Identification of an H6N6 swine influenza virus in southern China.

ABSTRACT: This is the first report of avian-like H6N6 swine influenza virus from swine in southern China. Phylogenetic analysis indicated that this virus might originate from domestic ducks. Serological surveillance suggested there had been sporadic H6 swine influenza infections in this area. Continuing study is required to determine if this virus could be established in the swine population and pose potential threats to public health.

Project description:Here, we report the complete genome sequence of an H6N6 avian influenza virus (AIV) isolated from a pigeon in Guangxi, southern China, in 2014. The eight RNA segment genes shared a high nucleotide identity (97 to 99%) with H6N6 subtypes of AIV isolated from ducks in the regions around Guangxi Province. The finding of this study will help us understand the ecology and molecular characteristics of H6 avian influenza virus in wild birds in southern China.

Pathogenicity and transmission of a swine influenza A(H6N6) virus.

Project description:Subtype H6 influenza A viruses (IAVs) are commonly detected in wild birds and domestic poultry and can infect humans. In 2010, a H6N6 virus emerged in southern China, and since then, it has caused sporadic infections among swine. We show that this virus binds to α2,6-linked and α2,3-linked sialic acids. Mutations at residues 222 (alanine to valine) and 228 (glycine to serine) of the virus hemagglutinin (HA) affected its receptor-binding properties. Experiments showed that the virus has limited transmissibility between ferrets through direct contact or through inhalation of infectious aerosolized droplets. The internal genes of the influenza A(H1N1)pdm09 virus, which is prevalent in swine worldwide, increases the replication efficiency of H6N6 IAV in the lower respiratory tract of ferrets but not its transmissibility between ferrets. These findings suggest H6N6 swine IAV (SIV) currently poses a moderate risk to public health, but its evolution and spread should be closely monitored.

Identification of a Triple-Reassortant H1N1 Swine Influenza Virus in a Southern China Pig.

Project description:We report here the complete genome sequence of a triple-reassortant H1N1 swine influenza virus strain, A/swine/Guangxi/BB1/2013 (H1N1) (GXBB1), isolated from a swine in the Guangxi Province of southern China in 2013. We obtained the complete genome sequence of the GXBB1 virus. Sequence analysis demonstrated that this H1N1 virus was a triple-reassortant swine influenza virus (SIV) whose genes originated from avian, human, and swine, respectively. Knowledge regarding the complete genome sequence of the GXBB1 virus will be useful for epidemiological surveillance.

Isolation and Phylogenetic Analysis of H1N1 Swine Influenza Virus from Sick Pigs in Southern China.

Project description:Two swine influenza (SI) H1N1 virus was isolated from a pig during a severe outbreak of respiratory disease in south China. The two H1N1 influenza viruses were classical SI virus. A/swine/Guangdong/L6/09 is classical SI virus of recent years, which is of the main SI virus in China. Howere, A/swine/Guangdong/L3/09 was closet to A/swine/Iowa/1931, which was the first isolated SI virus and had demonstrated significant pathogenicity in animal models. The results of phylogenetic analysis of A/swine/Guangdong/L3/09 showed a close relationship with the 1918 pandemic virus. The results suggested that the previous SI virus appeared again. Whether, it brought a new pandemic to pigs deserves more attention.

Complete genome sequence of an avian-like H4N8 swine influenza virus discovered in southern China.

Project description:We report here the complete genomic sequence of an avian-like H4N8 swine influenza virus containing an H5N1 avian influenza virus segment from swine in southern China. Phylogenetic analyses of the sequences of all eight viral RNA segments demonstrated that these are wholly avian influenza viruses of the Asia lineage. To our knowledge, this is the first report of interspecies transmission of an avian H4N8 influenza virus to domestic pigs under natural conditions.

Identification of host encoded microRNAs interacting with novel swine-origin influenza A (H1N1) virus and swine influenza virus.

Project description:The discovery of microRNAs (miRNAs) is a remarkable breakthrough in the field of life science, and they are important actors which regulate gene expression in diverse cellular processes. Recently, several reports indicated that miRNAs can also target viruses and regulate virus replication. Here we discovered 36 pig-encoded miRNAs and 22 human-encoded miRNAs which have putative targets in swine influenza virus (SIV) and Swine-Origin 2009 A/H1N1 influenza virus (S-OIV) genes respectively. Interestingly, the putative interactions of ssc-miR-124a, ssc-miR-136 and ssc-miR-145 with their SIV target genes had been found to be maintained almost throughout all of the virus evolution. Enrichment analysis of previously reported miRNA gene expression profiles revealed that three miRNAs are expressed at higher levels in human lung or trachea tissue. The hsa-miR-145 and hsa-miR-92a putatively target the HA gene and hsa-miR-150 putatively targets the PB2 gene. Analysis results based on the location distribution from which virus was isolated and sequence conservation imply that some putative miRNA-mediated host-virus interactions may characterize the location-specificity.

Novel swine influenza virus reassortants in pigs, China.

Project description:During swine influenza virus surveillance in pigs in China during 2006-2009, we isolated subtypes H1N1, H1N2, and H3N2 and found novel reassortment between contemporary swine and avian panzootic viruses. These reassortment events raise concern about generation of novel viruses in pigs, which could have pandemic potential.

Project description:Here, we report the complete genome sequence of an H6N6 avian influenza virus (AIV) isolated from Sansui Sheldrake ducks in Guizhou Province, China, in 2014. Phylogenetic analysis showed that the H6N6 virus was a reassortant virus derived from three different H6 subtype lineages. The finding of this study will help us understand the epidemiology and the evolutionary characteristics of H6 subtypes of AIV in ducks in southwestern China.
