Comparative Analysis of Maternal Colostrum and Colostrum Replacer Effects on Immunity, Growth, and Health of Japanese Black Calves.

ABSTRACT: Maternal colostrum (MC) is an important source of nutrients and immune factors for newborn calves. However, when colostrum is unavailable or of poor quality, a colostrum replacer (CR) may be a suitable alternative to MC. As stock-raising farmers must make informed decisions about colostrum feeding management, this study was conducted to determine the effect of feeding MC versus CR on the promotion of immunological status, growth, and health in pre-weaned Japanese black (JB) calves. Sixteen newborn JB calves were fed MC after birth, and 16 JB calves were fed CR. For the MC group, the numbers of γδ T cells, CD4⁺ cells, CD8⁺ cells, CD4⁺CD8⁺ cells, B cells, and MHC class II⁺ cells were significantly higher compared with the CR group. Furthermore, the expression levels of interleukin (IL)-1β-, IL-2-, and interferon-γ (IFN-γ)-encoding mRNAs were significantly higher in the MC group compared with the CR group. A lower incidence of disease in 1-month-old calves and higher carcass weight in the MC group were observed compared with the CR group. These results suggest that CR activates the immune system delayed in calves compared with MC. MC increases populations of various immunocompetent cells, which can reduce infection rates and improve body weight gain.

Project description:The objective of the experiment was to evaluate effects of increased milk replacer feeding on growth, intake, feed efficiency, and health parameters in stressed calves. Holstein bull calves (n = 120; approximately 3 to 8 d of age) were purchased from sale barns and dairy farms and housed in fiberglass hutches. In addition, wood shavings contaminated with coronavirus were mixed with clean shavings and added to each hutch before the start of the experiment. Calves were fed either a fixed amount (454 g/d) of a 20% crude protein (CP), 20% fat milk replacer to weaning at 28 d or a variable amount (454, 681, 908, and 454 g/d on d 0 to 7, 8 to 14, 15 to 31, and 32 to 41, respectively) of a milk replacer containing 28% CP and 17% fat without or with added dietary supplement containing bovine serum. Calves were also fed commercial calf starter and water ad libitum. Plasma IgG concentration in most calves on arrival at the facility was < 10 g/L. Intake, change in body weight, feed efficiency, morbidity and mortality, and selected plasma metabolites were determined. Body weight at 28 d, 56 d, daily body weight gain, intake of milk replacer, fecal scores, days with diarrhea, and days treated with antibiotics were increased with feeding variable amount of milk replacer over the 56-d study. Starter intake from d 1 to 56 was reduced from 919 to 717 g/d in calves fed fixed and variable amounts of milk replacer, respectively. Morbidity, measured as the number of days that calves had diarrhea, was increased by 53% when a variable amount of milk replacer was fed. Calves fed variable milk replacer were treated with antibiotics for 3.1 d compared with 1.9 d for calves fed 454 g of milk replacer/d. Concentrations of plasma glucose, urea N, and insulin-like growth factor-I were increased when calves were fed variable amount of milk replacer. Dietary supplement containing bovine serum had no effect on any parameter measured. There was no effect of milk replacer feeding on concentrations of nonesterified fatty acids, total protein, or growth hormone concentrations. Plasma tumor necrosis factor-alpha was highest in calves with the highest plasma IgG concentrations on the day of arrival and might be related to the calf's ability to identify pathogens in the environment. Under conditions of this study, calves fed variable amount of milk replacer and exposed to immunological challenge before weaning had greater BW gain, but also increased incidence of diarrhea that required added veterinary treatments.

Project description:The production of crossbred beef × dairy (B × D) calves is increasing; however, evaluation of pre-weaning feeding strategies for B × D calves is limited. We hypothesized that both male and female B × D calves fed added fat and protein through milk replacer would have increased muscle growth, muscle fiber cross-sectional area, average daily gain, BW, and morphometric measurements. We also hypothesized that calves fed the additional fat and protein milk replacer would have upregulation of regulatory genes involved in muscle hypertrophy. SimAngus × Holstein calves (n = 42) were assigned to one of two milk replacers: 30.0% crude protein (CP), 32% crude fat (CF) milk replacer (HPHF, n = 11 males, 11 females), or 22% CP, 20% CF milk replacer (CON, n = 10 males, 10 females) from 0 to 8 wk of age. B × D calves were weighed at birth and weekly thereafter. At week 2 and 8, longissimus muscle biopsies were collected for muscle fiber cross-sectional area (CSA) or gene expression analysis. Ultrasounds were performed at 4 and 8 wk of age to quantify ribeye area (REA), and backfat and rump fat thickness. Morphometric measurements, BW, CSA, and ultrasound data were analyzed with PROC MIXED with animal as the subject and fixed effects of milk replacer, age, sex, and their interactions. Gene expression data were analyzed in R Studio. Calves that consumed the HPHF milk replacer were heavier than calves consuming the CON milk replacer (HPHF: 70.7 ± 0.39 kg; CON: 68.5 ± 0.41 kg; P < 0.01). At 8 wk of age, HPHF calves tended to have 14% larger muscle fiber CSA than CON calves (P = 0.06). No differences due to diet were observed for REA or fat thickness (P ≥ 0.38). Expression of MyoD tended to be 34% greater in CON females than HPHF females at 2 wk (P = 0.06), but at 8 wk, HPHF females tended to express 39% more MyoD than CON females (P = 0.09). Myogenin expression was 3% greater in CON calves than HPHF calves at 2 wk (P = 0.02), and CON females tended to express 52% more IGF-1 than HPHF females (P = 0.07). Feeding a milk replacer with a protein and fat content similar to beef cow milk improves B × D calf growth compared with a conventional milk replacer with less protein and fat. Improvements in early growth may improve B × D carcass quality and quantity, with the potential to increase return to the producer.

Project description:Acquisition of adequate transfer of passive immunity (ATPI) by calves depends on the absorption of sufficient mass of colostral immunoglobulin G (IgG). Several studies report conflicting evidence regarding the ability of feeding pooled colostrum to achieve ATPI. Pooling colostrum is practical and efficient for some dairies, and recommendations are required to prevent failure of transfer of passive immunity (FTPI) in calves following pooling. This study aimed to determine the effect of pooling colostrum on serum IgG concentrations, FTPI, and preweaning mortality in calves. A prospective study was performed on two conventional Jersey dairy farms where heat treatment of colostrum occurred in the same colostrum processing kitchen. Four to 10 cows contributed to colostrum pools. A sample of the colostrum pool fed to the calves and serum from calves at 24-72 h was collected for IgG concentration determination by single radial immunodiffusion assay. Multivariable and logistic regression analyses were performed to evaluate factors that predicated serum IgG concentrations and the probability of FTPI, respectively. A Cox proportional hazard model analysis was performed to determine risk factors for mortality over the preweaning period. A total of 164 calves fed 28 colostrum pools were enrolled. Birth weight, number of colostrum feedings and pool IgG concentrations were significant predictors of calf serum IgG concentrations at 24-72 h, whereas the number of colostrum feedings and age at bleeding to determine passive transfer status were not significant predictors of calf serum IgG concentrations at 24-72 h. The prevalence of FTPI was 4.9%. Birth weight, pool IgG concentrations, number of colostrum feedings, and age at bleeding to check for passive transfer status were not significant predictors of the probability of FTPI at 24-72 h. The incidence of mortality was 4.3%. Passive transfer status was not a predictor of mortality. Our study demonstrates the effect of pooling colostrum on serum IgG concentrations even in herds where colostrum with higher median colostrum IgG concentrations is fed to calves. The results emphasize the recommendations to assess pooled colostrum IgG concentrations before feeding calves.
