New species of springtails in the Proisotoma genus complex from Vermont and New York, USA with descriptive notes on Ballistura alpa Christiansen & Bellinger 1980 (Hexapoda, Collembola, Isotomidae).

ABSTRACT: Three new species of Isotomidae springtails are described from the Lake Champlain Basin (Vermont and New York, USA), Lake Willoughby and Greater Averril Pond in Vermont. Subisotoma joyceisp. n. and Scutisotoma champisp. n. were collected in sandy beaches whereas Ballistura rossisp. n. was found only in a constructed wetland built and managed by the University of Vermont. Scutisotoma champisp. n. was found in Lakes Champlain and Willoughby, and Greater Averril Pond and is probably present in most lakes and large ponds in the area. Subisotoma joyceisp. n. was found only along the southern and eastern coast of South Hero, and the mainland coast facing eastern South Hero. Ballistura alpa is redescribed and transferred to the genus Pachyotoma based on the absence of tibiotarsal seta B4/B5, the presence of secondary cuticular granules, 4 prelabral setae, a full complement of guard setae on labial papilla E and in having a bifurcate outer maxillary lobe with 4 sublobal setae.

Project description:BackgroundSpringtails (Hexapoda: Collembola) are tiny organisms that lead a hidden life, mostly occuring deep in the soil and on leaf litter. They have a variety of interesting body morphology patterns, the most famous of which is the catapult-like structure that enables them to jump and flee from predators. This highly specialized jumping apparatus consists of a mobile furca, which when at rest fits into a trigger, "the retinaculum" on the ventral side of the abdomen. Despite the many studies that have attempted to investigate the jumping apparatus, the actual mechanisms involved in the jump, for example the way in which the furca is released by the retinaculum, how and where the mechanisms of spring and hydrostatic pressure originate, are still not properly understood. The morphology of the jumping apparatus of Orchesella cincta was investigated in detail using confocal laser scanning microscopy and MicroCT techniques for 3D reconstruction.ResultsThe morphology of O. cincta with both flexed and extended furca is analysed and described. The abdominal musculature involved in the jumping mechanism and relevant structures of the exoskeleton of retinaculum and furca are described in detail. With the data obtained in this study, hypotheses can be made about (1) where and how the spring and hydrostatic pressure mechanisms originate; (2) which muscles act on the extension and flexion of the furca; (3) which muscles act on the retinaculum and (4) how the retinaculum is released from the furca.ConclusionsThe comparative morphological study proved informative, and shows how springtail jumping involves mechanisms unique to this taxon. Hydrostatic pressure regulation possibly varies between animals with distinct segmentation, and those with fused segmentation. Interesting cuticular characters were revealed, such as basal plates and sclerites related to the construction of the spring mechanism. The present study establishes itself as a model option for future morphofunctional studies on springtail's jumping. Analysis of videos and images using a high speed camera will be useful for understanding how the jump develops through take-off, aerial and landing phases.

Project description:BackgroundMitogenomics data, i.e. complete mitochondrial genome sequences, are popular molecular markers used for phylogenetic, phylogeographic and ecological studies in different animal lineages. Their comparative analysis has been used to shed light on the evolutionary history of given taxa and on the molecular processes that regulate the evolution of the mitochondrial genome. A considerable literature is available in the fields of invertebrate biochemical and ecophysiological adaptation to extreme environmental conditions, exemplified by those of the Antarctic. Nevertheless, limited molecular data are available from terrestrial Antarctic species, and this study represents the first attempt towards the description of a mitochondrial genome from one of the most widespread and common collembolan species of Antarctica.ResultsIn this study we describe the mitochondrial genome of the Antarctic collembolan Cryptopygus antarcticus Willem, 1901. The genome contains the standard set of 37 genes usually present in animal mtDNAs and a large non-coding fragment putatively corresponding to the region (A+T-rich) responsible for the control of replication and transcription. All genes are arranged in the gene order typical of Pancrustacea. Three additional short non-coding regions are present at gene junctions. Two of these are located in positions of abrupt shift of the coding polarity of genes oriented on opposite strands suggesting a role in the attenuation of the polycistronic mRNA transcription(s). In addition, remnants of an additional copy of trnL(uag) are present between trnS(uga) and nad1. Nucleotide composition is biased towards a high A% and T% (A+T = 70.9%), as typically found in hexapod mtDNAs. There is also a significant strand asymmetry, with the J-strand being more abundant in A and C. Within the A+T-rich region, some short sequence fragments appear to be similar (in position and primary sequence) to those involved in the origin of the N-strand replication of the Drosophila mtDNA.ConclusionThe mitochondrial genome of C. antarcticus shares several features with other pancrustacean genomes, although the presence of unusual non-coding regions is also suggestive of molecular rearrangements that probably occurred before the differentiation of major collembolan families. Closer examination of gene boundaries also confirms previous observations on the presence of unusual start and stop codons, and suggests a role for tRNA secondary structures as potential cleavage signals involved in the maturation of the primary transcript. Sequences potentially involved in the regulation of replication/transcription are present both in the A+T-rich region and in other areas of the genome. Their position is similar to that observed in a limited number of insect species, suggesting unique replication/transcription mechanisms for basal and derived hexapod lineages. This initial description and characterization of the mitochondrial genome of C. antarcticus will constitute the essential foundation prerequisite for investigations of the evolutionary history of one of the most speciose collembolan genera present in Antarctica and other localities of the Southern Hemisphere.
