ABSTRACT: 4-Phospho-D-erythronate is an intermediate in synthesis of pyridoxal 5'-phosphate in some bacteria and an inhibitor of ribose 5-phosphate isomerase. Previous synthetic schemes for the preparation of 4-phospho-D-erythronate required expensive precursors and typically gave low yields. We report a straightforward synthesis of 4-phospho-D-erythronate from the inexpensive precursor D-erythronolactone in 5 steps with a preparatively useful yield of 22%.

An Amazingly Simple, Fast and Green Synthesis Route to Polyaniline Nanofibers for Efficient Energy Storage.

Project description:The major drawbacks of the conventional methods for preparing polyaniline (PANI) are the large consumptions of toxic chemicals and long process durations. This paper presents a remarkably simple and green route for the chemical oxidative synthesis of PANI nanofibers, utilizing sodium phytate as a novel and environmentally friendly plant derived dopant. The process shows a remarkable reduction in the synthesis time and usage of toxic chemicals with good dispersibility and exceedingly high conductivity up to 10 S cm-1 of the resulting PANI at the same time. A detailed characterization of the PANI samples has been made showing excellent relationships between their structure and properties. Particularly, the electrochemical properties of the synthesized PANI as electrode material for supercapacitors were analyzed. The PANI sample, synthesized at pre-optimized conditions, exhibited impressive supercapacitor performance having a high specific capacitance (Csp) (832.5 Fg-1 and 528 Fg-1 at 1 Ag-1 and 40 Ag-1, respectively) as calculated from galvanostatic charge/discharge (GCD) curves. A good rate capability with a capacitance retention of 67.6% of its initial value was observed. The quite low solution resistance (Rs) value of 281.0 × 10-3 Ohm and charge transfer resistance value (Rct) of 7.44 Ohm represents the excellence of the material. Further, a retention of 95.3% in coulombic efficiency after 1000 charge discharge cycles, without showing any significant degradation of the material, was also exhibited.

A simple route to highly active single-enzyme nanogels.

Project description:We have established a simple one-step synthesis of single-enzyme nanogels (SENs), i.e., nanobiocatalysts consisting of an enzyme molecule embedded in a hydrophilic, polymeric crosslinked nanostructure, as a most attractive approach to enhance the stability of enzymes. In contrast to earlier protocols, we demonstrate here that the addition of a small amount of sucrose makes the nanogel formation equally effective as earlier two-step protocols requiring enzyme pre-modification. This provides the dual advantage of skipping a synthetic step and preserving the surface chemistry of the enzymes, hence their native structure. Enzymes encapsulated in this way exhibit a high catalytic activity, similar to that of the free enzymes, in a markedly widened pH range. With our method, the thickness of the hydrogel layer can be finely tuned by careful adjustment of reaction parameters. This is most important because the shell thickness strongly affects both enzyme activity and stability, as we observe for a wide selection of proteins. Finally, a single-molecule analysis by means of two-color confocal fluorescence coincidence analysis confirms that our encapsulation method is highly efficient and suppresses the occurrence of nanoparticles lacking an enzyme molecule. The proposed method is therefore highly attractive for biocatalysis applications, ensuring a high activity and stability of the enzymes.

Metazoan opsin evolution reveals a simple route to animal vision.

Project description:All known visual pigments in Neuralia (Cnidaria, Ctenophora, and Bilateria) are composed of an opsin (a seven-transmembrane G protein-coupled receptor), and a light-sensitive chromophore, generally retinal. Accordingly, opsins play a key role in vision. There is no agreement on the relationships of the neuralian opsin subfamilies, and clarifying their phylogeny is key to elucidating the origin of this protein family and of vision. We used improved methods and data to resolve the opsin phylogeny and explain the evolution of animal vision. We found that the Placozoa have opsins, and that the opsins share a common ancestor with the melatonin receptors. Further to this, we found that all known neuralian opsins can be classified into the same three subfamilies into which the bilaterian opsins are classified: the ciliary (C), rhabdomeric (R), and go-coupled plus retinochrome, retinal G protein-coupled receptor (Go/RGR) opsins. Our results entail a simple scenario of opsin evolution. The first opsin originated from the duplication of the common ancestor of the melatonin and opsin genes in a eumetazoan (Placozoa plus Neuralia) ancestor, and an inference of its amino acid sequence suggests that this protein might not have been light-sensitive. Two more gene duplications in the ancestral neuralian lineage resulted in the origin of the R, C, and Go/RGR opsins. Accordingly, the first animal with at least a C, an R, and a Go/RGR opsin was a neuralian progenitor.

Simple, Axial Ligand-Mediated Route to Water-Soluble Iridium Corroles.

Project description:The synthesis and purification of water-soluble porphyrin-type compounds for photodynamic therapy and other medical applications is often a tedious exercise. Here, we have investigated the simple stratagem of adding a water-soluble axial ligand to the standard protocol for iridium insertion into simple meso-triarylcorroles. Early results showed that six-coordinate Ir[TpXPC](dna)2 derivatives, in which TpXPC = tris(para-X-phenyl)corrole (X = CF3, CN, H, and OMe) and dna = dinicotinic acid, are highly water-soluble. In the end, however, all axially nitrogen-ligated complexes proved unstable with respect to chromatographic purification and storage. Five-coordinate water-soluble phosphine adducts, fortunately, proved a great improvement. From the point of view of ease of purification and storage, the best products proved to be Ir[TpXPC](L), where X = CF3 and OMe and L = tris(2-carboxyethyl)phosphine (tcep) and trisodium tris(3-sulfonatophenyl)phosphine (tppts); carefully optimized synthetic protocols are presented for these four compounds.

Engineered mosaic protein polymers; a simple route to multifunctional biomaterials.

Project description:BackgroundEngineered living materials (ELMs) are an exciting new frontier, where living organisms create highly functional materials. In particular, protein ELMs have the advantage that their properties can be manipulated via simple molecular biology. Caf1 is a protein ELM that is especially attractive as a biomaterial on account of its unique combination of properties: bacterial cells export it as a massive, modular, non-covalent polymer which is resistant to thermal and chemical degradation and free from animal material. Moreover, it is biologically inert, allowing the bioactivity of each 15 kDa monomeric Caf1 subunit to be specifically engineered by mutagenesis and co-expressed in the same Escherichia coli cell to produce a mixture of bioactive Caf1 subunits.ResultsHere, we show by gel electrophoresis and transmission electron microscopy that the bacterial cells combine these subunits into true mosaic heteropolymers. By combining two separate bioactive motifs in a single mosaic polymer we demonstrate its utility by stimulating the early stages of bone formation by primary human bone marrow stromal cells. Finally, using a synthetic biology approach, we engineer a mosaic of three components, demonstrating that Caf1 complexity depends solely upon the variety of monomers available.ConclusionsThese results demonstrate the utility of engineered Caf1 mosaic polymers as a simple route towards the production of multifunctional biomaterials that will be useful in biomedical applications such as 3D tissue culture and wound healing. Additionally, in situ Caf1 producing cells could create complex bacterial communities for biotechnology.Graphical abstract

A Simple and Efficient Mechanochemical Route for the Synthesis of Salophen Ligands and of the Corresponding Zn, Ni, and Pd Complexes.

Project description:A number of salophen ligands and their Zn, Ni, and Pd complexes were synthesized by an efficient one-pot mechanosynthesis protocol. The reaction products were characterized by means of complementary solid-state techniques, i.e., powder X-ray diffraction, single-crystal X-ray diffraction, and solid-state NMR spectroscopy. Four new crystal structures of metal salophen complexes as DMSO solvates are here reported. The described simple and relatively fast (about 1 h for all derivatives) procedure is a good alternative to classical methods performed in organic solvents.

Simple Route to Synthesize Fully Conjugated Ladder Isomer Copolymers with Carbazole Units.

Project description:Two isomer polymers, P3 and P6, with fully conjugated ladder structures are presented by simple synthetic routes. The well-defined structures of fully conjugated ladder polymers P3 and P6 were ensured by the high yields of every reaction step. The fully rigid ladder structures were confirmed by nuclear magnetic resonance (NMR), fourier transform infrared spectroscopy (FTIR), and photophysical test. Polymers P3 and P6 with bulky alkyl side chains exhibit good solution processability and desirable thermostable properties. After the intramolecular cyclization reaction, the band gaps of polymers P3 and P6 become lower (2.86 eV and 2.66 eV, respectively) compared with polymers P1 and P4. This initial study provides insight for the rational design of fully ladder-conjugated isomeric polymers with well-defined structures.

Synthesis of Low-Cost and High-Performance Dual-Atom Doped Carbon-Based Materials with a Simple Green Route as Anodes for Sodium-Ion Batteries.

Project description:Sodium-ion batteries (SIBs) are promising alternatives to replace lithium-ion batteries as future energy storage batteries because of their abundant sodium resources, low cost, and high charging efficiency. In order to match the high energy capacity and density, designing an atomically doped carbonous material as the anode is presently one of the important strategies to commercialize SIBs. In this work, we report the preparation of high-performance dual-atom-doped carbon (C) materials using low-cost corn starch and thiourea (CH4N2S) as the precursors. The electronegativity and radii of the doped atoms and C are different, which can vary the embedding properties of sodium ions (Na+) into/on C. As sulfur (S) can effectively expand the layer spacing, it provides more channels for embedding and de-embedding Na+. The synergistic effect of N and S co-doping can remarkably boost the performance of SIBs. The capacity is preserved at 400 mAh g -1 after 200 cycles at 500 mA g-1; more notably, the initial Coulombic efficiency is 81%. Even at a high rate of high current of 10 A g-1, the cell capacity can still reach 170 mAh g-1. More importantly, after 3000 cycles at 1 A g-1, the capacity decay is less than 0.003% per cycle, which demonstrates its excellent electrochemical performance. These results indicate that high-performance carbon materials can be prepared using low-cost corn starch and thiourea.
