Project description:EFSA carries out the risk assessment of genetically modified plants for food and feed uses under Regulation (EU) No 503/2013. Exposure assessment - anticipated intake/extend of use shall be an essential element of the risk assessment of genetically modified feeds, as required by Regulation (EU) No 503/2013. Estimates of animal dietary exposure to newly expressed proteins should be determined to cover average consumption across all the different species, age, physiological and productive phases of farmed and companion animals, and identify and consider particular consumer groups with expected higher exposure. This statement is aimed at facilitating the reporting of the information that applicants need to provide on expected animal dietary exposure to newly expressed proteins and to increase harmonisation of the application dossiers to be assessed by the EFSA GMO Panel. Advice is provided on the selection of proper feed consumption and feed concentration data, and on the reporting of exposure's estimates. An overview of the different uncertainties that may be linked to the estimations is provided. This statement also explains how to access an Excel calculator which should be used in future applications as basis to provide a more consistent presentation of estimates of expected animal dietary exposure.

Project description:Nickel (Ni) is a silvery-white, hard, ductile metal existing in oxidation states; in biological systems, Ni2+ is the prevalent form. All analytical results used to estimate animal dietary exposure were reported as Ni', without providing information on specific chemical species. Considering the data provided by Member states, among FoodEx level 1 feed categories, the highest mean Ni levels were measured in 'Minerals and products derived thereof' (n = 72). High mean Ni concentrations were also observed in 'Compound feed' (n = 516), in particular in complementary feeds for fattening cattles, unspecified complementary feed and complementary feeds for fattening pigs. Within grains used as feed (n = 597), the highest mean Ni concentrations were measured in oats. In addition, Ni concentrations in hydrogenated vegetable oils/fats were reported by industry. Exposure to Ni in livestock and companion animals varied according to the animal species. When considering the diets with hydrogenated vegetable oils/fats based on the reported Ni concentrations, the mean exposures varied between 6.0 μg Ni/kg body weight (bw) per day in cats and 79 μg Ni/kg bw per day in laying hens and the high exposure levels varied between 11 μg Ni/kg bw per day in cats and 127 μg Ni/kg bw per day in rabbits. The mean exposure estimates considering the maximum concentration of Ni assumed from good manufacturing practice in hydrogenated vegetable oils/fats (50 mg Ni/kg) varied between 27 μg Ni/kg bw per day in cats and 255 μg Ni/kg bw per day in rabbits; for the high concentration scenarios, exposures varied between 30 μg Ni/kg bw per day and 307 μg Ni/kg bw per day in the same species. The estimated exposures to Ni are in line with the one reported in the 2015 EFSA opinion, using a worst-case scenario. When estimating exposure with a realistic scenario, using the reported Ni concentration in hydrogenated vegetable oils/fats, the exposure of livestock and companion animals is lower (approximately from 1.5 to 6 times, depending on the species) than the 2015 assessment.

Project description:Dietary risk assessment (DRA) of pesticides includes the estimation of chronic and acute exposures from crop residues, but assesses acute exposures only for pesticides with an acute reference dose (ARfD). Acute estimation uses high percentiles of food consumption surveys which are considerably higher than per capita lifetime averaged food consumption values which are used for chronic estimations. Assessing acute risks only for pesticides with an ARfD tacitly assumes that chronic risk assessment covers also intermittent occurring exposures which could significantly exceed chronic estimates. The present investigation conducted on 2200 rat studies from 436 pesticides provides evidence demonstrating that pesticides with and without ARfD have no-observed-adverse-effect levels (NOAELs) which remain statistically unchanged in developmental, subacute, subchronic, reproductive and chronic toxicity studies covering exposure durations between 2 and 104 weeks. DRA of pesticides without ARfD needs reconsideration in light of equally high toxic dose levels after short- and long-term exposures, suggesting that intermittent exposures could be toxic, if they repeatedly exceed the acceptable chronic daily intake (ADI; conceptually the human counterpart of chronic animal NOAEL). As such risks are currently not assessed for pesticides without ARfD, the current DRA concept, which automatically presumes the use of low chronic exposure estimates entirely covers the risks of not acutely toxic pesticides, needs reconsideration. Furthermore, risks to intermittent occurring high exposures are probably also insufficiently assessed for pesticides where the ARfD is significantly higher than the ADI. As an example, the maximum residue limit for bifenazate in peaches is discussed.

Project description:An estimated 11 million people in the US have home wells with unsafe levels of hazardous metals and nitrate. The national scope of the health risk from consuming this water has not been assessed as home wells are largely unregulated and data on well water treatment and consumption are lacking. Here, we assessed health risks from consumption of contaminated well water on the Crow Reservation by conducting a community-engaged, cumulative risk assessment. Well water testing, surveys and interviews were used to collect data on contaminant concentrations, water treatment methods, well water consumption, and well and septic system protection and maintenance practices. Additive Hazard Index calculations show that the water in more than 39% of wells is unsafe due to uranium, manganese, nitrate, zinc and/or arsenic. Most families' financial resources are limited, and 95% of participants do not employ water treatment technologies. Despite widespread high total dissolved solids, poor taste and odor, 80% of families consume their well water. Lack of environmental health literacy about well water safety, pre-existing health conditions and limited environmental enforcement also contribute to vulnerability. Ensuring access to safe drinking water and providing accompanying education are urgent public health priorities for Crow and other rural US families with low environmental health literacy and limited financial resources.

Project description:In this study, the 10-year dietary habits of patients diagnosed with cancer (n = 1155) were retrospectively analyzed, and the relationships between dietary (red meat, white meat, fish meat, French fries, bread, instant coffee, ready-to-drink coffee, Turkish coffee, and black tea) heterocyclic amines, polycyclic aromatic hydrocarbons, acrylamide, and N-nitrosamine-based risk scores and cancer types were statistically evaluated. The foods with the highest and lowest mean dietary heat-treatment contaminant risk scores were red meat and ready-to-drink coffee, respectively. There were statistically significant differences in the dietary heat-treatment contamination risk scores based on the cancer patients' demographic characteristics (sex, age, smoking, and body mass index) (p < 0.05). According to the cancer types, the systems with the highest and lowest dietary heat-treatment contaminant risk scores were determined as other (brain, thyroid, lymphatic malignancies, skin, oro- and hypopharynx, and hematology) and the reproductive (breast, uterus, and ovary) system, respectively. The relationship between instant coffee consumption and respiratory system cancer types, the frequency of consumption of French fries and urinary system cancer types and the consumption of meat products and gastrointestinal system cancer types were determined. It is thought that this study contains important findings regarding the relationship between dietary habits and cancer and will be a good source for other studies to be conducted in this context.

Project description:The ergot alkaloids (EAs) are mycotoxins produced by several species of fungi in the genus Claviceps. In Europe, Claviceps purpurea is the most widespread species and it commonly affects cereals such as rye, wheat, triticale, barley, millets and oats. Food and feed samples used to estimate human and animal dietary exposure were analysed for the 12 main C. purpurea EAs: ergometrine, ergosine, ergocornine, ergotamine, ergocristine, ergocryptine (α- and β-isomers) and their corresponding -inine (S)-epimers. The highest levels of EAs were reported in rye and rye-containing commodities. In humans, mean chronic dietary exposure was highest in 'Toddlers' and 'Other children' with maximum UB estimates of 0.47 and 0.46 μg/kg bw per day, respectively. The 95th percentile exposure was highest in 'Toddlers' with a maximum UB estimate of 0.86 μg/kg bw per day. UB estimations were on average fourfold higher than LB estimations. Average acute exposure (MB estimations) ranged from 0.02 μg/kg bw per day in 'Infants' up to 0.32 μg/kg bw per day estimated in 'Other children'. For the 95th percentile acute exposure, the highest estimate was for a dietary survey within the age class 'Other children' (0.98 μg/kg bw per day). Dietary exposure estimates for animals, assuming a mean concentration scenario, varied between 0.31-0.46 μg/kg bw per day in beef cattle and 6.82-8.07 μg/kg bw per day (LB-UB) in piglets, while exposure estimates assuming a high concentration scenario (95th percentile) varied between 1.43-1.45 μg/kg bw per day and 16.38-16.61 μg/kg bw per day (LB-UB) in the same species. A statistically significant linear relationship between the content of sclerotia and the levels of EAs quantified was observed in different crops (barley, oats, rye, triticale and wheat grains). However, the absence of sclerotia cannot exclude the presence of EAs as samples with no sclerotia identified showed measurable levels of EAs ('false negatives').

Project description:There is increasing concern that particulate contaminants including manufactured nanomaterials and nano- and microplastics taken up and or accumulating in lower-trophic level aquatic organisms results in dietary exposure of fish feeding on these organisms. Controlled feeding studies can help advance our understanding of dietary uptake, bioaccumulation, and associated effects of (nano)particulate contaminants in fish, and also provide information about their likelihood to be transferred along the trophic chain and or to act as vector for other, surface-adsorbed pollutants. However, traditional approaches to prepare test feed for dietary exposure studies where commercial fish feed such as flakes, granules or pellets are soaked or spray-spiked with dissolved chemicals are not well suitable for (nano-)particulate contaminants. Microplastics, which often have sizes in the µm to mm range, and manufactured nanomaterials, in particular those which are soluble, such as metal/metal oxide nanoparticles, have to be incorporated into the feed to avoid their dissociation and or dissolution before the feed is ingested by the animal to avoid undesired waterborne exposure, which may confound results.•Here we describe a methodological approach to produce worm-shaped food packages, that is a practical diet, of controlled diameter and length (in the millimeter range), which allows to prepare food rations with a weight in the order of a few milligrams and to adjust the food rations to the individual body wet weight of small experimental fish with high accuracy (±0.5 mg) without the need for weighing/proportioning the feed using a scale.•The method can be used to prepare test feed with internally incorporated particulate contaminants, such as manufactured nanomaterials and nano- and microplastics, to assess the latter's dietary uptake, bioaccumulation and associated toxicity in fish. We described two protocol variations: One using dry starting material, such as feed flakes, and one using liquid starting material, such as worm homogenate.•The method has been developed for academic research environments with no access to specialized equipment for test feed preparation, and uses utensils and inexpensive plastic ware belonging to the standard inventory of ecotoxicological research laboratories.

Project description:The dissipation behavior and dietary exposure risk assessment of four fungicides (dimethomorph, mandipropamid, myclobutanil, and metalaxyl) was performed in fruits and leaves of squash grown under greenhouse conditions. Squash fruit and leaf samples were randomly collected at 0, 3, 5, 7, and 14 days after the last pesticide application. Analysis was performed using ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS). The quick, easy, cheap, effective, rugged, and safe (QuEChERS) method was used for sample preparation. Recovery rates at two spiked levels (0.01 and 0.1 mg/kg) were found to be in the range of 76.4%-101.9% for the analyzed pesticides and their relative standard deviations were ≤4%. Pesticide half-lives were 2.1 and 4.9 days for dimethomorph, 4.6 and 8.1 days for mandipropamid, 4.7 and 8.2 days for myclobutanil, and 2.7 and 5 days for metalaxyl in squash fruit and leaf, respectively. Regarding the total surveyors, hazard quotient values for squash fruit and leaf were ≤1.03 × 10-3 and ≤2.39 × 10-3, respectively. These values in the case of true consumers were ≤3.14 × 10-3 and ≤3.91 × 10-1, respectively.

Project description:The European Commission asked EFSA for a scientific opinion on the risks to animal health related to nitrite and nitrate in feed. For nitrate ion, the EFSA Panel on Contaminants in the Food Chain (CONTAM Panel) identified a BMDL 10 of 64 mg nitrate/kg body weight (bw) per day for adult cattle, based on methaemoglobin (MetHb) levels in animal's blood that would not induce clinical signs of hypoxia. The BMDL 10 is applicable to all bovines, except for pregnant cows in which reproductive effects were not clearly associated with MetHb formation. Since the data available suggested that ovines and caprines are not more sensitive than bovines, the BMDL 10 could also be applied to these species. Highest mean exposure estimates of 53 and 60 mg nitrate/kg bw per day in grass silage-based diets for beef cattle and fattening goats, respectively, may raise a health concern for ruminants when compared with the BMDL 10 of 64 mg nitrate/kg bw per day. The concern may be higher because other forages might contain higher levels of nitrate. Highest mean exposure estimates of 2.0 mg nitrate/kg bw per day in pigs' feeds indicate a low risk for adverse health effects, when compared with an identified no observed adverse effect level (NOAEL) of 410 mg nitrate/kg bw per day, although the levels of exposure might be underestimated due to the absence of data on certain key ingredients in the diets of this species. Due to the limitations of the data available, the CONTAM Panel could not characterise the health risk in species other than ruminants and pigs from nitrate and in all livestock and companion animals from nitrite. Based on a limited data set, both the transfer of nitrate and nitrite from feed to food products of animal origin and the nitrate- and nitrite-mediated formation of N-nitrosamines and their transfer into these products are likely to be negligible.
