These chains are also linked through O-H?O hydrogen bonds thus building up a supra-molecular two-dimensional framework extending parallel to (001). The organic cations cross-link the anionic layers by way of multiple N-H?O inter-actions, leading to a cohesive network.

Project description:In the title compound, C(7)H(10)N(+)·H(2)PO(4) (-)·H(3)PO(4), there is a clear distinction between the P-O/P=O and P-OH bond lengths. In the crystal, the H(2)PO(4) (-) anions and H(3)PO(4) mol-ecules are linked by O-H⋯O hydrogen bonds, leading to layers propagating in the bc plane. The organic cations are located between these layers and inter-act with them by way of N-H⋯O hydrogen bonds.

Project description:The asymmetric unit of the title compound, C(6)H(5)N(2) (+)·H(2)PO(4) (-)·C(6)H(4)N(2)·H(3)PO(4), contains one 4-cyano-pyridinium cation, one H(2)PO(4) (-) anion, one independent isonicotinonitrile mol-ecule and one independent H(3)PO(4) mol-ecule. The dihedral angle between the mean planes of the separate protonated and unprotonated pyridine rings is 9.93?(8)°. In the crystal, N-H?O and O-H?N hydrogen bonds and weak C-H?O and C-H?N inter-molecular inter-actions connect the organic mol-ecules into a two-dimensional network parallel to the ac plane. O-H?O hydrogen-bonding inter-actions involving the H(2)PO(4) (-) anions and H(3)PO(4) mol-ecules provide additional support from the inorganic groups Weak ?-? stacking inter-actions between the pyridine rings of neighbouring organic mol-ecules [centroid-centroid distances = 3.711?(4) and 3.784?(2)?Å] further link the layers into a three-dimensional network.

Project description:The title compound, C(10)H(15)N(2) (+)·H(2)PO(4) (-), is built up from 4-phenyl-piperazin-1-ium cations and dihydrogen phosphate anions. The inter-connection between two adjacent anions is assured by two strong O-H⋯O hydrogen bonds, which lead to the formation of infinite wave-like chains which spread along the a axis. The organic cations connect these chains via N-H⋯O hydrogen bonds. The crystal cohesion and stability are ensured by electrostatic and van der Waals inter-actions which, together with N-H⋯O and O-H⋯O hydrogen bonds, build up a two-dimensional network.

Project description:The P atom in the title mol-ecule, C(24)H(28)N(3)O(2)P, is in a distorted tetra-hedral P(=O)(N)(N)(2) environment. The phosphoryl group and the NH unit adopt a syn orientation with respect to each other and the N atoms have sp(2) character. The P-N bonds in the P(O)[N(CH(3))(CH(2)C(6)H(5))](2) unit are shorter than the P-N bond in the C(=O)NHP(=O) fragment. An intra-molecular C-H⋯O hydrogen bond occurs. In the crystal, pairs of P=O⋯H-N hydrogen bonds form centrosymmetric dimers. C-H⋯O contacts are also observed. Four C atoms of two benzene rings are disordered over two alternative sites with an occupancy ratio of 0.523 (12):0.427 (12).

Project description:In the title compound, C(16)H(22)N(3)OP·H(2)O, the P atom adopts a distorted tetra-hedral environment with the bond angles around the P atom in the range 99.98 (7)-116.20 (7)°. The P-N bond length in the [(CH(3))(2)N]P(O) fragment [1.6392 (14) Å] is slightly shorter than two other P-N bonds [1.6439 (15) and 1.6530 (14) Å]. In the (CH(3))(2)NP(O) fragment, one of the methyl groups is syn to the P=O bond, whereas the other one is anti to the P=O bond [C-N-P=O torsion angles = 4.80 (17) and -174.57 (15)°]. In the crystal, the water mol-ecules form hydrogen bonds to the O atoms of the P=O bond of two different mol-ecules and act as acceptors for the two amino H atoms of the same mol-ecule. As a result, chains parallel to [010] are formed.

The hydrogen-bonded dimers are connected by another O-H⋯O hydrogen bond with the hydroxyl groups of the cations, constructing a columner structure along the a axis. A number of C-H⋯O interactions are also present.

Supramolecular Synthon Promiscuity in Phosphoric Acid-Dihydrogen Phosphate Ionic Cocrystals.

Project description:Approximately 80% of active pharmaceutical ingredients (APIs) studied as lead candidates in drug development exhibit low aqueous solubility, which typically results in such APIs being poorly absorbed and exhibiting low bioavailability. Salts of ionizable APIs and, more recently, pharmaceutical cocrystals can address low solubility and other relevant physicochemical properties. Pharmaceutical cocrystals are amenable to design through crystal engineering because supramolecular synthons, especially those sustained by hydrogen bonds, can be anticipated through computational modeling or Cambridge Structural Database (CSD) mining. In this contribution, we report a combined experimental and CSD study on a class of cocrystals that, although present in approved drug substances, remains understudied from a crystal engineering perspective: ionic cocrystals composed of dihydrogen phosphate (DHP) salts and phosphoric acid (PA). Ten novel DHP:PA ionic cocrystals were prepared from nine organic bases (4,4'-bipyridine, 5-aminoquinoline, 4,4'-azopyridine, 1,4-diazabicyclo[2.2.2]octane, piperazine, 1,2-bis(4-pyridyl)ethane, 1,2-bis(4-pyridyl)xylene, 1,2-di(4-pyridyl)-1,2-ethanediol, and isoquinoline-5-carboxylic acid) and one anticonvulsant API, lamotrigine. From the resulting crystal structures and a CSD search of previously reported DHP:PA ionic cocrystals, 46 distinct hydrogen bonding motifs (HBMs) have been identified between DHP anions, PA molecules, and, in some cases, water molecules. Our results indicate that although DHP:PA ionic cocrystals are a challenge from a crystal engineering perspective, they are formed reliably and, given that phosphoric acid is a pharmaceutically acceptable coformer, this makes them relevant to pharmaceutical science.
