A co-fermentation strategy to consume sugar mixtures effectively.

ABSTRACT: We report a new approach for the simultaneous conversion of xylose and glucose sugar mixtures into products by fermentation. The process simultaneously uses two substrate-selective strains of Escherichia coli, one which is unable to consume glucose and one which is unable to consume xylose. The xylose-selective (glucose deficient) strain E. coli ZSC113 has mutations in the glk, ptsG and manZ genes while the glucose-selective (xylose deficient) strain E. coli ALS1008 has a mutation in the xylA gene. By combining these two strains in a single process, xylose and glucose are consumed more quickly than by a single-organism approach. Moreover, we demonstrate that the process is able to adapt to changing concentrations of these two sugars, and therefore holds promise for the conversion of variable sugar feed streams, such as lignocellulosic hydrolysates.

Children's propensity to consume sugar and fat predicts regular alcohol consumption in adolescence.

Project description:OBJECTIVE:The present study investigated the association between sugar and fat intake in childhood in relation to alcohol use in adolescence. We hypothesized that early exposure to diets high in fat and sugar may affect ingestive behaviours later in life, including alcohol use.Design/Setting/SubjectsChildren from the European IDEFICS/I.Family cohort study were examined at ages 5-9 years and followed up at ages 11-16 years. FFQ were completed by parents on behalf of children, and later by adolescents themselves. Complete data were available in 2263 participants. Children's propensities to consume foods high in fat and sugar were calculated and dichotomized at median values. Adolescents' use of alcohol was classified as at least weekly v. less frequent use. Log-binomial regression linked sugar and fat consumption in childhood to risk of alcohol use in adolescence, adjusted for relevant covariates. RESULTS:Five per cent of adolescents reported weekly alcohol consumption. Children with high propensity to consume sugar and fat were at greater risk of later alcohol use, compared with children with low fat and low sugar propensity (relative risk=2·46; 95 % CI 1·47, 4·12), independent of age, sex and survey country. The association was not explained by parental income and education, strict parenting style or child's health-related quality of life and was only partly mediated by sustained consumption of sugar and fat into adolescence. CONCLUSIONS:Frequent consumption of foods high in fat and sugar in childhood predicted regular use of alcohol in adolescence.

Interactions in plasticizer mixtures used for sugar replacement.

Project description:In our quest for novel ingredients to be used in sugar replacement strategies, we have investigated the thermodynamics of polycarboxylic acids, such as citric acid. We have demonstrated the applicability of the Flory-Huggins (FH) theory to describe the thermodynamics of polycarboxylic acids solutions. Moreover, for citric acid we can describe the complete phase diagram with the theory. It shows that polycarboxylic acids have similar plasticizing and hygroscopic properties as sugars and polyols. Regarding mixtures of polycarboxylic acids and carbohydrates, the FH theory is able to describe a) the water activity of the mixtures, b) the solubility of ternary mixtures of acids and sugars, c) the lowering of the deliquescence point for binary mixtures of crystals, and d) the melting point depression in eutectic mixtures. Unexpectingly, our investigations show there is a strong non-zero FH interaction parameter between carboxylic acids and carbohydrates. In our prior sugar replacement strategy we have assumed zero interactions between plasticizers. Here, we will readdress this assumption. Carefull investigations of solid-liquid equilibrium of eutectic mixtures involving polycarboxylic acids and/or carbohydrates, shows nearly zero interaction in eutectic mixtures consisting only of two carbohydrates or two polycarboxylic acids. We now hold the hypothesis that there is strong non-zero interaction if the mixture contains plasticizers strongly differing in the amount of hydrogen bonding groups. This strong interaction explains why these mixtures, like polycarboxylic acids and carbohydrates, are excellent candidates as deep eutectic solvents. Furthermore, we conclude that polycarboxylic acids are useful additions to the toolbox of sugar replacers, albeit that there are some limitations to their amounts used.

Lactate oxidase/catalase-displaying nanoparticles efficiently consume lactate in the tumor microenvironment to effectively suppress tumor growth.

Project description:The aggressive proliferation of tumor cells often requires increased glucose uptake and excessive anaerobic glycolysis, leading to the massive production and secretion of lactate to form a unique tumor microenvironment (TME). Therefore, regulating appropriate lactate levels in the TME would be a promising approach to control tumor cell proliferation and immune suppression. To effectively consume lactate in the TME, lactate oxidase (LOX) and catalase (CAT) were displayed onto Aquifex aeolicus lumazine synthase protein nanoparticles (AaLS) to form either AaLS/LOX or AaLS/LOX/CAT. These complexes successfully consumed lactate produced by CT26 murine colon carcinoma cells under both normoxic and hypoxic conditions. Specifically, AaLS/LOX generated a large amount of H2O2 with complete lactate consumption to induce drastic necrotic cell death regardless of culture condition. However, AaLS/LOX/CAT generated residual H2O2, leading to necrotic cell death only under hypoxic condition similar to the TME. While the local administration of AaLS/LOX to the tumor site resulted in mice death, that of AaLS/LOX/CAT significantly suppressed tumor growth without any severe side effects. AaLS/LOX/CAT effectively consumed lactate to produce adequate amounts of H2O2 which sufficiently suppress tumor growth and adequately modulate the TME, transforming environments that are favorable to tumor suppressive neutrophils but adverse to tumor-supportive tumor-associated macrophages. Collectively, these findings showed that the modular functionalization of protein nanoparticles with multiple metabolic enzymes may offer the opportunity to develop new enzyme complex-based therapeutic tools that can modulate the TME by controlling cancer metabolism.

Functionally redundant but dissimilar microbial communities within biogas reactors treating maize silage in co-fermentation with sugar beet silage.

Project description:Numerous observations indicate a high flexibility of microbial communities in different biogas reactors during anaerobic digestion. Here, we describe the functional redundancy and structural changes of involved microbial communities in four lab-scale continuously stirred tank reactors (CSTRs, 39°C, 12 L volume) supplied with different mixtures of maize silage (MS) and sugar beet silage (SBS) over 80 days. Continuously stirred tank reactors were fed with mixtures of MS and SBS in volatile solid ratios of 1:0 (Continuous Fermenter (CF) 1), 6:1 (CF2), 3:1 (CF3), 1:3 (CF4) with equal organic loading rates (OLR 1.25 kgVS m(-3) d(-1) ) and showed similar biogas production rates in all reactors. The compositions of bacterial and archaeal communities were analysed by 454 amplicon sequencing approach based on 16S rRNA genes. Both bacterial and archaeal communities shifted with increasing amounts of SBS. Especially pronounced were changes in the archaeal composition towards Methanosarcina with increasing proportion of SBS, while Methanosaeta declined simultaneously. Compositional shifts within the microbial communities did not influence the respective biogas production rates indicating that these communities adapted to environmental conditions induced by different feedstock mixtures. The diverse microbial communities optimized their metabolism in a way that ensured efficient biogas production.

Parallel evolution of the make-accumulate-consume strategy in Saccharomyces and Dekkera yeasts.

Project description:Saccharomyces yeasts degrade sugars to two-carbon components, in particular ethanol, even in the presence of excess oxygen. This characteristic is called the Crabtree effect and is the background for the 'make-accumulate-consume' life strategy, which in natural habitats helps Saccharomyces yeasts to out-compete other microorganisms. A global promoter rewiring in the Saccharomyces cerevisiae lineage, which occurred around 100 mya, was one of the main molecular events providing the background for evolution of this strategy. Here we show that the Dekkera bruxellensis lineage, which separated from the Saccharomyces yeasts more than 200 mya, also efficiently makes, accumulates and consumes ethanol and acetic acid. Analysis of promoter sequences indicates that both lineages independently underwent a massive loss of a specific cis-regulatory element from dozens of genes associated with respiration, and we show that also in D. bruxellensis this promoter rewiring contributes to the observed Crabtree effect.

Characterization of eutectic mixtures of sugars and sugar-alcohols for cryopreservation.

Project description:Natural Deep Eutectic Systems (NADES) composed of sugar and sugar alcohols have been studied and applied in a variety of biological applications. Understanding their interaction with water across dilution and temperature is inherently important for maximizing the utility of NADES. Herein a wide range of sugar:sugar-alcohol molar ratios were synthesized and characterized by viscosity, molar excess volume, differential scanning calorimetry, water activity, and confocal Raman cryomicroscopy. NADES were found to have greater viscosity, reduced heat of fusion, greater absolute molar excess volume, lower water activity, and stronger hydrogen bonding of water than non-NADES mixtures. This is hypothesized to be due to cumulatively stronger hydrogen bonding interactions between components in pure and diluted NADES with the strongest interactions in the water-rich region. This work provides useful data and further understanding of hydrogen bonding interaction strength for a wide range of molar ratios in pure to well-diluted forms.

Adaptability of the Saccharomyces cerevisiae yeasts to wine fermentation conditions relies on their strong ability to consume nitrogen.

Project description:Saccharomyces cerevisiae strains are genetically diverse, largely as a result of human efforts to develop strains specifically adapted to various fermentation processes. These adaptive pressures from various ecological niches have generated behavioral differences among these strains, particularly in terms of their nitrogen consumption capacities. In this work, we characterize this phenotype by the specific quantity of nitrogen consumed under oenological fermentation conditions using a new approach. Indeed, unlike previous studies, our experiments were conducted in an environment containing excess nitrogen, eliminating the nitrogen limitation/starvation factor that is generally observed in fermentation processes. Using these conditions, we evaluated differences in the nitrogen consumption capacities for a set of five strains from diverse origins. The strains presented extremely different phenotypes and variations in their capacities to take up nitrogen from a wine fermentation environment. These variations reflect the differences in the nitrogen uptake capacities between wine and non-wine strains. Finally, the strains differed in their ability to adapt to the nitrogen composition of the environment, leading to variations in the cellular stress states, fermentation performances and the activity of the nitrogen sensing signaling pathway.
