Trans-3,3',4,5'-Tetra-meth-oxy-stilbene.

ABSTRACT: The title compound, C(18)H(20)O(4), was synthesized by a Wittig-Horner reaction of diethyl 3,4-dimeth-oxy-benzyl-phosphate and 3,5-dimeth-oxy-benzaldehyde. In the crystal, the dihedral angle between the two aromatic rings is 2.47 (12)°. All the meth-oxy groups are almost coplanar with the aromatic ring to which they are attached [C-C-O-C torsion angles = -2.8 (3), -5.2 (4), -176.3 (2) and -178.0 (2)°].

Project description:In the crystal structure, the title compound, C(24)H(22)O(8), lies on a twofold rotation axis and the asymmetric unit comprises one half-mol-ecule. The dihedral angle formed by the coumarin unit with the symmetry-related part is 74.78 (14)°. One of the meth-oxy groups attached to the coumarin unit is considerably twisted, making an angle of 87.17 (17)° with respect to the coumarin unit; the other is twisted by 0.66 (19)°. No classical hydrogen bonds are found in the sturcture; only a weak C-H⋯π inter-action and short intra-molecular O⋯O contacts [2.683 (2)-2.701 (2) Å] are observed.

Project description:Blood-brain barrier (BBB) disruption is a major pathophysiological event of ischemic stroke. Brain microvascular endothelial cells are critical to maintain homeostasis between central nervous system and periphery. Resveratrol protects against ischemic stroke. 3,3',4,5'-tetramethoxy-trans-stilbene (3,3',4,5'-TMS) and 3,4',5-trimethoxy-trans-stilbene (3,4',5-TMS) are resveratrol derivatives with addition of methoxy groups, showing better pharmacokinetic performance. We aimed to explore their protective effects and underlying mechanisms. Oxygen-glucose deprivation (OGD) model was applied in bEnd.3 cell line, mouse brain microvascular endothelium to mimic ischemia. The cells were pre-treated with 3,3',4,5'-TMS or 3,4',5-TMS (1 and 5 μM, 24 h) and then subjected to 2-h OGD injury. Cell viability, levels of proinflammatory cytokines and reactive oxygen species (ROS), and protein expressions were measured by molecular assays and fluorescence staining. OGD injury triggered cell death, inflammatory responses, ROS production and nuclear factor-kappa B (NF-κB) signalling pathway. These impairments were remarkably attenuated by the two stilbenes, 3,3',4,5'-TMS and 3,4',5-TMS. They also alleviated endothelial barrier injuries through upregulating the expression of tight junction proteins. Moreover, 3,3',4,5'-TMS and 3,4',5-TMS activated 5' adenosine monophosphate-activated protein kinase (AMPK) and endothelial nitric oxide synthase (eNOS). Overall, 3,3',4,5'-TMS and 3,4',5-TMS exert protective effects against OGD damage through suppressing cell death, inflammatory responses, oxidative stress, as well as BBB disruption on bEnd.3 cells.

Project description:The asymmetric unit of the title compound, C(36)H(30)O(4), contains two crystallographically independent mol-ecules of similar geometry. In both mol-ecules, the meth-oxy-meth-oxy groups are disordered over two positions with refined site occupancies of 0.613 (3):0.387 (3) and 0.589 (4):0.411 (4). The dihedral angles between the naphthalene planes within the same mol-ecule are 71.72 (7) and 71.73 (8)°. In the crystal, neighbouring mol-ecules are linked by inter-molecular C-H⋯O hydrogen bonds, forming double chains parallel to the c axis.

While the monohydrate is mainly planar, in the title compound there is a slight twist in the two benzene rings at each end of the complex [dihedral angle = 13.14 (6)°]. All the atoms of the meth-oxy substitutents are in the plane of the ring to which they are attached (r.m.s. deviation = 0.0079 Å) except for one of the meth-oxy C atoms, which deviates slightly [0.309 (4) Å]. In the crystal, weak C-H⋯O inter-molecular inter-actions link the mol-ecules.

Project description:IN THE TITLE COMPOUND [SYSTEMATIC NAME: 1-(2,4-dimethoxyphenyl)-3-(3,4-dimethoxyphenyl)prop-2-en-1-one], C(19)H(20)O(5), the dihedral angle between the benzene rings is 26.88 (5)°. One of the meth-oxy groups is twisted slightly away from the plane [C-O-C-C torsion angle = -12.8 (3)°] while the others are almost co-planer [C-O-C-C torsion angles = -3.2 (3), 2.6 (3) and -3.6 (3)°]. The crystal packing is stabilized by inter-molecular C-H⋯O inter-actions. A weak intra-molecular C-H⋯O inter-action occurs.
