reproduce under all scenarios. There was a decrease in the duration of lifespan and female fecundity at RCP8.5 during June and August, but this may be compensated for by the high intrinsic rate of increase, which implies faster population growth and shorter generation time.ConclusionOur study and other reports reveal the high adaptability of T. evansi to a wide range of summer temperatures; this may explain its current distribution. We anticipate that global warming will favor the spread of T. evansi and may further expand its distribution to a large area of the globe. These findings should be of ecological and practical relevance for designing prevention and control strategies.
