Versatile small-molecule motifs for self-assembly in water and the formation of biofunctional supramolecular hydrogels.

ABSTRACT: This feature article introduces new structural motifs (referred as "samogen") that serve as the building blocks of hydrogelators for molecular self-assembly in water to result in a series of supramolecular hydrogels. Using a compound that consists of two phenylalanine residues and a naphthyl group (also abbreviated as NapFF (1) in this text) as an example of the samogens, we demonstrated the ability of the samogens to convert bioactive molecules into molecular hydrogelators that self-assemble in water to result in nanofibers. By briefly summarizing the properties and applications (e.g., wound healing, drug delivery, controlling cell fate, typing bacteria, and catalysis) of these molecular hydrogelators derived from the samogens, we intend to illustrate the basic requirements and promises of the small-molecule hydrogelators for applications in chemistry, materials science, and biomedicine.

Supramolecular Self-Assembly To Control Structural and Biological Properties of Multicomponent Hydrogels.

Project description:Self-assembled nanofibers are ubiquitous in nature and serve as inspiration for the design of supramolecular hydrogels. A multicomponent approach offers the possibility of enhancing the tunability and functionality of this class of materials. We report on the synergistic multicomponent self-assembly involving a peptide amphiphile (PA) and a 1,3:2,4-dibenzylidene-d-sorbitol (DBS) gelator to generate hydrogels with tunable nanoscale morphology, improved stiffness, enhanced self-healing, and stability to enzymatic degradation. Using induced circular dichroism of Thioflavin T (ThT), electron microscopy, small-angle neutron scattering, and molecular dynamics approaches, we confirm that the PA undergoes self-sorting, while the DBS gelator acts as an additive modifier for the PA nanofibers. The supramolecular interactions between the PA and DBS gelators result in improved bulk properties and cytocompatibility of the two-component hydrogels as compared to those of the single-component systems. The tunable mechanical properties, self-healing ability, resistance to proteolysis, and biocompatibility of the hydrogels suggest future opportunities for the hydrogels as scaffolds for tissue engineering and drug delivery vehicles.

Antiviral supramolecular polymeric hydrogels by self-assembly of tenofovir-bearing peptide amphiphiles.

Project description:The development of long-acting antiviral therapeutic delivery systems is crucial to improve the current treatment and prevention of HIV and chronic HBV. We report here on the conjugation of tenofovir (TFV), an FDA approved nucleotide reverse transcriptase inhibitor (NRTI), to rationally designed peptide amphiphiles (PAs), to construct antiviral prodrug hydrogelators (TFV-PAs). The resultant conjugates can self-assemble into one-dimensional nanostructures in aqueous environments and consequently undergo rapid gelation upon injection into 1× PBS solution to create a drug depot. The TFV-PA designs containing two or three valines could attain instantaneous gelation, with one displaying sustained release for more than 28 days in vitro. Our studies suggest that minor changes in peptide design can result in differences in supramolecular morphology and structural stability, which impacted in vitro gelation and release. We envision the use of this system as an important delivery platform for the sustained, linear release of TFV at rates that can be precisely tuned to attain therapeutically relevant TFV plasma concentrations.

Supramolecular gel formation regulated by water content in organic solvents: self-assembly mechanism and biomedical applications.

Project description:As one of the most important and fruitful methods, supramolecular self-assembly has a significant advantage in designing and fabricating functional soft materials with various nanostructures. In this research, a low-molecular-weight gelator, N,N'-di(pyridin-4-yl)-pyridine-3,5-dicarboxamide (PDA-N4), was synthesized and used to construct self-assembled gels via a solvent-mediated strategy. It was found that PDA-N4 could form supramolecular gels in mixed solvents of water and DMSO (or DMF) at high water fraction (greater than or equal to 50%). By decreasing the water fraction from 50% to 30%, the gel, suspension and solution phases appeared successively, indicating that self-assembled aggregates could be efficiently modulated via water content in organic solvents. Moreover, the as-prepared PDA-N4 supramolecular gels not only displayed solid-like behavior, and pH- and thermo-reversible characteristics, but also showed a solution-gel-crystal transition with the extension of aging time. Further analyses suggested that both the crystal and gel had similar assembled structures. The intermolecular hydrogen bonding between amide groups and the π-π stacking interactions between pyridine groups played key roles in gel formation. Additionally, the release behavior of vitamin B12 (VB12) from PDA-N4 gel (H2O/DMSO, v/v = 90/10) was evaluated, and the drug controlled release process was consistent with a first-order release mechanism. The human umbilical venous endothelial cell culture results showed that the PDA-N4 xerogel has good cytocompatibility, which implied that the gels have potential biological application in tissue engineering and controlled drug release.

Transient Supramolecular Hydrogels Formed by Aging-Induced Seeded Self-Assembly of Molecular Hydrogelators.

Project description:Here, transient supramolecular hydrogels that are formed through simple aging-induced seeded self-assembly of molecular gelators are reported. In the involved molecular self-assembly system, multicomponent gelators are formed from a mixture of precursor molecules and, typically, can spontaneously self-assemble into thermodynamically more stable hydrogels through a multilevel self-sorting process. In the present work, it is surprisingly found that one of the precursor molecules is capable of self-assembling into nano-sized aggregates upon a gentle aging treatment. Importantly, these tiny aggregates can serve as seeds to force the self-assembly of gelators along a kinetically controlled pathway, leading to transient hydrogels that eventually spontaneously convert into thermodynamically more stable hydrogels over time. Such an aging-induced seeded self-assembly process is not only a new route toward synthetic out-of-equilibrium supramolecular systems, but also suggests the necessity of reporting the age of self-assembling building block solutions in other self-assembly systems.

Structuring supramolecular hyaluronan hydrogels via peptide self-assembly for modulating the cell microenvironment.

Project description:The use of synthetic extracellular matrices (ECMs) in fundamental in vitro cell culture studies has been instrumental for investigating the interplay between cells and matrix components. To provide cells with a more native environment in vitro, it is desirable to design matrices that are biomimetic and emulate compositional and structural features of natural ECMs. Here, the supramolecular fabrication of peptide-hyaluronan (HA) hydrogels is presented as potential ECM surrogates, combining native HA and rationally designed cationic amphipatic peptides [(KI)nK, lysine (K), isoleucine (I), n = 2-6] whose mechanical properties and microstructure are tunable by the peptide sequence. (KI)nK peptides adopt β-sheet configuration and self-assemble into filamentous nanostructures triggered by pH or ionic strength. The self-assembly propensity of (KI)nK peptides increases with the sequence length, forming single phase hydrogels (shorter peptides) or with phase separation (longer peptides) in presence of the anionic polyelectrolyte HA through electrostatic complexations. The gel phase formed in (KI)nK-HA complexes exhibits viscoelastic behavior and triggers the formation of human mesenchymal stem cell (MSC) spheroids which disassemble over the time. It is anticipated that these (KI)nK-HA hydrogels with tunable physical and biochemical properties offer a promising platform for in vitro applications and in stem cell therapy.

Multistep sequence-controlled supramolecular polymerization by the combination of multiple self-assembly motifs

Enzyme-instructed self-assembly of the stereoisomers of pentapeptides to form biocompatible supramolecular hydrogels.

Project description:This article reports enzyme-instructed self-assembly (EISA) of stereoisomers of pentapeptides as a simple approach for generating biocompatible supramolecular hydrogels as potential soft bionanomaterials. Peptide-based supramolecular hydrogels are emerging as a new type of biomaterials. The use of tyrosine phosphate offers a trigger for enzymatic hydrogelation, and the incorporation of D-amino acids can increase the proteolytic stability of peptides. This work compared four phosphorpeptides that are stereoisomers in terms of rate of dephosphorylation, proteolytic stability, and cell compatibility. The results show that the naphthyl (Nap)-capped pentapeptides, containing the amino acid sequence of Phe-Phe-Gly-Glu-pTyr, are able to undergo EISA to form the hydrogels consisting the nanofibres of the dephosphorylated pentapeptides. The naphthyl-capped D-phosphopentpeptides, contrasting to a naphthyl-capped D-phosphotripeptide (Nap-D-Phe-D-Phe-D-pTyr), are largely cell compatible. This result, suggesting that the sequence of phophopeptides also dedicates the cell compatibility of the peptide assemblies resulted from EISA, provides useful insights for developing supramolecular hydrogels as potential biomaterials with tailored properties.

Dual self-assembly of supramolecular peptide nanotubes to provide stabilisation in water.

Project description:Self-assembling peptides have the ability to spontaneously aggregate into large ordered structures. The reversibility of the peptide hydrogen bonded supramolecular assembly make them tunable to a host of different applications, although it leaves them highly dynamic and prone to disassembly at the low concentration needed for biological applications. Here we demonstrate that a secondary hydrophobic interaction, near the peptide core, can stabilise the highly dynamic peptide bonds, without losing the vital solubility of the systems in aqueous conditions. This hierarchical self-assembly process can be used to stabilise a range of different β-sheet hydrogen bonded architectures.

Ultrafast Interfacial Self-Assembly toward Supramolecular Metal-Organic Films for Water Desalination.

Project description:Supramolecular metal-organic materials are considered as the ideal candidates for membrane fabrication due to their excellent film forming characteristics, diverse metal centers and ligand sources, and designable structure and function. However, it remains challenging to rapidly construct highly permeable supramolecular metal-organic membranes with high salt rejection. Herein, a novel ultrafast interfacial self-assembly strategy to prepare supramolecular metal-organic films through the strong coordination interaction between highly active 1,3,5-triformylphloroglucinol (TFP) ligands and Fe3+ , Sc3+ , or Cu2+ at the organic-aqueous interface is reported. Benefiting from the self-completing and self-limiting characteristics of this interfacial self-assembly, the new kind of supramolecular membrane with optimized composition can be assembled within 3.5 min and exhibits ultrathin, dense, defect-free features, and thus shows an excellent water permeance (21.5 L m-2 h-1 bar-1 ) with a high Na2 SO4 rejection above 95%, which outperforms almost all of the non-polyamide membranes and commercially available nanofiltration membranes. This strong-coordination interfacial self-assembly method will open up a new way for the development of functional metal-organic supramolecular films for high-performance membrane separation and beyond.

Self-Assembly of Unprotected Dipeptides into Hydrogels: Water-Channels Make the Difference.

Project description:Unprotected dipeptides are attractive building blocks for environmentally friendly hydrogel biomaterials by virtue of their low-cost and ease of preparation. This work investigates the self-assembling behaviour of the distinct stereoisomers of Ile-Phe and Phe-Ile in phosphate buffered saline (PBS) to form hydrogels, using transmission electron microscopy (TEM), attenuated total reflectance infrared spectroscopy (ATR-IR), circular dichroism (CD), and oscillatory rheometry. Each peptide purity and identity was also confirmed by 1 H- and 13 C-NMR spectroscopy and HPLC-MS. Finally, single-crystal XRD data allowed the key interactions responsible for the supramolecular packing into amphipathic layers or water-channels to be revealed. The presence of the latter in the crystal structure is a distinctive feature of the only gelator of this work that self-organizes into stable hydrogels, with fast kinetics and the highest elastic modulus amongst its structural isomers and stereoisomers.
