Project description:Five new species of Scolytus Geoffroy, 1762 (Coleoptera: Curculionidae: Scolytinae) are described from Peru, namely Scolytus woodi, Scolytus carveli, Scolytus vagabundus, Scolytus lindemani, Scolytus mozolevskae .The following new synonym is established: Scolytus angustatus Browne, 1970 (= Scolytus facialis Schedl, 1973, syn. n.) New records of the Scolytus species in Loreto, Junin ,Cusco and Madre de Dios Provinces are given and the biology of the genus representatives is discussed.

A taxonomic monograph of Nearctic Scolytus Geoffroy (Coleoptera, Curculionidae, Scolytinae).

Project description:The Nearctic bark beetle genus Scolytus Geoffroy was revised based in part on a molecular and morphological phylogeny. Monophyly of the native species was tested using mitochondrial (COI) and nuclear (28S, CAD, ArgK) genes and 43 morphological characters in parsimony and Bayesian phylogenetic analyses. Parsimony analyses of molecular and combined datasets provided mixed results while Bayesian analysis recovered most nodes with posterior probabilities >90%. Native hardwood- and conifer-feeding Scolytus species were recovered as paraphyletic. Native Nearctic species were recovered as paraphyletic with hardwood-feeding species sister to Palearctic hardwood-feeding species rather than to native conifer-feeding species. The Nearctic conifer-feeding species were monophyletic. Twenty-five species were recognized. Four new synonyms were discovered: Scolytuspraeceps LeConte, 1868 (= Scolytusabietis Blackman, 1934; = Scolytusopacus Blackman, 1934), Scolytusreflexus Blackman, 1934 (= Scolytusvirgatus Bright, 1972; = Scolytuswickhami Blackman, 1934). Two species were reinstated: Scolytusfiskei Blackman, 1934 and Scolytussilvaticus Bright, 1972. A diagnosis, description, distribution, host records and images were provided for each species and a key is presented to all species.

Collecting and preserving bark and ambrosia beetles (Coleoptera: Curculionidae: Scolytinae & Platypodinae).

Project description:This protocol describes the different methods to collect and preserve bark and ambrosia beetles, detailing collecting tools, recording relevant data, and optimizing step-by-step methods to extract beetles from twigs, branches, bark, and trunks. It elaborates on trapping techniques, tools, lures, baits, and beetle preservation. The main rule of manual collecting is to not attempt to pry the insect out of the wood or bark, but instead, remove the wood/bark away from the beetle: gently and systematically. The main rule of trapping is that there is no general attractant; instead, attractants and traps should reflect the ecology of the targeted beetle taxa.

Molecular cytogenetic studies in the ladybird beetle Henosepilachnaargus Geoffroy, 1762 (Coleoptera, Coccinellidae, Epilachninae).

Project description:The ladybird Henosepilachnaargus Geoffroy, 1762 has been cytogenetically studied. In addition we have conducted a review of chromosome numbers and the chromosomal system of sex determination available in the literature in species belonging to the genus Henosepilachna and in its closely related genus Epilachna. Chromosome number of Henosepilachnaargus was 2n=18, including the sex chromosome pair, a common diploid chromosome number within the tribe Epilachnini. The study of prophase I meiotic chromosomes showed the typical Xyp "parachute" bivalent as in the majority of species of Coccinellidae. C-banding and fluorescent staining with AT-specific DAPI fluorochrome dye have been carried out for the first time in H. argus. C-banding technique revealed that heterochromatic blocks are pericentromerically located and DAPI staining showed that this heterochromatin is AT rich. Fluorescence in situ hybridizations using rDNA and the telomeric TTAGG sequence as probes have been carried out. FISH using rDNA showed that the nucleolar organizing region is located on the short arm of the X chromosome. FISH with the telomeric sequence revealed that in this species telomeres of chromosomes are composed of the pentanucleotide TTAGG repeats. This is the first study on the telomeric sequences in Coccinellidae.

Patterns of Seed Penetration by the Date Stone Beetle Coccotrypes dactyliperda (Coleoptera, Curculionidae, Scolytinae).

Project description:The cryptic spermatophagus date stone beetle (Coccotrypes dactyliperda Fabricius, 1801) tunnels into palm seeds for oviposition and subsequent establishment of brood galleries. Based on behavioral observations this paper describes the biomechanical and gustatory parameters that govern the initial excavation of the tunnels and the subsequent establishment of the galleries. When tunneling into Canary Islands Date Palm (Phoenix canariensis Chabaud, 1882) seeds, penetration principally occurs at the dorsal side of the seed, in particular the groove, which allows the beetle to gain the required traction. Tunneling is executed in a circular fashion with clockwise or counter-clockwise repositioning in approximately one-eighth to one-quarter turns. Biomechanically, the three pairs of legs provide thrust (hind legs), pivoting (mid legs), and lateral control (fore legs). Gustatory cues, the nature of which remains unclear at this point, prevent the beetle from tunneling through to the other side.

Project description:BackgroundIn 2020 and 2021, chalcidoid parasitoids (Hymenoptera, Chalcidoidea) of bark beetles in pine (Pinus spp.) plantations were studied in Bulgaria. Samples (cuttings of stems and branches) of pine trees infested by bark beetles were collected from seven plantations of Pinussylvestris and P.nigra in Bulgaria. From each sampling plot, five cuttings were collected and placed in photoeclectors in laboratory conditions (18-22ºC). Emerged bark beetles and parasitoids were regularly gathered and fixed in ethanol.New informationSix parasitoid species - Dinotiscuscolon, Metacolusazureus, M.unifasciatus, Rhopalicusquadratus, R.tutela (Chalcidoidea: Pteromalidae) and Heydeniapretiosa (Chalcidoidea, Heydeniidae) were reared from five bark beetle hosts (Ipsacuminatus, Pityogenesbistridentatus, Pityophthoruspityographus, Tomicuspiniperda and T.minor). Amongst them, three species (H.pretiosa, M.azureus and R.quadratus) are recorded as new for Bulgarian fauna.

Rapid Molecular Identification of Scolytinae (Coleoptera: Curculionidae).

Project description:Routine identification of bark and ambrosia beetles is done using morphology. For people lacking the necessary taxonomic knowledge, proper identification of a novel specimen can be challenging and time consuming. This study compares the usefulness of four genetic markers (28S, EF-1a, ITS2, and COI) and five primer pairs (D2F1/D3R2, eflafor1/eflarev1, ets149/efa754, ITS2F/ITS2R, and LCO1490/HCO2198) to identify Scolytinae beetles, and outlines a molecular identification strategy, with results possible in two days. Markers COI and EF-1a were selected based on the ability of the respective primers to amplify DNA from multiple genera (Coptoborus, Xyleborus, Hypothenemus, Theoborus, and Araptus) and the ability of the resulting sequences to provide accurate and unambiguous matches in GenBank. BLASTn analysis of EF-1a sequences (both primer pairs) correctly identified four out of the five genera and COI sequences identified at least one sample of every genus tested and was the only primer pair to correctly identify Araptus specimens. Further, 28S sequences successfully identified Coptoborus, Xyleborus, and Theoborus but not Hypothenemus or Araptus. The low number of EF-1a (1), 28S (7), and ITS2 (0) sequences from Araptus individuals present in GenBank compared with COI (137) is likely the reason that only the latter marker was capable of identifying members of this genus. ITS2 sequences were insufficient to identify any of the samples tested. This study also determined the minimum quantity of DNA that could be used for molecular identification. Primers D2F1 and D3R2, which had the highest rate of amplification in all genera tested, could yield an informative sequence with as little as 0.00048 ng of DNA, however, at least 0.0024 ng was needed for reliable amplification.

Five new species of Triotemnus (Coleoptera, Curculionidae, Scolytinae) from Morocco and Yemen.

Project description:Fivenew species of the genus Triotemnus from Morocco and Yemen are described. Triotemnus is a new genus of Scolytinae for the Yemen region. External morphology of the new species and all morphologically related species of the genus were studied. While the new species from Morocco are morphologically similar to the known species from the corresponding region, all three newly described species from Yemen, mainly two of them living in Socotra, are morphologically very different from all other known species of the genus. Geographical distribution and the probability of endemicity are discussed.
