ABSTRACT: In the penta-cyclic triterpenoide skeleton of the title mol-ecule, C(27)H(43)NO(2) [systematic name: (3E,3aS,5aR,5bR,7aR,11R,11aR,11bR,13aR,13bR)-5a,5b,10,10,13b-penta-methyl-icosa-hydro-1H-11,7a-(epoxy-methano)cyclo-penta-[a]chrysen-3-one oxime], the five-membered ring A has an envelope conformation, while the six-membered rings B-E adopt chair conformations. Rings A and B are cis-fused. The hydroximino group has an E configuration. Strong inter-molecular O-H?O hydrogen bonds link the mol-ecules into helical chains.

Project description:The title triterpene, C(30)H(50)O(2), is an 18α-oleanane derivative prepared by the Wagner-Meerwein rearrangement of betulin with Bi(OTf)(3).xH(2)O (OTF is trifluoromethanesulfonate). There are two symmetry-independent mol-ecules in the asymmetric unit that show no significant differences concerning bond lengths and angles. The conformation of the six-membered rings is close to a chair form, while the five-membered epoxide rings adopt envelope conformations. All rings are trans-fused. In the crystal, mol-ecules are held together by O-H⋯O hydrogen bonds. A quantum-mechanical ab initio Roothan Hartree-Fock calculation on the isolated mol-ecule gives values for bond lengths and valency angles close to the experimental values. The calculations also reproduce well the mol-ecular conformation with calculated puckering parameters that match well the observed values.

Oxidative Functionalization of Trinor-18α-olean-17(22)-ene Derivatives. Annulation of the E-Ring by an Intramolecular Aldol Reaction.

Project description:cis-Dihydroxylation of trinor-18α-olean-17(22)-ene 2 with osmium tetroxide led to diol 9. Its cleavage with lead tetraacetate gave tetracyclic ketoaldehyde 10. By comparison, the ozonation of trinor-18α-olean-17(22)-ene 2 in the presence of p-toluenesulfonic acid gave the corresponding ketoacetal 12. Both products were subjected to an intramolecular aldol reaction under the acidic conditions and yielded unusual triterpenes bearing a bicyclo[4.3.1]decane fragment (22). Further manipulation of the protective groups afforded compounds useful in triterpene synthesis, especially in the preparation of potentially biologically active saponins based on a tetracyclic terpene core.

Project description:In the title mol-ecule, C13H9NO, the fluorene system and the oxime group non-H atoms are essentially coplanar, with a maximum deviation from the fluorene mean plane of 0.079 (2) Å for the oxime O atom. A short intra-molecular C-H⋯O generates an S(6) ring. In the crystal, mol-ecules related by a twofold screw axis are connected by O-H⋯N hydrogen bonds, forming [100] chains Within these chains, mol-ecules related by a unit translation along [100] show π-π stacking inter-actions between their fluorene ring systems with an inter-planar distance of 3.347 (2) Å. The dihedral angle between the fluorene units of adjacent mol-ecules along the helix is 88.40 (2)°. There is a short C-H⋯π contact between the fluorene groups belonging to neighbouring chains.

The five-membered pyrrolidine ring adopts a twist conformation on the N-C(tetra-substituted) bond. In the crystal, mol-ecules are linked via pairs of N-H⋯O hydrogen bonds, forming inversion dimers with an R 2 (2)(8) ring motif. The dimers are linked via pairs of C-H⋯O hydrogen bonds, forming ribbons enclosing R 2 (2)(12) ring motifs lying in a plane parallel to (01-1).

Differential effects of the novel neurosteroid hypnotic (3β,5β,17β)-3-hydroxyandrostane-17-carbonitrile on electroencephalogram activity in male and female rats.

Project description:BackgroundWe recently showed that a neurosteroid analogue, (3β,5β,17β)-3-hydroxyandrostane-17-carbonitrile (3β-OH), induced hypnosis in rats. The aim of the present study was to evaluate the hypnotic and anaesthetic potential of 3β-OH further using electroencephalography.MethodsWe used behavioural assessment and cortical electroencephalogram (EEG) spectral power analysis to examine hypnotic and anaesthetic effects of 3β-OH (30 and 60 mg kg-1) administered intraperitoneally or intravenously to young adult male and female rats.ResultsWe found dose-dependent sex differences in 3β-OH-induced hypnosis and EEG changes. Both male and female rats responded similarly to i.p. 3β-OH 30 mg kg-1. However, at the higher dose (60 mg kg-1, i.p.), female rats had two-fold longer duration of spontaneous immobility than male rats (203.4 [61.6] min vs 101.3 [32.1] min), and their EEG was suppressed in the low-frequency range (2-6 Hz), in contrast to male rats. Although a sex-dependent hypnotic effect was not confirmed after 30 mg kg-1 i.v., female rats appeared more sensitive to 3β-OH with relatively small changes within delta (1-4 Hz) and alpha (8-13 Hz) bands. Finally, 3β-OH had a rapid onset of action and potent hypnotic/anaesthetic effect after 60 mg kg-1 i.v. in rats of both sexes; however, all female rats and only half of the male rats reached burst suppression, an EEG pattern usually associated with profound inhibition of thalamocortical networks.ConclusionsBased on its behavioural effects and EEG signature, 3β-OH is a potent hypnotic in rats, with female rats being more sensitive than male rats.

Project description:The title compound, C(12)H(13)NO(2), represents a conformationally restricted 2-pyridone analogue of 1,4-dihydro-pyridine-type calcium antagonists and was selected for a crystal structure determination in order to explore some aspects of drug-receptor inter-action. In the mol-ecule, two stereogenic centres are of opposite chirality, whereas a racemate occurs in the crystal. It was found that the formally aminic N atom of the heterocycle is essentially sp(2)-hybridized with the lone-pair electrons partially delocalized through conjugation with the adjacent carbonyl bond. As a result, the central pyridone ring assumes an unsymmetrical half-chair conformation. The critical 4-phenyl ring is fixed in a pseudo-axial and perpendicular orientation [dihedral angle 85.8?(1)°] with respect to the pyridone ring via an oxygen bridge. In the crystal a pair of centrosymmetric N-H?O hydrogen bonds connect mol-ecules of opposite chirality into a dimer. The dimers are packed by hydrophobic van der Waals inter-actions.
