Two new species of the mealybug genus Paracoccus from Jiangxi, South China (Hemiptera, Coccomorpha, Pseudococcidae).

ABSTRACT: Two new mealybug species, Paracoccusgillianwatsonae Zhang, sp. nov. and P.wui Zhang, sp. nov., collected from Jiangxi, South China, are described and illustrated based on the morphology of adult females. Paracoccusgillianwatsonae is similar to P.burnerae (Brain, 1915), but it differs in having fewer pairs of cerarii, and in lacking both ventral oral collar tubular ducts on the margins of the head and translucent pores on the hind femur. Paracoccuswui resembles P.keralae Williams, 2004 and P.neocarens (Lit, 1992), but it differs in lacking ventral oral collar tubular ducts on the margins of the head and in having multilocular disc-pores usually in double rows at the posterior edges of abdominal segments V and VI. A key to the Paracoccus species found in China is provided.

Project description:A study of the mealybug genus Planococcus Ferris, 1950 (Hemiptera, Coccomorpha, Pseudococcidae) known from China is presented and 12 species are recognised. Of these, Planococcuscamelliae Zhang, sp. nov. is described as new to science based on the morphology of the adult female, and P.bambusifolii (Takahashi, 1951) is recorded from China for the first time. Molecular analyses based on the mitochondrial gene cytochrome c oxidase subunit I (COI) of the new species and a key to species of the genus Planococcus in China are also given.

Three new species of mealybug (Hemiptera, Coccomorpha, Pseudococcidae) on persimmon fruit trees (Diospyros kaki) in southern Brazil.

Project description:Brazil has the greatest insect diversity in the world; however, little is known about its scale insect species (Hemiptera: Coccomorpha). Mealybugs (Pseudococcidae) have been found in at least 50% of persimmon orchards Diospyros kaki L. in the southern part of the country. In this study three new mealybug species on persimmon trees located in the Serra Gaúcha Region, RS, Brazil, namely, Anisococcus granarae Pacheco da Silva & Kaydan, sp. n., Ferrisia kaki Kaydan & Pacheco da Silva, sp. n. and Pseudococcus rosangelae Pacheco da Silva & Kaydan, sp. n. are described. In addition, an identification key for the genera occurring on fruit orchards and vineyards in Brazil is provided, together with illustrations and molecular data for the new species.

Project description:A new mealybug species Atrococcus rushuiensis Zhang, sp. nov., collected under the leaf sheath of Sporobolus fertilis (Poaceae) in Fuzhou City, Jiangxi Province, China, is described and illustrated. A new combination is introduced, transferring Allotrionymus shanxiensis Wu to the genus Atrococcus as A. shanxiensis (Wu), comb. nov. A key is presented for the species of Atrococcus recorded from China.

Molecular and morphological identification of mealybug species (Hemiptera: Pseudococcidae) in Brazilian vineyards.

Project description:Mealybugs (Hemiptera: Pseudococcidae) are pests constraining the international trade of Brazilian table grapes. They damage grapes by transmitting viruses and toxins, causing defoliation, chlorosis, and vigor losses and favoring the development of sooty mold. Difficulties in mealybug identification remain an obstacle to the adequate management of these pests. In this study, our primary aim was to identify the principal mealybug species infesting the major table grape-producing regions in Brazil, by morphological and molecular characterization. Our secondary aim was to develop a rapid identification kit based on species-specific Polymerase Chain Reactions, to facilitate the routine identification of the most common pest species. We surveyed 40 sites infested with mealybugs and identified 17 species: Dysmicoccus brevipes (Cockerell), Dysmicoccus sylvarum Williams and Granara de Willink, Dysmicoccus texensis (Tinsley), Ferrisia cristinae Kaydan and Gullan, Ferrisia meridionalis Williams, Ferrisia terani Williams and Granara de Willink, Phenacoccus baccharidis Williams, Phenacoccus parvus Morrison, Phenacoccus solenopsis Tinsley, Planococcus citri (Risso), Pseudococcus viburni (Signoret), Pseudococcus cryptus Hempel, four taxa closely related each of to Pseudococcus viburni, Pseudococcus sociabilis Hambleton, Pseudococcus maritimus (Ehrhorn) and Pseudococcus meridionalis Prado, and one specimen from the genus Pseudococcus Westwood. The PCR method developed effectively identified five mealybug species of economic interest on grape in Brazil: D. brevipes, Pl. citri, Ps. viburni, Ph. solenopsis and Planococcus ficus (Signoret). Nevertheless, it is not possible to assure that this procedure is reliable for taxa that have not been sampled already and might be very closely related to the target species.

First records of the genus Pelionella Kaydan, 2015 in East Asia, with description of a new species (Hemiptera, Coccomorpha, Pseudococcidae).

Project description:Two mealybug species (Hemiptera: Coccomorpha: Pseudococcidae), Pelionella osakaensissp. n. and P. manifecta (Borchsenius, 1949), are described and illustrated based on adult female specimens collected in Japan, on the Japanese mugwort Artemisia indica Willd. var. maximowiczii (Nakai) H. Hara (Asteraceae). These are the first records of the occurrence of Pelionella species in East Asia. The new species is similar to P. grassiana (Goux, 1989) and P. proeminens (Goux, 1990), but differs in lacking multilocular pores with double loculi rings on the venter and in possessing dorsal cerarii and a circulus. The Japanese population of P. manifecta is morphologically slightly different from the Azerbaijani and French populations in lacking large-type oral-collar tubular ducts associated with clusters formed by multilocular pores and oral-collar ducts on ventral abdominal segments III and IV. A modified key to species of the genus Pelionella Kaydan, 2015, is provided.

Mealybug (Hemiptera: Pseudococcidae) Species Associated with Cacao Mild Mosaic Virus and Evidence of Virus Acquisition.

Project description:Theobroma cacao is affected by viruses on every continent where the crop is cultivated, with the most well-known ones belonging to the Badnavirus genus. One of these, cacao mild mosaic virus (CaMMV), is present in the Americas, and is transmitted by several species of Pseudococcidae (mealybugs). To determine which species are associated with virus-affected cacao plants in North America, and to assess their potential as vectors, mealybugs (n = 166) were collected from infected trees in Florida, and identified using COI, ITS2, and 28S markers. The species present were Pseudococcus jackbeardsleyi (38%; n = 63), Maconellicoccus hirsutus (34.3%; n = 57), Pseudococcus comstocki (15.7%; n = 26), and Ferrisia virgata (12%; n = 20). Virus acquisition was assessed by testing mealybug DNA (0.8 ng) using a nested PCR that amplified a 500 bp fragment of the movement protein-coat protein region of CaMMV. Virus sequences were obtained from 34.6 to 43.1% of the insects tested; however, acquisition did not differ among species, X2 (3, N = 166) = 0.56, p < 0.91. This study identified two new mealybug species, P. jackbeardsleyi and M. hirsutus, as potential vectors of CaMMV. This information is essential for understanding the infection cycle of CaMMV and developing effective management strategies.

Project description:A new species of the genus Formicoccus Takahashi (Hemiptera, Coccomorpha, Pseudococcidae) collected from the holoparasitic plant Balanophorafungosa J. R. & G. Forst (Balanophoraceae), on Ishigaki Island, Japan, is described as Formicoccusyoshinoi Tanaka, sp. nov. based on the morphology of adult females. This species is similar to F.formicarius (1900) and F.erythrinae Williams, 2004, but differs from them by having fewer than six cerarii, and only one type of ventral oral collar tubular duct distributed on the medial area of the posterior abdominal segments. Keys to the Oriental species of the genus Formicoccus are provided.

Induction of leaf curling in cassava plants by the cassava mealybug Phenacoccus manihoti (Hemiptera: Pseudococcidae)

Project description:Certain phytophagous insects can induce leaf curling in their host plants that may provide protected and nutrient-rich habitats. However, the mechanisms of this induction remain poorly understood. The cassava mealybug, Phenacoccus manihoti Matile-Ferrero (Hemiptera: Pseudococcidae), is a serious pest of the cassava and causes leaf curling. To reveal the mechanisms of leaf-curl induction, we first inoculated varying numbers of mealybugs in different locations, namely, the apical meristem and the stem, on cassava seedlings. Second, we performed transcriptome analysis using the total RNA extracted from leaves. The results showed that a single insect was able to induce leaf curling, but the intensity and frequency of the leaf curling were positively correlated with the number of insects. Furthermore, the leaf curling occurred when the mealybugs fed on or close to the apical meristem but not when they fed on the stem. Transcriptome analysis identified a total of 3,931 differentially expressed genes (DEGs) from intact plants and the plants inoculated with mealybugs at different time points. GO analysis of the biological processes revealed that the DEGs contained a series of factors for leaf development of the adaxial–abaxial axis, and auxin biosynthesis and polarity. This suggests that alterations in these functions may cause leaf curling.
