Previous descriptions of adult Austroplatypus incompertus (Schedl) are completed by the addition of descriptions and illustrations of the adults and, in particular, their maxillary palps. I describe and illustrate the non-adult phases of the life cycle and provide a key to the larval instars. The sexual dimorphism of Austroplatypus incompertus is atypical and includes a latitudinal cline which obeys Bergmann's rule. The taxonomic position of the genus within the Platypodinae is clarified. Project description:External secretions play a vital role in external immune defense. However, the functions and components of these exudates are largely unknown in the red palm weevil, Rhynchophorus ferrugineus (Olivier) (Coleoptera: Curculionidae). In order to determine their role in external immunity, the immunosuppressive efficacy of the secretions in vitro against microbes, including bacteria and fungi, was clarified. In the present study, we found that these secretions had antimicrobial activity in vitro, implying external immunizing potency against pathogens. Surprisingly, all liquid phases of secretions could not significantly inhibit the growth of microbes in vitro compared to solid phases. To explain this phenomenon, the composition and emission differentia of secretions from the exocrine glands associated with different developmental stages, secretory regions, and phases were identified and analyzed based on metabonomics techniques. A total of more than 200 compounds, including quinines, phenols, aldehydes, acids, alcohols, saccharides, ketones, esters, amines, salts, ureas, and heterocycles, were identified in the secretions of larvae and adults. The liquid phase shared a number of metabolites with the solid phase, but the emission types and amounts were significantly different in the two phases, resulting in differences in external immunological activity. Tyrosine and p-benzoquinone were the dominant metabolites in all of the secretions, accounting for approximately 11.29% of emissions, with the portion in the solid phase being generally higher than that in the liquid phase. Moreover, only p-benzoquinone was entirely significantly upregulated in the solid phase compared to the liquid phase. Therefore, metabolome analysis suggested that p-benzoquinone, which may potentially be developed to be a valuable marker for determining external immunity, was considered to be the main substance responsible for external immune functions. This hypothesis was further demonstrated by the antimicrobial activity of p-benzoquinone.

Project description:As yet little is known of the bionomics of weevils of the genus Cleopomiarus Pierce, 1919; current knowledge is limited to data on the morphology and biology of the preimaginal stages of certain species. This paper includes original information on the life cycle of Cleopomiarusmicros (Germar, 1821). It presents the morphology of the egg, last larva (L3) and pupa. Data on the host plant (Jasionemontana L.) and breeding plant (Campanulapatula L.) and on the oviposition and phenology of the species are updated. The anatomy of the third-stage larva of C.micros shares certain traits with other species of the tribe Mecinini Gistel, 1848. Comparison of the morphology of preimaginal stages of C.micros with those previously described for other species of the genera Cleopomiarus and Miarus Schönherr, 1826 - previously considered the same genus - reveals species differences in larval body length, colour of the body and epicranium, and chaetotaxy of head and body.

Project description:Routine identification of bark and ambrosia beetles is done using morphology. For people lacking the necessary taxonomic knowledge, proper identification of a novel specimen can be challenging and time consuming. This study compares the usefulness of four genetic markers (28S, EF-1a, ITS2, and COI) and five primer pairs (D2F1/D3R2, eflafor1/eflarev1, ets149/efa754, ITS2F/ITS2R, and LCO1490/HCO2198) to identify Scolytinae beetles, and outlines a molecular identification strategy, with results possible in two days. Markers COI and EF-1a were selected based on the ability of the respective primers to amplify DNA from multiple genera (Coptoborus, Xyleborus, Hypothenemus, Theoborus, and Araptus) and the ability of the resulting sequences to provide accurate and unambiguous matches in GenBank. BLASTn analysis of EF-1a sequences (both primer pairs) correctly identified four out of the five genera and COI sequences identified at least one sample of every genus tested and was the only primer pair to correctly identify Araptus specimens. Further, 28S sequences successfully identified Coptoborus, Xyleborus, and Theoborus but not Hypothenemus or Araptus. The low number of EF-1a (1), 28S (7), and ITS2 (0) sequences from Araptus individuals present in GenBank compared with COI (137) is likely the reason that only the latter marker was capable of identifying members of this genus. ITS2 sequences were insufficient to identify any of the samples tested. This study also determined the minimum quantity of DNA that could be used for molecular identification. Primers D2F1 and D3R2, which had the highest rate of amplification in all genera tested, could yield an informative sequence with as little as 0.00048 ng of DNA, however, at least 0.0024 ng was needed for reliable amplification.

Project description:A detailed description of the mature larva and pupa of Rhinusaneta (Germar, 1821) and new diagnostic features of this species are presented. The development cycle of R.neta in the standard conditions lasts almost 60 days: an 11-day egg period, a 29-day larval period, and an 18-day pupal period, on average. The larvae are parasitised by hymenopterans of the superfamily Chalcidoidea. Similarities and differences with Rhinusabipustulata and other species of this genus are presented.

Project description:The mature larvae and pupae of Lixus (Ortholixus) bituberculatus Smreczy?ski, 1968 and Lixus (Dilixellus) neglectus Fremuth, 1983 (Curculionidae: Lixinae: Lixini) are described and compared with known larvae of 21 other Lixus and 2 Hypolixus taxa. The mature larva and pupa of Lixus bituberculatus are the first immature stages described representing the subgenus Ortholixus. The larva of Lixus neglectus, in the subgenus Dilixellus, is distinguished from the known larvae of four species in this subgenus by having more pigmented sclerites on the larval body. All descriptions of mature larvae from the tribe Lixini, as do all known species from the tribe Cleonini, fit the diagnosis of the mature larva of the Lixinae subfamily. Furthermore, new biological information of these species in the Czech Republic, Slovakia and Romania is provided. For Lixus bituberculatus, a chicory, Cichorium intybus L. (Asteraceae), is identified as a host plant, and Lixus neglectus is found on dock Rumex thyrsiflorus Fingerh. (Polygonaceae). Both species are probably monophagous or oligophagous. Adults of Lixus bituberculatus often inhabit host plants growing in active, dry and sunny pastures with sparse patches without vegetation, being mostly active during the night in April/May and then again in September, when the highest activity levels are observed. Adults of Lixus neglectus inhabit dry grasslands on sandy soils with host plants, being active during the day from May to September, with the highest level of activity in May/June and September. The larvae of both species are borers in the stem and root of the host plant, and they pupate in root or root neck. Adults leave the pupation cells at the end of summer and do not hibernate in the host plants. Finally, Romania is a new geographic record for Lixus bituberculatus.

Project description:Last instar larva and pupa of Metadonus vuillefroyanus (Capiomont, 1868) (Curculionidae: Hyperini) are described and compared with known larvae of the other 43 hyperine taxa. The thorn-like setae located on distinct black protuberances on the larval body are characteristic features of the genus Metadonus and the subgenus Eririnomorphus of the genus Hypera. The biological singularity of this species was studied and described. The variable colouration of larvae has been confirmed in association with the variability of the host plant's colouration at the studied localities. This species' reported inability to spin cocoons has been disproven. A different type of cocoon with two layers, where the inner layer consists of proteins from Malpighian tubules while the outer layer contains soil particles, is described. This type of cocoon is unique compared with those known from other hyperines, which usually pupate on or above the ground and do not use substrate particles in building their cocoons.

Project description:The ambrosia beetle Xyleborus volvulus Fabricius has been reported as a potential vector of the plant pathogen Raffaelea lauricola T.C. Harr., Fraedrich & Aghayeva that is affecting avocado orchards in South Florida. In this study, we examined its life cycle, process of gallery formation, gallery structure, and fungal associates by rearing one generation on avocado sawdust medium under control conditions. The adult foundress excavated a vertical tunnel that constituted the main gallery with a length of 2.5?cm, followed by the construction of up to six secondary galleries with a total length of 4.4?cm. The time period for one generation (egg to adult) was 28?days. Teneral males emerged 3 days after the emergence of the first females. The F1 generation did not significantly contribute to gallery expansion. Four species of Raffaelea and nine yeast species were recovered from galleries and beetles. Raffaelea arxii and Candida berthetii were the most frequent symbionts recovered from new adults and galleries. Candida berthetii dominated during the early stages of the gallery development, whereas R. arxii was most frequent in later stages. Other Raffaelea species were inconsistently isolated from galleries, which suggests a strong association between Xyleborus volvulus and both R. arxii and C. berthetii These results suggest that R. arxii is the primary nutritional symbiont of X. volvulus and that yeast species may be pioneer colonizers that assist with the growth of fungal symbionts.IMPORTANCE Ambrosia beetles cultivate fungi in tunnels bored into weakened host trees. This obligate interaction is required for their survival as beetles feed on these symbiotic fungi, and the fungi benefit from transportation by the beetles. Xyleborus volvulus carries many nonpathogenic symbionts; however, recently the acquisition of Raffaelea lauricola (the causal agent of a lethal vascular disease of lauraceous trees) by this beetle has altered its status from wood degrader to potential pest in avocado. We conducted a study to understand the relationship of this beetle and its fungal associates. Our results show that X. volvulus has a multipartite flexible association with different Raffaelea species. The lack of fidelity in the mutualistic association may explain the acquisition of R. lauricola Knowing the beetle biology and its mutualistic interactions furthers an understanding of the beetle's role as a potential vector and in disease transmission.

Project description:The oak flea weevil, Rhynchaenusmaculosus Yang et Zhang 1991, is a newly emerging pest that severely damages oak (genus Quercus) in China. The first R. maculosus outbreak occurred in 2020 and caused spectacular damage to all oak forests in Jilin province, northeast China. The lack of key morphological characters complicates the identification of this native pest, especially in larva and pupa stages. This is problematic because quick and accurate species identification is crucial for early monitoring and intervention during outbreaks. Here, we provided the first detailed morphological description of R. maculosus at four life stages. Additionally, we used DNA barcodes from larva and pupa specimens collected from three remote locations for molecular identification. The average pairwise divergence of all sequences in this study was 0.51%, well below the 2% to 3% (K-2-parameter) threshold set for one species. All sample sequences matched the R. maculosus morphospecies (KX657706.1 and KX657707.1), with 99.23% to 100% (sequence identity, E value: 0.00) matching success. The tree based on barcodes placed the specimens into the Rhynchaenus group, and the phylogenetic relationship between 62 sequences (30 samples and 32 from GeneBank) had high congruence with the morphospecies taxa. The traditional DNA barcodes were successfully transformed into quick response codes with larger coding capacity for information storage. The results showed that DNA barcoding is reliable for R. maculosus identification. The integration of molecular and morphology-based methods contributes to accurate species identification of this newly emerging oak pest.

Project description:Anthonomus grandis Boheman is a key pest in cotton crops in the New World. Its larval stage develops within the flower bud using it as food and as protection against its predators. This behavior limits the effectiveness of its control using conventional insecticide applications and biocontrol techniques. In spite of its importance, little is known about its genome sequence and, more important, its specific expression in key organs like the midgut. Total mRNA isolated from larval midguts was used for pyrosequencing. Sequence reads were assembled and annotated to generate a unigene data set. In total, 400,000 reads from A. grandis midgut with an average length of 237 bp were assembled and combined into 20,915 contigs. The assembled reads fell into 6,621 genes models. BlastX search using the NCBI-NR database showed that 3,006 unigenes had significant matches to known sequences. Gene Ontology (GO) mapping analysis evidenced that A. grandis is able to transcripts coding for proteins involved in catalytic processing of macromolecules that allows its adaptation to very different feeding source scenarios. Furthermore, transcripts encoding for proteins involved in detoxification mechanisms such as p450 genes, glutathione-S-transferase, and carboxylesterases are also expressed. This is the first report of a transcriptomic study in A. grandis and the largest set of sequence data reported for this species. These data are valuable resources to expand the knowledge of this insect group and could be used in the design of new control strategies based in molecular information.
