N-(Pyrimidin-2-yl)aniline.

ABSTRACT: There are two molecules in the asymmetric unit of the title compound, C(10)H(9)N(3), with inter-ring dihedral angles of 31.1?(1) and 35.3?(1)°. The bridging C-N-C bond angles are 128.2?(1) and 129.1?(1)°. In the crystal, the two independent mol-ecules are linked into a dimer by two N-H?N hydrogen bonds.

Project description:Four independent mol-ecules comprise the asymmetric unit of the title co-crystal, C(10)H(9)N(3)·C(7)H(5)NO(4), two for each component. Small conformational differences are noted for the benzoic acid derivatives, notably in the twists of the carb-oxy-lic acid residue out of the plane of the benzene ring to which it is connected [torsion angles = 167.62?(17) and 174.54?(17)°]. In the aniline derivative, the major difference is observed in the dihedral angles formed between the CN(3) and phenyl least-squares planes [1.51?(5) and 6.25?(6)°]. Pairs of mol-ecules associate via O-H?N and N-H?O hydrogen bonds leading to eight-membered {?HOCO?HNCN} hetero-synthons. The two-mol-ecule aggregates are consolidated in the crystal structure by C-H?O(nitro) and ?-? inter-actions [shortest centroid-centroid distance between benzene rings = 3.6242?(10)?Å].

Optimization of 3-Pyrimidin-4-yl-oxazolidin-2-ones as Allosteric and Mutant Specific Inhibitors of IDH1

Project description:High throughput screening and subsequent hit validation identified 4-isopropyl-3-(2-((1-phenylethyl) amino)pyrimidin-4-yl)oxazolidin-2-one as a potent inhibitor of IDH1R132H. Synthesis of the 4 separate diastereomers identified the (S,S)-diastereomer (IDH125) as the most potent isomer. This also showed reasonable cellular activity and excellent selectivity vs IDH1wt. Initial SAR exploration identified the key tolerances and potential for optimization. X-ray crystallography identified a functionally relevant allosteric binding site amenable to inhibitors which can penetrate the blood-brain barrier, and aided rational optimization. Potency improvement and modulation of the physico-chemical properties identified (S)-3-(2-(((S)-1-(5-(4-fluoro-3-methylphenyl)pyrimidin-2-yl)ethyl)amino)pyrimidin-4-yl)-4-isopropyloxazolidin-2-one (IDH889) with good in-vivo exposure and in-vivo activity in a mutant IDH1 xenograft mouse model. identified the (S,S)-diastereomer (IDH125) as the most potent isomer. Project description:Bis[2-(4,6-dimethyl-pyrimidin-2-ylsulfan-yl)eth-yl]amine under hydro-thermal conditions has unexpectedly been transformed into the title compound, C(32)H(44)N(10)S(4). In the title mol-ecule, the zigzag 3,10-diaza-6,7-disulfanyldodecyl skeleton has two dimethyl-pyrimidinylsulfanyl groups at both ends, and the aza atoms each carry a dimethyl-pyrimidinyl unit. The N atoms in the skeleton show a planar coordination.
