KNb(1.75)V(0.25)PS(10).

ABSTRACT: The title compound, potassium diniobium vanadium phospho-rus deca-sulfide, KNb(1.75)V(0.25)PS(10), was obtained by reaction of the elements with a eutectic mixture of KCl/LiCl. It is isostructural with the quaternary KNb(2)PS(10), but the Nb sites are occupied by statistically disordered Nb (87.5%) and V (12.5%) atoms. The structure is composed of anionic (∞) (1)[M(2)PS(10)](-) chains (M = Nb/V) separated from each other by K(+) ions. The chain is composed of [MS(8)] distorted bicapped trigonal prisms and [PS(4)] tetra-hedra. There are no inter-chain bonding inter-actions. The crystal used for the X-ray analysis was a racemic twin.

Project description:The reaction of cadmium nitrate and sodium nitrite in the presence of 1,10-phenanthroline yields the mixed nitrate-nitrite title complex, [Cd(NO(2))(1.75)(NO(3))(0.25)(C(12)H(8)N(2))(2)]. The metal ion is bis-chelated by two N-heterocycles as well as by the nitrate/nitrite ions in a distorted dodeca-hedral CdN(4)O(4) coordination environment. One nitrite group is ordered; the other is disordered with respect to a nitrate group (ratio 0.75:0.25) concerning the O atom that is not involved in bonding to the metal ion.

Cubic ZrW(1.75)Mo(0.25)O(8) from a Rietveld refinement based on neutron powder diffraction data.

Project description:The solid solution in the system Zr-Mo-W-O with composition ZrW(1.75)Mo(0.25)O(8) (zirconium tungsten molybdenum octa-oxide) was prepared by solid-state reactions as a polycrystalline material. Its structure has cubic symmetry (space group P2(1)3) at room temperature. The structure contains a network of corner-sharing ZrO(6) octa-hedra (.3. symmetry) and MO(4) (M = W, Mo) tetra-hedra (.3. symmetry). Along the main threefold axis of the cubic unit cell, the MO(4) tetra-hedra are arranged in pairs forming M(2)O(8) units in which the M1O(4) tetra-hedra have larger distortions in terms of bond distances and angles than the M2O(4) tetra-hedra. These units are disordered over two possible orientations, with the M-O(terminal) vectors pointing to the [111] or [] directions. The reversal of the orientations of the M(2)O(8) units results from the concerted flips of these units. The time-averaged proportions of flipped and unflipped M(2)O(8) units were determined and the fraction of unflipped M(2)O(8) units is about 0.95. The order degree of the M(2)O(8) unit orientation is about 0.9. During the reversal process, the M-atom site has a migration about 0.93 Å, one of the O-atom sites has a 0.25 Å migration distance, whereas two other O-atom sites migrate marginally (≃ 0.08 Å). The results prove the constraint strategy to be a reasonable approach based on the ratcheting mechanism.

Redetermination of AgNb(2)PS(10) revealing a silver deficiency.

Project description:In comparison with a previous crystallographic study [Goh et al. (2002 ▶). J. Solid State Chem.168, 119-125] of the title compound, silver diniobium tris-(disulfide) tetra-thio-phosphate(V), that reports a full occupation of the silver position and isotropic displacement parameters for the atoms, the current redetermination reveals a silver deficiency with a site-occupation factor of 0.88 (1) and reports all atoms with anisotropic displacement parameters. The structure of Ag(0.88)Nb(2)PS(10) is composed of (∞) (1)[Nb(2)PS(10)] chains, which are built up from pairs of distorted bicapped trigonal-prismatic [NbS(8)] polyhedra forming [Nb(2)S(12)] dimers and of tetra-hedral [PS(4)] groups. These chains are connected via the statistically disordered Ag(+) ions, forming double layers. Adjacent layers are stacked solely through van der Waals forces into a three-dimensional structure. Short and long Nb-Nb distances [2.880 (1) and 3.770 (2) Å, respectively] alternate along the chain and S(2) (2-) and S(2-) anionic species are observed.

Project description:The quinter-nary thio-phosphate Cs(0.5)Ag(0.5)Nb(2)PS(10), cesium silver tris-(disulfido)[tetra-thio-phosphato(V)]diniobate(IV), has been prepared from the elements using a CsCl flux. The crystal structure is made up of (∞) (1)[Nb(2)PS(10)] chains expanding along [010]. These chains are built up from bicapped trigonal-prismatic [Nb(2)S(12)] units and tetra-hedral [PS(4)] groups and are linked through a linear S-Ag-S bridge, forming a two-dimensional layer. These layers then stack on top of each other, completing the three-dimensional structure with an undulating van der Waals gap. The disordered Cs(+) ions reside on sites with half-occupation in the voids of this arrangement. Short [2.8843 (5) Å] and long [3.7316 (4) Å] Nb-Nb distances alternate along the chains, and anionic S(2) (2-) and S(2-) species are observed. The charge balance of the com-pound can be represented by the formula [Cs(+)](0.5)[Ag(+)](0.5)[Nb(4+)](2)[PS(4) (3-)][S(2) (2-)](3).

SUPERFAMILY 1.75 including a domain-centric gene ontology method.

Project description:The SUPERFAMILY resource provides protein domain assignments at the structural classification of protein (SCOP) superfamily level for over 1400 completely sequenced genomes, over 120 metagenomes and other gene collections such as UniProt. All models and assignments are available to browse and download at http://supfam.org. A new hidden Markov model library based on SCOP 1.75 has been created and a previously ignored class of SCOP, coiled coils, is now included. Our scoring component now uses HMMER3, which is in orders of magnitude faster and produces superior results. A cloud-based pipeline was implemented and is publicly available at Amazon web services elastic computer cloud. The SUPERFAMILY reference tree of life has been improved allowing the user to highlight a chosen superfamily, family or domain architecture on the tree of life. The most significant advance in SUPERFAMILY is that now it contains a domain-based gene ontology (GO) at the superfamily and family levels. A new methodology was developed to ensure a high quality GO annotation. The new methodology is general purpose and has been used to produce domain-based phenotypic ontologies in addition to GO.

Inhibitory effect of S-nitroso-N-acetylpenicillamine on the basolateral 10-pS Cl- channel in thick ascending limb.

Project description:We have previously reported that L-arginine, a nitric oxide synthase substrate, inhibits the basolateral 10-pS Cl- channel through the cGMP/PKG signaling pathway in the thick ascending limb (TAL). As a NO releasing agent, the effect of S-nitroso-N-acetyl-penicillamine (SNAP) on the channel activity was examined in thick ascending limb of C57BL/6 mice in the present study. SNAP inhibited the basolateral 10-pS Cl- channel in a dose-dependent manner with an IC50 value of 6.6 μM. The inhibitory effect of SNAP was abolished not only by NO scavenger (carboxy-PTIO) but also by blockers of soluble guanylate cyclase (ODQ or LY-83583), indicating that the cGMP-dependent signaling pathway is involved. Moreover, the inhibitory effect of SNAP on the channel was strongly attenuated by a protein kinase G (PKG)-specific inhibitor, KT-5823, but not by the PDE2 inhibitor, BAY-60-7550. We concluded that SNAP inhibited the basolateral 10-pS Cl- channels in the TAL through a cGMP/PKG signaling pathway. As the 10-pS Cl- channel is important for regulation of NaCl absorption along the nephron, these data suggest that SNAP might be served as a regulator to prevent high-salt absorption related diseases, such as hypertension.

Project description:Lotilaner ophthalmic solution 0.25% (XDEMVY™), a gamma-aminobutyric acid-gated chloride channel (GABA-Cl) inhibitor selective for mites, is being developed by Tarsus Pharmaceuticals for the treatment of Demodex blepharitis and meibomian gland dysfunction in patients with Demodex lid infestation (Demodex-induced meibomianitis). On 24 July 2023, lotilaner ophthalmic solution 0.25% received its first approval in the USA for the treatment of Demodex blepharitis. The agent is also currently in phase 3 development for Demodex blepharitis in China and phase 2 development for Demodex-induced meibomianitis in the USA. This article summarizes the milestones in the development of lotilaner ophthalmic solution 0.25% leading to this first approval for the treatment of Demodex blepharitis in the USA.
