ABSTRACT: On exposure to dichlorodicyanoquinone in wet dichloromethane at room temperature, equatorial 4-O-benzyl ethers are removed with moderate selectivity in the presence of other benzyl ethers in glycopyranosides and glycothiopyranosides.

Project description:In the title compound, C14H20O5, an inter-mediate in the synthesis of oligosaccharides, the glycosidic [H-C-O-C(H3)] torsion angle ?H is 52.3° and the exo-cyclic [H-C-O-C(H2)] torsion angle ?H is -11.7°. The hexa-pyran-ose ring has a chair conformation. In the crystal, mol-ecules are linked by O-H?O hydrogen bonds, forming chains propagating along [010]. Enclosed within the chains are R 3 (3)(12) ring motifs involving three mol-ecules. The chains are linked via C-H?? inter-actions, forming a three-dimensional network.

Project description:Oxidative activation of benzyl or cinnamyl ether bearing allylsilane derivatives using a catalytic amount of DDQ and 2 equivalents of CAN in the presence of PPTS provided functionalized 4-methylenetetrahydropyrans in good yields and excellent diastereoselectivity. The reaction could be applied to the synthesis of a variety of substituted tetrahydropyran derivatives.

Visible-Light-Mediated Oxidative Debenzylation Enables the Use of Benzyl Ethers as Temporary Protecting Groups.

Project description:The cleavage of benzyl ethers by catalytic hydrogenolysis or Birch reduction suffers from poor functional group compatibility and limits their use as a protecting group. The visible-light-mediated debenzylation disclosed here renders benzyl ethers temporary protective groups, enabling new orthogonal protection strategies. Using 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) as a stoichiometric or catalytic photooxidant, benzyl ethers can be cleaved in the presence of azides, alkenes, and alkynes. The reaction time can be reduced from hours to minutes in continuous flow.

Beyond benzyl grignards: facile generation of benzyl carbanions from styrenes.

Project description:Benzylic functionalization is a convenient approach towards the conversion of readily available aromatic hydrocarbon feedstocks into more useful molecules. However, the formation of carbanionic benzyl species from benzyl halides or similar precursors is far from trivial. An alternative approach is the direct reaction of a styrene with a suitable coupling partner, but these reactions often involve the use of precious-metal transition-metal catalysts. Herein, we report the facile and convenient generation of reactive benzyl anionic species from styrenes. A Cu(I)-catalyzed Markovnikov hydroboration of the styrenic double bond by using a bulky pinacol borane source is followed by treatment with KOtBu to facilitate a sterically induced cleavage of the C-B bond to produce a benzylic carbanion. Quenching this intermediate with a variety of electrophiles, including CO(2), CS(2), isocyanates, and isothiocyanates, promotes C-C bond formation at the benzylic carbon atom. The utility of this methodology was demonstrated in a three-step, two-pot synthesis of the nonsteroidal anti-inflammatory drug (±)-flurbiprofen.

Synthesis of alkynyl ethers and low-temperature sigmatropic rearrangement of allyl and benzyl alkynyl ethers.

Project description:Alpha-alkoxy ketones 3 can be transformed into 1-alkynyl ethers 5 by a two-step procedure involving formation of the enol triflate or phosphate and base-induced elimination. Performing the same reaction sequence with allylic alcohols (R2OH, R2 = allyl) furnishes instead gamma,delta-unsaturated carboxylic acid derivatives 6, derived from [3,3]-sigmatropic rearrangement of the intermediate allyl alkynyl ethers at -78 degrees C and trapping of the subsequently formed ketene with nucleophiles (Nu-H). Benzyl alkynyl ether 5 (R2 = benzyl) rearranges to indanone 7 upon heating to 60 degrees C.

Project description:The title compound, C13H24O9·H2O, a structural model for part of bacterial O-anti-gen polysaccharides from Shigella flexneri and Escherichia coli, crystallizes with four independent disaccharide mol-ecules and four water mol-ecules in the asymmetric unit. The conformation at the glycosidic linkage joining the two rhamnosyl residues is described by the torsion angles φH of 39, 30, 37 and 37°, and ψH of -32, -35, -31 and -32°, which are the major conformation region known to be populated in an aqueous solution. The hexo-pyran-ose rings have the 1 C 4 chair conformation. In the crystal, the disaccharide and water mol-ecules are associated through O-H⋯O hydrogen bonds, forming a layer parallel to the bc plane. The layers stack along the a axis via hydro-phobic inter-actions between the methyl groups.

A New Method for the Cleavage of Nitrobenzyl Amides and Ethers.

Project description:A mild and efficient o- and p-nitrobenzyl cleavage protocol was developed. o- and p-Nitrobenzyl groups were easily removed from a variety of substrates using 20% aqueous NaOH in methanol at 75 °C, presumably via oxidation at the benzylic position by oxygen dissolved in the solution. These easily introducible and removable nitrobenzyl groups can serve as valuable protecting groups for the synthesis of multifunctional, complex molecules.
