Project description:It is found that hydrogelation of peptides enhances the transverse relaxation rate R2 of water protons but has no effect on the longitudinal relaxation rate R1 and the diffusion coefficient D. The magnitude of water proton R2 enhancement increases linearly with the shear modulus G of hydrogels.

Project description:A pair of mutually attractive but self-repulsive decapeptides, with alternating charged/neutral amino acid sequence patterns, was found to co-assemble into a viscoelastic material upon mixing at a low total peptide concentration of 0.25 wt %. Circular dichroism spectroscopy of individual decapeptide solutions revealed their random coil conformation. Transmission electron microscopy images showed the nanofibrillar network structure of the hydrogel. Dynamic rheological characterization revealed its high elasticity and shear-thinning nature. Furthermore, the co-assembled hydrogel was capable of rapid recoveries from repeated shear-induced breakdowns, a property desirable for designing injectable biomaterials for controlled drug delivery and tissue engineering applications. A systematic variation of the neutral amino acids in the sequence revealed some of the design principles for this class of biomaterials. First, viscoelastic properties of the hydrogels can be tuned through adjusting the hydrophobicity of the neutral amino acids. Second, the beta-sheet propensity of the neutral amino acid residue in the peptides is critical for hydrogelation.

Project description:The data presented here are related to the research paper entitled "Green Earth pigments dispersions: water dynamics at the interfaces". The nuclear magnetic resonance (NMR) relaxometry data are provided for various aqueous Green Earth (GE) pigments dispersions with volume fraction spanning approximately from 0.1 to 0.5. For two of them (Cyprus GE and Bohemian GE), the NMR relaxation profiles from 10 kHz to 30 MHz (1H frequency) is given for several temperatures spanning from 293 to 318K. In addition, the X-ray diffraction pattern is provided for France GE (Kremer pigments) for the identification of the main mineral component. The nitrogen gas isotherms are provided for Cyprus GE and Bohemian GE.

Project description:In the cytosolic environment, protein crowding and Brownian motions result in numerous transient encounters. Each such encounter event increases the apparent size of the interacting molecules, leading to slower rotational tumbling. The extent of transient protein complexes formed in live cells can conveniently be quantified by an apparent viscosity, based on NMR-detected spin-relaxation measurements, that is, the longitudinal (T1) and transverse (T2) relaxation. From combined analysis of three different proteins and surface mutations thereof, we find that T2 implies significantly higher apparent viscosity than T1. At first sight, the effect on T1 and T2 seems thus nonunifiable, consistent with previous reports on other proteins. We show here that the T1 and T2 deviation is actually not a inconsistency but an expected feature of a system with fast exchange between free monomers and transient complexes. In this case, the deviation is basically reconciled by a model with fast exchange between the free-tumbling reporter protein and a transient complex with a uniform 143 kDa partner. The analysis is then taken one step further by accounting for the fact that the cytosolic content is by no means uniform but comprises a wide range of molecular sizes. Integrating over the complete size distribution of the cytosolic interaction ensemble enables us to predict both T1 and T2 from a single binding model. The result yields a bound population for each protein variant and provides a quantification of the transient interactions. We finally extend the approach to obtain a correction term for the shape of a database-derived mass distribution of the interactome in the mammalian cytosol, in good accord with the existing data of the cellular composition.

Project description:Unlike the constant nature of elastic coefficients of isotropic bulk materials, the Young's (E) and shear (μ) moduli of nano-structured (NS) gyroid metamaterials change with relative density (ρ), but at different rates depending on the cell size of the structure. These elastic behaviors displayed by E and μ cause crossover/inversion of these two moduli, such that μ of the NS gyroid metamaterials is greater than E for the structures with ρ < 0.23. This peculiar elastic behavior causes NS gyroid metamaterials to display high μ/E values (~1.0), which are more than 250% larger than the typical values of the bulk material (~0.38), indicating that the NS gyroid metamaterial, even if it is light, is resistant to shear deformation. Here, we report the results of molecular dynamics simulations performed to elucidate the reason for unusually high μ/E values in NS gyroid metamaterials.

Project description:The kinetically stable heptadentate gadolinium complex Gd.pDO3A (1.Gd) demonstrates significant 31P nuclear magnetic resonance (NMR) relaxation enhancement of biologically relevant phosphate species; adenosine triphosphate (ATP), phosphocreatine (PCr) and inorganic phosphate. Gd.pDO3A (1.Gd) binds these species in fast exchange, enabling the relaxation of the bulk phosphate species in solution. This gives rise to 31P relaxation enhancements up to 250-fold higher than those observed for 31P relaxation enhancements with the commercial MRI contrast agent Gd.DOTA (DOTAREM), 2. Gd.pDO3A-like complexes may have potential applications as 31P magnetic resonance contrast agents, since shortening the T1 relaxation time of phosphate species would reduce the time needed to acquire 31P-MR spectra.

Project description:Protein side chain dynamics play a vital role in many biological processes, but differentiating mobile from rigid side chains remains a technical challenge in structural biology. Solution NMR spectroscopy is ideally suited for this but suffers from limited signal-to-noise, signal overlap, and a need for fractional 13C or 2H labeling. Here we introduce a simple strategy measuring initial 1H relaxation rates during a 1H TOCSY sequence like DIPSI-2, which can be appended to the beginning of any multi-dimensional NMR sequence that begins on 1H. The TOCSY RF field compels all 1H atoms to behave similarly under the influence of strong coupling and rotating frame cross-relaxation, so that differences in relaxation rates are due primarily to side chain mobility. We apply the scheme to a thermostable mutant Pin1 WW domain and demonstrate that the observed 1H relaxation rates correlate well with two independent NMR measures of side-chain dynamics, cross-correlated 13C relaxation rates in 13CβH2 methylene groups and maximum observable 3J couplings sensitive to the χ1 side chain dihedral angle (3JHα,Hβ, 3JN,Hβ, and 3JCO,Hβ). The most restricted side chains belong to Trp26 and Asn40, which are closely packed to constitute the folding center of the WW domain. None of the other conserved aromatic residues is as immobile as the first tryptophan side chain of the WW domain. The proposed 1H relaxation methodology should make it relatively easy to measure side chain dynamics on uniformly 15N- or 13C-labeled proteins, so long as chemical shift assignments are obtainable.

Project description:We propose an integrated method combining low-frequency mechanics with optical imaging to map the shear modulus within the biological tissue. Induced shear wave propagating in tissue is tracked in space and time using phase-sensitive optical coherence tomography (PhS-OCT). Local estimates of the shear-wave speed obtained from tracking results can image the local shear modulus. A PhS-OCT system remotely records depth-resolved, dynamic mechanical waves at an equivalent frame rate of ?47??kHz with the high spatial resolution. The proposed method was validated by examining tissue-mimicking phantoms made of agar and light scattering material. Results demonstrate that the shear wave imaging can accurately map the elastic moduli of these phantoms.

Project description:In a description of materials for orthotropic panels with a soft and/or corrugated core, it is important to correctly determine all constitutive parameters. In laboratory practice, the determination of transverse shear modulus is often overlooked. This paper presents a method for determining this property based on a plate torsion test and a correctly formulated analytical description. It has been proved that the transverse shear effect in some cases cannot be omitted because it significantly influences the mechanical behavior of corrugated board. The method of transverse shear modeling used so far can be modified to eliminate dimensionless, physically unjustified coefficient and replace them with coefficients that have a physical basis. It is shown here that such modification leads to results with lower error. The effective modeling of transverse shear effects enables a more conscious design of corrugated board structures, where the final goal is to obtain packaging with high strength and durability but low material consumption.

Project description:Dynamic elastography is an attractive method to evaluate tissue biomechanical properties. Recently, it was extended from US- and MR-based modalities to optical ones, such as optical coherence tomography for three-dimensional (3-D) imaging of propagating mechanical waves in subsurface regions of soft tissues, such as the eye. The measured group velocity is often used to convert wave speed maps into 3-D images of the elastic modulus distribution based on the assumption of bulk shear waves. However, the specific geometry of OCE measurements in bounded materials such as the cornea and skin calls into question elasticity reconstruction assuming a simple relationship between group velocity and shear modulus. We show that in layered media the bulk shear wave assumption results in highly underestimated shear modulus reconstructions and significant structural artifacts in modulus images. We urge the OCE community to be careful in using the group velocity to evaluate tissue elasticity and to focus on developing robust reconstruction methods to accurately reconstruct images of the shear elastic modulus in bounded media.
