Bio-Functional, Lanthanide-Labeled Polymer Particles by Seeded Emulsion Polymerization and their Characterization by Novel ICP-MS Detection.

ABSTRACT: We present the synthesis and characterization of monodisperse, sub-micron poly(styrene) (PS) particles loaded with up to and including 10(7) lanthanide (Ln) ions per particle. These particles have been synthesized by seeded emulsion polymerization with a mixture of monomer and a pre-formed Ln complex, and analyzed on a particle-by-particle basis by a unique inductively coupled plasma mass cytometer. Seed particles were prepared by surfactant-free emulsion polymerization (SFEP) to obtain large particle sizes in aqueous media. Extensive surface acid functionality was introduced using the acid-functional initiator ACVA, either during seed latex synthesis or in the second stage of polymerization. The loading of particles with three different Ln ions (Eu, Tb, and Ho) has proven to be close to 100 % efficient on an individual and combined basis. Covalent attachment of metal-tagged peptides and proteins such as Neutravidin to the particle surface was shown to be successful and the number of bound species can be readily determined. We believe these particles can serve as precursors for multiplexed, bead-based bio-assays utilizing mass cytometric detection.

Project description:Starch-based emulsion microgel particles with different starch (15 and 20 wt %) and oil contents (0-15 wt %) were synthesized, and their lubrication performance under physiological conditions was investigated. Emulsion microgels were subjected to skin mimicking or oral cavity mimicking conditions, i.e., smooth hydrophobic polydimethylsiloxane ball-on-disc tribological tests, in the absence or presence of salivary enzyme (?-amylase). In the absence of enzyme, emulsion microgel particles (30-60 vol % particle content) conserved the lubricating properties of emulsion droplets, providing considerably lower friction coefficients (? ? 0.1) in the mixed lubrication regime compared to plain microgel particles (0 wt % oil). Upon addition of enzyme, the lubrication performance of emulsion microgel particles became strongly dependent on the particles' oil content. Microgel particles encapsulating 5-10 wt % oil showed a double plateau mixed lubrication regime having a lowest friction coefficient ? ? 0.03 and highest ? ? 0.1, the latter higher than with plain microgel particles. An oil content of 15 wt % was necessary for the microgel particles to lubricate similarly to the emulsion droplets, where both systems showed a normal mixed lubrication regime with ? ? 0.03. The observed trends in tribology, theoretical considerations, and the combined results of rheology, light scattering, and confocal fluorescence microscopy suggested that the mechanism behind the low friction coefficients was a synergistic enzyme- and shear-triggered release of the emulsion droplets, improving lubrication. The present work thus demonstrates experimentally and theoretically a novel biolubricant additive with stimuli-responsive properties capable of providing efficient boundary lubrication between soft polymeric surfaces. At the same time, the additive should provide an effective delivery vehicle for oil soluble ingredients in aqueous media. These findings demonstrate that emulsion microgel particles can be developed into multifunctional biolubricant additives for future use in numerous soft matter applications where both lubrication and controlled release of bioactives are essential.

Project description:A new analytical procedure for the speciation of chromium (Cr) in plants by high performance liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) was developed using a strong anion-exchange Mono Q column for the separation of the Cr species. To optimize the analytical procedure, Cr complexes were first synthesized from Cr-nitrate with the addition of an excess of ligand (90°C). Cr-oxalate, Cr-malate, Cr-citrate, Cr-aconitate and Cr-quinate complexes and Cr-nitrate (pH 6.5) were chromatographically separated from Cr(VI) by applying linear gradient elution from 100% water to 100% NH4Cl at a flow rate of 1.5 ml min-1 in 10 min. The column recoveries ranged from 100 to 104%. The exception was Cr-aconitate (column recovery 33%), where a quantitative synthesis was not possible. Good repeatability of the measurements (relative standard deviations better than ± 3%) and low limits of detection (below 0.37 ng ml-1 Cr) were achieved for the individual Cr species. The developed analytical procedure was applied to Cr speciation for dandelions (Taraxacum officinale) grown in soil with a high Cr content and a study of the uptake and metabolism of Cr species in dandelions grown in soil with a low Cr content treated with solutions of Cr(VI) or Cr-nitrate (5000 ng ml-1 Cr, pH 6.5) for 48 h. The separated Cr species were quantified by post-column isotope dilution ICP-MS, while the identification was based on retention times and was also supported by mass spectra obtained with high resolution mass spectrometry (HR-MS). The data indicate that for dandelions grown in Cr-rich soil and that treated with Cr-nitrate (pH 6.5), the Cr was mainly accumulated in the roots, while in plants treated with Cr(VI) (pH 6.5), the Cr was evenly distributed between the roots and the leaves. The Cr species found in dandelion roots and leaves were Cr-aconitate, Cr-malate, and Cr-quinate. The results revealed that Cr(VI) was completely reduced and metabolized to Cr(III) complexes. LA-ICP-MS data showed that the Cr in a leaf of dandelion grown in Cr-rich soil was localized mainly at the apex of the leaf.

Project description:Incorporating unmodified silica nanoparticles onto polymer latexes to fabricate aqueous polymer dispersions without relying on electrostatic attraction during the Pickering emulsion polymerization process still faces challenges. For negatively charged silica nanoparticles to successfully adsorb onto polymer latexes, particularly in an anionic initiator emulsion polymerization system, they have remained elusive without the use of auxiliary monomers and cationic initiators. This study investigates various experimental parameters, such as emulsion polymerization temperature, monomer solubility, salt concentration, and cation type, to elucidate the factors influencing the adsorption of unmodified silica nanoparticles in Pickering emulsion polymerization. While poly(methyl methacrylate) (PMMA)/SiO2 hybrid latexes can be obtained under pH conditions of 5-6 and at temperatures of 65 °C or below, the loading rate of silica nanoparticles decreases as the reaction temperature increases, resulting in bare PMMA latexes without silica nanoparticle adsorption at temperatures exceeding 70 °C. Introducing styrene (St) into the monomer mixture with methyl methacrylate in a ratio of up to 10 wt % leads to a gradual decrease in silica nanoparticle loading rate, from 27.3 to 8.2 wt %, attributed to the low solubility of St in water. Furthermore, the presence of sodium ions (Na+) is found to be crucial for silica nanoparticle adsorption onto PMMA latexes, as the sodium ions have a stabilizing effect on both the silica nanoparticles and the silica nanoparticle-armored latexes. These findings highlight the complex nature of Pickering emulsion polymerization in the presence of unmodified silica nanoparticles, demonstrating that the loading rate of silica nanoparticles onto polymer latexes is influenced by various factors. These insights pave the way for developing aqueous polymer dispersions with high silica nanoparticle loading rates onto polymer latexes, which is a desirable trait in the coating industry.
