New and little known Isotomidae (Collembola) from the shore of Lake Baikal and saline lakes of continental Asia.

ABSTRACT: Collembola of the family Isotomidae from the shores of Lake Baikal and from six saline lake catenas of the Buryat Republic (Russia) and Inner Mongolia Province (China) were studied. Pseudanurophorus barathrum Potapov & Gulgenova, sp. nov. and Parisotoma baicalica Potapov & Gulgenova, sp. nov. from Baikal and Ephemerotoma buryatica Potapov, Huang & Gulgenova, sp. nov. and Folsomia mongolica Huang & Potapov, sp. nov. from saline lakes are described here. A morphological description of epitokous males of Scutisotoma acorrelata Potapov, Babenko & Fjellberg, 2006 is given. A list of 23 species of the family Isotomidae found in the shores of studied lakes is provided based on literature sources and newly collected material.

Project description:Three new species, Hypogastrura variata sp. nov., Xenylla aculeata sp. nov., and X. arnei sp. nov., are described based on material from coastal communities of the Sea of Okhotsk, northeast Russia. Taxonomic remarks concerning a little-known species, H. yosii Stach, 1964, found in coastal wrack sediments on Kunashir Island, Kuriles are also given. COI sequences of the above species are analysed, thus allowing for their species statuses to be confirmed.

Bellisotoma, a new genus of Isotomidae from North America (Hexapoda, Collembola).

Project description:A new genus of Isotomidae, Bellisotoma gen. n., is described. The new genus is a member of the Proisotoma genus complex and is characterized by a combination of having a bidentate mucro with wide dorsal lamellae that join clearly before the end of mucronal axis without forming a tooth and one strong ventral rib with basal notch that articulates with dens; having abundant chaetotaxy on both faces of dens; and abundant tergal sensilla. Bellisotoma gen. n. shows a furcula adapted to a neustonic mode of life, and may be a Isotopenola-like derivative adapted to neustonic habitats. Subisotoma joycei Soto-Adames & Giordano, 2011 and Ballistura ewingi James, 1933 are transferred to the new genus.

New species of Isotomiella Bagnall, 1939 from Southeast of Brazil (Collembola, Isotomidae).

Project description:Two new species of the genus Isotomiella Bagnall, 1939 are described and illustrated, the first: Isotomiella macedoisp. n., based on males and females, from the "Parque Nacional da Serra dos Órgãos" (Teresópolis municipality, State of Rio de Janeiro) differs from the other by tibiotarsus III thickened and blunt and two antero-lateral chaetae of labrum strongly thickened. The second species Isotomiella uaisp. n. from "Serra da Gandarela", (Caeté municipality, State of Minas Gerais) differs from the other by presence of short sensilla on antennal IV and tergites, two anterolabral chaetae thickened and falcate mucro.

Generic switch-over during ontogenesis in Dimorphacanthella gen. n. (Collembola, Isotomidae) with barcoding evidence.

Project description:A new genus Dimorphacanthella is established for Tetracanthella anommatos Chen and Yin, 1984 and Dimorphacanthella mediasetasp. n. from China. The new genus exhibits an unusual metamorphosis: small juveniles, previously called Uzelia anommatos Yue & Yin, 1999 get the second pair of anal spines resulted from moulting and become "Tetracanthella". Species identity of forms with two and four anal spines is proved by barcoding analysis. The derivation of anal spines is compared among genera having four anal spines.

Taxonomy of the Cryptopygus complex. I. Pauropygus - a new worldwide littoral genus (Collembola, Isotomidae).

Project description:In this paper, we describe the new genus Pauropygus gen. n. which includes three minute species, blind and unpigmented, living in interstitial littoral habitats in tropical or subtropical countries. Two of these species are new to science (type species Pauropygus projectus sp. n. from New Caledonia and Pauropygus pacificus sp. n. from China); the third one, originally described in the genus Cryptopygus (Cryptopygus caussaneli Thibaud, 1996), has a larger pantropical distribution. We synonymize here Cryptopygus riebi Barra, 1997 from South Africa with Pauropygus caussaneli. Two paratypes of the Mexican species Cryptopygus axayacatl Palacios & Thibaud, 2001 turned also to be Pauropygus caussaneli, while the holotype and remaining paratypes of this species support its placement in Proisotomodes. Among the Cryptopygus complex, Pauropygus gen. n. is easily recognized by characters of mouthparts (presence of two large projections on pleural fold, basolateral field with 6 chaetae, modified mouthparts) and reduced sensillar chaetotaxy (tergal sensilla 2-3,0-1/0-1,0-1,1-2,1-2,1-3, microsensilla reduced in number: 00/0-100, with sensilla situated in p-row on the abdomen). Small size, absence of eyes and pigment are also shared by all its species. The three species belonging to the genus differ by sensillar chaetotaxy.

Cryptic diversity in the ubiquist species Parisotoma notabilis (Collembola, Isotomidae): a long-used chimeric species?

Project description:Parisotoma notabilis is the most common species of Collembola in Europe and is currently designated as ubiquist. This species has been extensively used in numerous studies and is considered as well characterized on a morphological ground. Despite the homogeneity of its morphology, the sequencing of the barcoding fragment (5' end of COI) for several populations throughout Europe and North America revealed four distinct genetic lineages. The divergence found between these lineages was similar to the genetic distance among other species of the genus Parisotoma included in the analysis. All four lineages have been confirmed by the nuclear gene 28S. This congruence between mitochondrial and nuclear signals, as well as the geographical distribution pattern of lineages observed in Europe, supports the potential specific status of these lineages. Based on specimens from the type locality (Hamburg), the species name was successfully assigned to one of these lineages. This finding raises several problems as Parisotoma notabilis has been widely used in many ecological studies. Accumulation of new data for the different lineages detected, especially ecological information and life history traits, is needed to help resolve this situation.

Bacterioneuston in Lake Baikal: Abundance, Spatial and Temporal Distribution.

Project description:An aquatic surface microlayer covers more than 70% of the world's surface. Our knowledge about the biology of the surface microlayer of Lake Baikal, the most ancient lake on Earth with a surface area of 31,500 km², is still scarce. The total bacterial abundance, the number of cultured heterotrophic temporal bacteria, and the spatial distribution of bacteria in the surface microlayer and underlying waters of Lake Baikal were studied. For the first time, the chemical composition of the surface microlayer of Lake Baikal was determined. There were significant differences and a direct relationship between the total bacterial abundance in the surface microlayer and underlying waters of Lake Baikal, as well as between the number of cultured heterotrophic bacteria in studied water layers in the period of summer stratification. In the surface microlayer, the share of cultured heterotrophic bacteria was higher than in the underlying waters. The surface microlayer was characterized by enrichment with PO?3-, total organic carbon and suspended particulate matter compared to underlying waters. A direct relationship was found between the number of bacteria in the surface microlayer and environmental factors, including temperature, total organic carbon and suspended particulate matter concentration.

Pseudotrypsin: A Little-Known Trypsin Proteoform.

Project description:Trypsin is the protease of choice for protein sample digestion in proteomics. The most typical active forms are the single-chain ?-trypsin and the two-chain ?-trypsin, which is produced by a limited autolysis of ?-trypsin. An additional intra-chain split leads to pseudotrypsin (?-trypsin) with three chains interconnected by disulfide bonds, which can be isolated from the autolyzate by ion-exchange chromatography. Based on experimental data with artificial substrates, peptides, and protein standards, ?-trypsin shows altered kinetic properties, thermodynamic stability and cleavage site preference (and partly also cleavage specificity) compared to the above-mentioned proteoforms. In our laboratory, we have analyzed the performance of bovine ?-trypsin in the digestion of protein samples with a different complexity. It cleaves predominantly at the characteristic trypsin cleavage sites. However, in a comparison with common tryptic digestion, non-specific cleavages occur more frequently (mostly after the aromatic residues of Tyr and Phe) and more missed cleavages are generated. Because of the preferential cleavages after the basic residues and more developed side specificity, which is not expected to occur for the major trypsin forms (but may appear anyway because of their autolysis), ?-trypsin produces valuable information, which is complementary in part to data based on a strictly specific trypsin digestion and thus can be unnoticed following common proteomics protocols.

Phytocenosis biodiversity at various water levels in mesotrophic Lake Arakhley, Lake Baikal basin, Russia.

Project description:Small lakes have lower water levels during dry years as was the case in 2000-2020. We sought to show the biodiversity of plant communities at various water levels in Lake Arakhley. Changes in moisture content are reflected in the cyclical variations of the water level in the lake, which decreased approximately 2 m in 2017-2018. These variations affect the biological diversity of the aquatic ecosystems. We present the latest data on the state of the plant communities in this mesotrophic lake located in the drainage basin of Lake Baikal. Lake Arakhley is a freshwater lake with low mineral content and a sodium hydrocarbonate chemical composition. Changes in the nutrient concentration were related to precipitation; inflow volume and organic matter were autochtonous at low water levels. The most diverse groups of phytoplankton found in the lake were Bacillariophyta, Chlorophyta, and Chrysophyta. High biodiversity values indicate the complexity and richness of the lake's phytoplankton community. A prevalence of Lindavia comta was observed when water levels were low and Asterionella formosa dominated in high-water years. The maximum growth depth of lacustrine vegetation decreased from 11.0 m to 4.0 m from 1967 to 2018. Decreasing water levels were accompanied by a reduction in the littoral zone, altering the communities of aquatic plants. The hydrophyte communities were monodominant in the dry years and were represented by Ceratophyllum demersum. The vegetation cover of the lake was more diverse in high-water years and variations in the lake's water content altered the composition of biogenic substances. These changes were reflected in the lake's phytocenosis.
