There is increasing evidence from non-human animals that males adjust their ejaculate expenditure according to the risk of sperm competition. In this study we show that, after controlling for lifestyle factors known to influence semen quality, human males viewing images depicting sperm competition had a higher percentage of motile sperm in their ejaculates. Project description:PurposeCell-free mRNAs (cfmRNAs) were quantitatively measured in human seminal plasma and its relationship with semen quality was investigated.MethodsHerein, a prospectively, controlled investigation was performed to study seminal plasma HSPA2 and uPA cfmRNA alterations between 21 asthenozoospermic patients and 16 normozoospermic individuals. Standard semen analysis was performed and seminal plasma cfmRNAs content was measured by real-time quantitative PCR. In addition, the regression analysis between seminal plasma cfmRNAs expression and semen parameters was performed.ResultsSeminal plasma HSPA2, but not uPA cfmRNA indicated significant difference between normozoospermia and asthenozoospermia men (P?=?0.02444 and 0.07811, respectively). Negative correlation between HSPA2 cfmRNA and sperm motility (R (2)?=?0.213, P?=?0.004) as well as sperm concentration (R (2)?=?0.133, P?=?0.026) were revealed. However, no correlation was found between seminal plasma uPA cfmRNA content and semen parameters.ConclusionsOur data suggest that seminal plasma HSPA2 cfmRNA is different between asthenozoospermic and normozoospermic individuals and it might be an indicator for semen quality.

Project description:The two databases here described were generated to evaluate the role of affective content while assessing image quality (Corchs et al., 2018) [1]. The databases are composed of images JPEG-compressed together with the subjective quality scores collected during psychophysical experiments. To reduce interferences in quality perception due to image semantic, we have restricted the semantic content, choosing only close-ups of face images, and we have considered only two emotion categories (happy and sad). We have selected 23 images with happy faces and 23 images with sad faces of high quality. For what concerns image quality we have considered JPEG-distortion with 4 levels of compression, corresponding to q-factors 10, 15, 20, 30. The first image database, hereafter called MMSP-FaceA, is thus composed of 230 images (23+23) × 5 quality levels (including the original high quality pristine images). To better consider only interferences in quality perception due to affective content, we have generated a second image database where the background of images belonging to MMSP-FaceA has been cut off. This second image database is labelled as MMSP-FaceB. Psychophysical experiments were conducted, on a controlled web-based interface, where participants rated the image quality of the two databases in a five point scale. The two final databases MMSP-FaceA and MMSP-FaceB are thus composed of 230 images each, together with the raw quality scores assigned by the observers, and are available at our laboratory web site: www.mmsp.unimib.it/download.

Project description:BackgroundAlcohol consumption and problems are associated with interpersonal difficulties. We used a twin design to assess in men the degree to which genetic or environmental influences contributed to the covariance between alcohol consumption and problems, romantic quality and social support.MethodThe sample included adult male-male twin pairs (697 monozygotic and 487 dizygotic) for whom there were interview-based data on: alcohol consumption (average monthly alcohol consumption in the past year); alcohol problems (lifetime alcohol dependence symptoms); romantic conflict and warmth; friend problems and support; and relative problems and support.ResultsKey findings were that genetic and unique environmental factors contributed to the covariance between alcohol consumption and romantic conflict; genetic factors contributed to the covariance between alcohol problems and romantic conflict; and common and unique environmental factors contributed to the covariance between alcohol problems and friend problems.ConclusionsRecognizing and addressing the overlapping genetic and environmental influences that alcohol consumption and problems share with romantic quality and other indicators of social support may have implications for substance use prevention and intervention efforts.

Project description:The objective of this study was to compare the testicular development, semen quality, and spermatogenic cell apoptosis of roosters reared in colony, single, and large cages. Rohman parental layers (n = 540) were randomly allocated into cages of rearing system groups (135 males and 405 females). The experimental period was 70 to 210 d of age. We compared testicular development and plasma main reproductive hormones (Follicle-stimulating hormone; Luteinizing hormone; Testosterone; Estrogen2;) from d 70 to 210 of roosters among the three systems. In addition, routine semen quality indexes, apoptosis of testicular spermatogenic cells and sperm apoptosis of breeding roosters under three rearing systems on d 175 and d 210 were evaluated. Roosters during the growing period (from d 70 to 140) have rapid testis growth and increasing main reproductive hormones in plasma. At the peak of sexual maturity (d 210), in colony cage, the females have a positive effect and promote the testis development of males. However, the stocking density in colony cage has no effect on testicular development; compared with the single and large cage. Roosters reared in the natural mating system had better semen quality, particularly in semen volume, density, and viability; the hatching % of fertilized eggs and healthy chicks were higher for the colony than single and large cages. Furthermore, the sperm density was higher for colony than single and large cages, which was related to the apoptosis of spermatogonia and spermatocyte, not the apoptosis of mature sperm. This study provided the basic data for the reproductive performance research of chicken reared in the colony cages.

Project description:ObjectiveTo examine the impact of men's diet on outcomes of infertility treatment with assisted reproductive technology (ART) using an empirical score representing the relation of diet with semen quality.DesignProspective cohort study.SettingFertility center at an academic medical center.PatientsWe included 296 men (688 semen samples) to identify an empirical dietary pattern and 231 couples (406 ART cycles) to investigate the association of this diet pattern with ART outcomes.InterventionsMen's diet was assessed at baseline using a validated questionnaire. An empirical dietary pattern reflecting the overall relation of diet with semen quality was identified using reduced rank regression.Main outcome measuresThe primary outcome was live birth per treatment cycle. The secondary outcomes were fertilization, implantation, and clinical pregnancy.ResultsMen had a median baseline age and body mass index of 36.8 years and 26.9 kg/m2, respectively. Although the empirical diet pattern was significantly associated with all semen parameters, the empirical diet score was not related to any clinical outcome of infertility treatment after ART. The adjusted probabilities of relevant clinical outcomes in the lowest and highest quartiles of the empirical score were 0.62 (0.50-0.73) and 0.55 (0.45-0.66) for implantation, 0.57 (0.46-0.69) and 0.50 (0.40-0.61) for clinical pregnancy, and 0.49 (0.37-0.62) and 0.36 (0.25-0.48) for live birth. Analyses excluding couples with a diagnosis of male factor infertility and, separately, excluding intracytoplasmic sperm injection cycles yielded similar results.ConclusionsA dietary score representing the overall association of diet with semen quality parameters was not associated with ART outcomes.

Project description:We need data sets of images and subjective scores to develop robust no reference (or blind) visual quality metrics for consumer applications. These applications have many uncontrolled variables because the camera creates the original media and the impairment simultaneously. We do not fully understand how this impacts the integrity of our subjective data. We put forward two new data sets of images from consumer cameras. The first data set, CCRIQ2, uses a strict experiment design, more suitable for camera performance evaluation. The second data set, VIME1, uses a loose experiment design that resembles the behavior of consumer photographers. We gather subjective scores through a subjective experiment with 24 participants using the Absolute Category Rating method. We make these two new data sets available royalty-free on the Consumer Digital Video Library. We also present their integrity analysis (proposing one new approach) and explore the possibility of combining CCRIQ2 with its legacy counterpart. We conclude that the loose experiment design yields unreliable data, despite adhering to international recommendations. This suggests that the classical subjective study design may not be suitable for studies using consumer content. Finally, we show that Hoßfeld-Schatz-Egger α failed to detect important differences between the two data sets.

Project description:BackgroundAmbient air pollution is associated with systemic increases in oxidative stress, to which sperm are particularly sensitive. Although decrements in semen quality represent a key mechanism for impaired fecundability, prior research has not established a clear association between air pollution and semen quality. To address this, we evaluated the association between ambient air pollution and semen quality among men with moderate air pollution exposure.MethodsOf 501 couples in the LIFE study, 467 male partners provided one or more semen samples. Average residential exposure to criteria air pollutants and fine particle constituents in the 72 days before ejaculation was estimated using modified Community Multiscale Air Quality models. Generalized estimating equation models estimated the association between air pollutants and semen quality parameters (volume, count, percent hypo-osmotic swollen, motility, sperm head, morphology and sperm chromatin parameters). Models adjusted for age, body mass index, smoking and season.ResultsMost associations between air pollutants and semen parameters were small. However, associations were observed for an interquartile increase in fine particulates ≤2.5 µm and decreased sperm head size, including -0.22 (95% CI -0.34, -0.11) µm2 for area, -0.06 (95% CI -0.09, -0.03) µm for length and -0.09 (95% CI -0.19, -0.06) µm for perimeter. Fine particulates were also associated with 1.03 (95% CI 0.40, 1.66) greater percent sperm head with acrosome.ConclusionsAir pollution exposure was not associated with semen quality, except for sperm head parameters. Moderate levels of ambient air pollution may not be a major contributor to semen quality.

Project description:BackgroundHigh content screening (HCS) is a powerful method for the exploration of cellular signalling and morphology that is rapidly being adopted in cancer research. HCS uses automated microscopy to collect images of cultured cells. The images are subjected to segmentation algorithms to identify cellular structures and quantitate their morphology, for hundreds to millions of individual cells. However, image analysis may be imperfect, especially for "HCS-unfriendly" cell lines whose morphology is not well handled by current image segmentation algorithms. We asked if segmentation errors were common for a clinically relevant cell line, if such errors had measurable effects on the data, and if HCS data could be improved by automated identification of well-segmented cells.ResultsCases of poor cell body segmentation occurred frequently for the SK-BR-3 cell line. We trained classifiers to identify SK-BR-3 cells that were well segmented. On an independent test set created by human review of cell images, our optimal support-vector machine classifier identified well-segmented cells with 81% accuracy. The dose responses of morphological features were measurably different in well- and poorly-segmented populations. Elimination of the poorly-segmented cell population increased the purity of DNA content distributions, while appropriately retaining biological heterogeneity, and simultaneously increasing our ability to resolve specific morphological changes in perturbed cells.ConclusionImage segmentation has a measurable impact on HCS data. The application of a multivariate shape-based filter to identify well-segmented cells improved HCS data quality for an HCS-unfriendly cell line, and could be a valuable post-processing step for some HCS datasets.

Project description:It is generally thought that a single ejaculate is a bad predictor of semen quality of a subject, because of significant intra-individual variation. Therefore, we investigated the degree to which the results of a first semen analysis differ from that of a second analysis among men from a general population in Norway. In addition, we analysed how the two different semen results mirrored the overall semen quality assessment. A total of 199 volunteers participated in the study and delivered two semen samples with an interval of 6 months. The semen parameters were determined according to the World Health Organization (WHO) 1999 guidelines, which were also used to determine whether semen quality was normal or abnormal. In addition, the DNA fragmentation index (DFI) was determined using the Sperm Chromatin Structure Assay. The two samples from each individual were very similar with regard to standard semen parameters and DFI (r(s:) 0.67-0.72), and there were no significant systematic differences between the two samples. The result of the first sample (normal/abnormal) was highly predictive of the overall conclusion based on the two samples (sperm concentration: in 93% of the cases (95% confidence interval [CI]: 89%-96%); sperm motility: in 85% of the cases (95% CI: 79%-89%); overall semen quality: in 85% of the cases (95% CI: 80%-90%). In epidemiological studies, one ejaculate is a sufficient indicator of semen quality in a group of subjects. In a clinical situation, when the question is whether the semen quality is normal or not, the first ejaculate will, in at least 85% of cases, give a correct overall conclusion.

Project description:Artificial insemination (AI) is the preferred method for reproduction in the majority of the intensive pig production systems Worldwide. To this end, fresh extended ready-to-use semen doses are either purchased from AI-centres or produced by boars kept on-farm. For profitable semen production, it is necessary to obtain a maximum amount of high quality semen from each boar. This paper reviews current knowledge on factors that may affect semen quality by influencing the boar or the semen during processing. Genetic markers could be used for early detection of boars with the highest fertility potential. Genetic selection for fast growth might jeopardize semen quality. Early detection of boars no longer fit for semen production might be possible by ultrasonography of the testes. Seasonal variation in sperm quality could be associated with changes in photoperiod and heat stress during summer. Comfortable housing, with appropiate bedding material to avoid locomotion problems is essential. In some areas, cooling systems may be necessary to avoid heat stress. The sperm quality can be manipulated by feeding strategies aiming, for instance, to increase sperm resistance to oxidative stress and extend storage duration. High collection frequency will negatively influence sperm quality. Also, if collection is not hygienically performed it will result in bacterial contamination of the semen doses. The concern over bacterial contamination has risen not only because of its negative effect on semen quality but also due to the detection of antimicrobial resistance in isolates from extended semen. Moreover, bacterial and viral pathogens must be monitored because they affect semen production and quality and constitute a risk of herd infection. During processing, boar sperm are submitted to many stress factors that can cause oxidative stress and capacitation-like changes potentially reducing their fertility potential. Dilution rate or dilution temperature affects the quality of the semen doses. Some packaging might preserve semen better than others and some plastic components might be toxic for sperm. Standard operation procedures and quality assurance systems in AI centres are needed.
