ABSTRACT: In the cystal structure of the title compound, (2-hy-droxy-ethyl)trimethylammonium dihydrogen phosphate, C(5)H(14)NO(+)·H(2)PO(4) (-), two anions create dimeric structures via two O-H?O hydrogen bonds. The hydrogen-bonded dimers are connected by another O-H?O hydrogen bond with the hydroxyl groups of the cations, constructing a columner structure along the a axis. A number of C-H?O interactions are also present.

Choline Dihydrogen Phosphate Destabilizes G-Quadruplexes and Enhances Transcription Efficiency <i>In Vitro</i> and in Cells.

Project description:G-quadruplexes in disease-related genes are associated with various biological processes and regulate disease progression. Although methods involving ligands and other techniques are available to stabilize G-quadruplexes, approaches for destabilizing G-quadruplexes remain limited. Here, we evaluated whether G-quadruplexes can be destabilized using choline dihydrogen phosphate (choline dhp), a highly biocompatible hydrated ionic liquid. Circular dichroism spectral measurements at increasing temperatures revealed that choline dhp destabilized G-quadruplexes more effectively than did KCl-containing solutions. Thermodynamic analysis indicated that destabilization occurred via an entropic contribution, suggesting that choline ions did not coordinate with the G-quartets, because of their large radii. Subsequently, plasmid DNAs containing G-quadruplexes were constructed, and transcription reactions were performed in nuclear extracts from living cells. G-quadruplexes repressed transcription, whereas the addition of choline dhp increased transcription. Although ionic liquids often inactivate biomolecules, choline dhp can be used to culture various cells. Furthermore, the transcription of template DNA containing the G-quadruplex was greatly enhanced in living MDA-MD-231 cells (aggressive human breast cancer cells) cultured with choline dhp. Our results show that choline dhp destabilizes G-quadruplexes in cells, indicating that choline dhp can regulate gene expression. Thus, choline dhp may be useful for regulating target disease-related genes.

In 2CADHP, the dihydrogen phosphate anions form a double-stranded anionic chain generated parallel to the [010] direction through O-H...O hydrogen bonds, whereas in 4CADHP they form a two-dimensional supramolecular net extending parallel to the crystallographic (001) plane into which the cations are linked through strong N-H...O hydrogen bonds.

A long chain-like stacking of dihydrogenphosphate anions along the c-axis direction is constructed by O-H⋯O hydrogen bonds. Also along the c-axis direction, π-π stacking between inversion-related pyridinium rings [centroid-centroid distance = 3.8051 (10) Å] forms columnar stacks of cations.

Project description:In the crystal structure of the title compound, C(12)H(12)N(+)·H(2)PO(4) (-), the dihydrogen phosphate anions and the 2-phenyl-anilinium cations are associated via O-H⋯O and N-H⋯O hydrogen bonds so as to build inorganic layers around the x = 1/2 plane. The organic entities are anchored between these layers through C-H⋯O hydrogen bonds, forming a three-dimensional infinite network. The dihedral angle between the aromatic rings is 44.7 (4)°.

Comparable stability of Hoogsteen and Watson-Crick base pairs in ionic liquid choline dihydrogen phosphate.

Project description:The instability of Hoogsteen base pairs relative to Watson-Crick base pairs has limited biological applications of triplex-forming oligonucleotides. Hydrated ionic liquids (ILs) provide favourable environments for a wide range of chemical reactions and are known to impact the stabilities of Watson-Crick base pairs. We found that DNA triplex formation was significantly stabilized in hydrated choline dihydrogen phosphate as compared with an aqueous buffer at neutral pH. Interestingly, the stability of Hoogsteen base pairs was found to be comparable with that of Watson-Crick base pairs in the hydrated IL. Molecular dynamics simulations of a DNA triplex in the presence of choline ions revealed that the DNA triplex was stabilized because of the binding of choline ion around the third strand in the grooves. Our finding will facilitate the development of new DNA materials. Our data also indicate that triplex formation may be stabilized inside cells where choline ions and their derivatives are abundant in vivo.

The anion chains are connected by water mol-ecules, which lie on crystallographic twofold rotation axes, through O-H?O hydrogen bonds. These hydrogen bonds are almost perpendicular to the other hydrogen bonds which create an assembled structure of anions. In addition, C-H?O hydrogen bonds between anions and cations are observed.

2-Amino-pyrimidinium dihydrogen phosphate monohydrate.

Project description:In the title compound, C(4)H(6)N(3) (+)·H(2)O(4)P(-)·H(2)O, the pyrimidin-ium ring is essentially planar, with an r.m.s. deviation of 0.0016 Å. In the structure, pairs of symmetry-related anions are connected into centrosymmetric clusters via strong O-H⋯O hydrogen bonds forming six-membered rings with an R(2) (2)(6) motif. These clusters are inter-connected via water mol-ecules through OW-H⋯O hydrogen bonds, building an infinite layer parallel to the ab plane. Moreover, infinite chains of 2-amino-pyrimidinium cations spread along the a-axis direction. These chains are connected to the inorganic layer through N-H⋯O, C-H⋯O and C-H⋯N hydrogen bonds, which, together with electrostatic and van der Waals inter-actions, contribute to the cohesion and stability of the network in the crystal structure.
